Project description:Ticks are blood-feeding arachnids that are known to transmit various pathogenic microorganisms to their hosts. During blood feeding, ticks activate their metabolism and immune system to efficiently utilise nutrients from the host's blood and complete the feeding process. In contrast to insects, in which the fat body is known to be a central organ that controls essential metabolic processes and immune defence mechanisms, the function of the fat body in tick physiology is still relatively unexplored. To fill this gap, we sought to uncover the repertoire of genes expressed in the fat body associated with trachea (FB/Tr) by analyzing the transcriptome of individual, partially fed (previtellogenic) Ixodes ricinus females . The resulting catalog of individual mRNA sequences reveals a broad repertoire of transcripts encoding proteins involved in nutrient storage and distribution, as well as components of the tick immune system. To gain a detailed insight into the secretory products of FB/Tr specifically involved in inter-tissue transport and humoral immunity, the transcriptomic data were complemented with the proteome of soluble proteins in the hemolymph of partially fed female ticks. Among these proteins, the hemolipoglyco-carrier proteins were predominant. When comparing immune peptides and proteins from the fat body with those produced by hemocytes, we found that the fat body serves as a unique producer of certain immune components. Finally, time-resolved transcriptional regulation of selected immune transcripts from the FB/Tr was examined in response to experimental challenges with model microbes and analysed by RT-qPCR. Overall, our data show that the fat body of ticks, similar to insects, is an important metabolic tissue that also plays a remarkable role in immune defense against invading microbes. These findings improve our understanding of tick biology and its impact on the transmission of tick-borne pathogens.

Project description:Anopheline mosquitoes transmit Plasmodium parasites to humans, and are responsible for an estimated 219 million cases of malaria, leading to over 400,000 deaths annually. The mosquito’s immune system limits Plasmodium infection in several ways, and hemocytes, the insect white blood cells, are key players in these defense responses. However, the full functional diversity of mosquito hemocytes and their developmental trajectories have not been established. We use bulk RNA sequencing (scRNA-seq) to analyze the transcriptional profiles of hemocytes, of guts, and of carcasses of mosquito hemocytes in response to blood feeding or infection with Plasmodium. Data from three independent biological replicates for each condition and time-point (day 0, 1, 2, 3, and 7 after sugar-feeding, blood-feeding or P. berghei infection).

Transcriptomes from 5,383 cells (collected 1, 3, and 7 days after feeding) revealed nine major cell clusters.

Project description:Staphylococcus aureus is a common human opportunistic pathogen which causes a wide range of infections from superficial skin infections to invasive infections including pneumonia and bloodstream infections. Here we describe how an inoculum of S. aureus activates the cellular and humoral response of Galleria mellonella larvae while growing and disseminating throughout the host, forming nodules and ultimately killing the host. An inoculum of S. aureus (2×106/ larva) decreased larval viability at 24 (80 ± 5.77%), 48 (55.93 ± 5.55%) and 72 (10.23 ± 2.97%) hours and this was accompanied by significant proliferation and disseminated of S. aureus between 6 hours to 48 hours and the formation of nodules in the host. The hemocyte (immune cell) densities increased between 4 and 24 hours and cells isolated from larvae after 6 and 24 hours exposure to heat killed S. aureus (2×106/ larva) showed altered killing kinetics as compared to control larvae. Alterations in the humoral immune response of larvae 6 and 24 hours post infection was also determined by quantitative shotgun proteomics. The proteome of 6 hour infected larvae was enriched for antimicrobial proteins, members of the prophenoloxidase cascade and a range of peptidoglycan recognition proteins. By 24 hours there was a significant increase in a range of antimicrobial peptides with anti-staphylococcal activity and proteins associated with nodule formation. The results presented here find S. aureus interacts with the larval host immune response, induces its expression and also forms nodules which are a hallmark of soft tissue infections during human infection.

Project description:The squid, Euprymna scolopes, and the luminescent bacterium, Vibrio fischeri, form a highly specific beneficial light organ symbiosis. Not only does the host have to select V. fischeri from the environment, but it must also prevent subsequent colonization by non-symbiotic microorganisms. Host macrophage-like hemocytes are believed to play a role in mediating the symbiosis with V. fischeri. Previous studies have shown that the colonization state of the light organ influences the host’s hemocyte response to the symbiont. To further understand the molecular mechanisms behind this process, two quantitative mass spectrometry-based proteomic techniques, isobaric tags for relative and absolute quantification (iTRAQ) and label-free spectral counting, were used to compare and quantify the adult hemocyte proteomes from colonized (sym) and uncolonized (antibiotic-treated/cured) squid. Overall, iTRAQ allowed for the quantification of 1,024 proteins with 2 or more peptides. Thirty-seven unique proteins were determined to be significantly different between sym and cured hemocytes (p-value < 0.05), with 20 more abundant proteins and 17 less abundant in sym hemocytes. The label-free approach resulted in 1,241 proteins that were identified in all replicates. Of 185 unique proteins present at significantly different amounts in sym hemocytes (as determined by spectral counting), 92 were more abundant and 93 were less abundant. Comparisons between iTRAQ and spectral counting revealed that 30 of the 37 proteins quantified by iTRAQ exhibited similar trends identified by the label-free method. Both proteomic techniques mutually identified 16 proteins that were significantly different between the two groups of hemocytes (p-value <0.05). The presence of V. fischeri in the host light organ influenced the abundance of proteins associated with the cytoskeleton, adhesion, lysosomes, proteolysis, and the innate immune response. These data provide evidence that colonization by V. fischeri alters the hemocyte proteome and reveals proteins that may be important for maintaining host-symbiont specificity.

Project description:Mussels are a group of sessile and filter-feeding invertebrates, and are key structural elements of many marine and freshwater ecosystems. The mussel haemolymph is the first defense line to pathogen infection and inflammation with an innate immune response. Despite the importance of this body fluid in determining the physiological condition of the animal, little is known about the molecular mechanisms underlying the cellular and humoral responses. Proteomic characterization of this body fluid has been hampered by the scarce Mytilus sp genomic data available. To systematic characterize the marine mussel Mytilus edulis hemolymph proteome, we have applied a mass spectrometry (nanoLC-MS/MS) strategy integrating genomics and transcriptomics data for proteins identification. After sample analysis and first identification based on MS/MS data comparison, proteins with unknown functions were annotated with blast using public database (nrNCBI) information. Overall 654 haemolymph proteins were identified with high confidence. The gene ontology analysis revealed that the majority of haemolymph proteins participate in primary cellular metabolic processes: energy production and metabolism of biomolecules. Nevertheless it also revealed a protein complement whose functions could be related to oxidative stress defense, xenobiotic detoxification, drug metabolism and immune response.

Project description:Toll pathway is a key mediator of antiplasmodial immunity and its mechanism of action is dependent on hemocytes (mosquito white blood cells). Anopheles gambiae were injected with dsRNA for Cactus, an inhibitor of the Toll pathway. Cactus silencing over activates the Toll pathway. The goal of this experiment is to evaluate hemocyte transcriptional changes between control mosquitoes injected with a non-related dsRNA (dsLacZ and dsCactus injected mosquitoes. We also divide hemocytes from control and experimental conditions into 2 groups: bound (hemocytes that attach to glass - granulocyte populations) and unbound (hemocytes that remain in suspension - prohemocytes and oenocytoids). Files from Unbound fractions are labelled: UB and from bound fractions are labelled B. Some of the samples were run twice and have 2 gz files.

Project description:This study assessed the development of disseminated candidiasis within Galleria mellonella larvae and characterised the proteomic responses of Candida albicans to larval hemolymph. Infection of larvae with an inoculum of 1 × 106 yeast cell reduced larval viability 24 (53.33 ± 3.33%), 48 (33.33 ± 3.33%) and 72 (6.66 ± 3.33%) hour post infection. C. albicans infection quickly disseminated from the site of inoculation and the presence of yeast and hyphal forms were found in nodules extracted from infected larvae at 6 and 24 hours. A range of proteins secreted during infection of G. mellonella were detected in larval hemolymph and these were enriched for biological processes such as interaction with host and pathogenesis. The candicidal activity of hemolymph after immediate incubation of yeast cells resulted in a decrease in yeast cell viability (0.23 ± 0.03 × 106, p < 0.05) as compared to control (0.99 ± 0.01 × 106). extracellular (in vivo) proteome of C. albicans in larval hemolymph were assessed. C. albicans responds to incubation in hemolymph ex vivo by the induction of an oxidative stress response, a decrease in proteins associated with protein synthesis and an increase in glycolytic proteins.
