ABSTRACT: Paired serological proteomes from CHB patients undergoing short-term and long-term anti-viral treatment were profiled using 4D-DIA-based proteomics technology.The serological proteomic landscapes following AVT were systematically analyzed and compared between fibrosis regressors and non-regressors.

Longitudinal serological proteomic dynamics during antiviral therapy (AVT) in chronic hepatitis B (CHB) patients with liver fibrosis remain poorly characterized. Chronic allergen exposure drives accumulation of long-lived IgE plasma cells in the bone marrow, giving rise to serological memory

Project description:Summary: Long-lived IgE plasma cells reside in the bone marrow of allergic mice and atopic humans, confer IgE serological memory and produce allergen-specific IgE that can drive anaphylaxis. Abstract: Immunoglobulin E (IgE) plays an important role in allergic diseases. Nevertheless, the source of IgE serological memory remains controversial. We re-examined the mechanism of serological memory in allergy using a dual-reporter system to track IgE plasma cells (PCs) in mice. Short-term allergen exposure resulted in the generation of IgE plasma cells that resided mainly in secondarylymphoid organs and produced IgE that was unable to degranulate mast cells. In contrast, chronic allergen exposure led to the generation of long-lived IgE plasma cells that were primarily derived from sequential class switching of IgG1, accumulated in the bone marrow (BM) and produced IgE capable of inducing anaphylaxis. Most importantly, IgE plasma cells were found in the BM of human allergic, but not non-allergic donors, and allergen-specific IgE produced by these cells was able to induce mast cell degranulation when transferred to mice. These data demonstrate that longlived IgE BMPCs arise during chronic allergen exposure and establish serological memory in both mice and humans.

The quality of DIA analysis is dependent on the quality of the spectral library. We used retinal rat tissue to create a spectral library of rats retina proteome. This data set may therefore be valuable for the future analysis of retinal proteins and research projects that focus on ocular disorders or function.

Proteomic signatures of the serological response to flu vaccination in a large human cohort study

Project description:This project investigates the serum proteomic signatures of the serological response to flu vaccination. We profiled the serum proteome of 160 flu vaccine recipients from a cohort study conducted in 2019-2020 season, with DIA-MS based proteomics. We compared the protein level between low and high responders based on the response measure seroconversion, as well as the adjusted seroconversion which excludes the confoudning effects of all the semographic factors recorded for these participants. We did the analysis for the composite seroconversion as well as for the individual strains. We also investigated the complex impact of age.

Proteomics Landscape of Diverse Serological Patterns in COVID-19

Project description:Serological test is a powerful tool to help clinicians assess the patients’ status during COVID-19 treatment. Highly diverse serological outcomes as exemplified by the negative or overly high IgG and IgM expressions are frequently reported but never elucidated interpreted with molecular insights. We classed a patient cohort (N=144) into four groups based on their longitudinal serological patterns and compared their differences in clinical indicators across a two-year manifestation. Thereafter, four patients from each group (N=16) with 111 serum samples collected during the 10 weeks since disease onset were enrolled for proteomic analyses.

Immunometabolic adaptation in monocytes underpins functional changes during pregnancy

Project description:Metabolic heterogeneity is a determinant of immune cell function. The normal physiological metabolic reprogramming of pregnancy that ensures the fuel requirements of mother and baby are met might also underpin changes in innate and adaptive immunity that occur with pregnancy and manifest as altered responses to pathogens and changes to autoimmune disease symptoms. Here, we use peripheral blood monocytes, susceptible to microenvironmentally determined reprogramming of metabolism, to determine if pregnancy at term provokes an altered metabolic profile that underpins functional change. Focusing on late gestation where any effect will be most profound, we reveal that monocytes lose M2-like and gain M1-like properties accompanied by reductions in mitochondrial mass, maximal respiration and cardiolipin content in pregnancy; glycolysis is unperturbed. We establish that muramyl dipeptide (MDP)-stimulated cytokine production relies on oxidative metabolism then show that reduced monocyte oxidative metabolism with pregnancy compromises the production of TNF and IL-6 in response to MDP but not LPS. Overall, mitochondrially centred metabolic capabilities of late gestation monocytes are downregulated revealing natural plasticity in monocyte phenotype and function that could reveal targets for improving pregnancy outcomes but also yield new therapeutic approaches to diverse metabolic and/or immune-mediated diseases beyond pregnancy.
