Combined cysteine oxidation (C6-CPT/CysPAT) proteomics and phosphoproteomics of pervanadate-treated of HEK293T cells


ABSTRACT: HEK293T cells were treated with pervanadate for 0, 5 or 15 minutes to evaluate the effects on the phosphorylation and cysteine oxidation status across the proteome. Samples were labelled withTMT 18plex and C6-CPT/CysPAT labelling was used to allow the simultaneous enrichment of both the phosphoproteome and total vs. oxidised cysteine proteome.

Dynamic regulation of protein tyrosine phosphorylation (pTyr) by kinases and phosphatases enables cells to sense and respond to environmental changes. The widely used chemical pervanadate induces the accumulation of pTyr in mammalian cell lines.
