Replication-Associated Chromatin assembly promotes Pluripotency Exit and Differentiation
Ontology highlight
ABSTRACT: We map the proteomic composition of newly replicated nascent and mature chromatin in induced pluripotent stem (iPS) cells and during their differentiation along the presomitic mesoderm (PSM) pathway. We identify hundreds of proteins whose association with the replisome is significantly different between iPS and PSM cells. There are 40 samples in total including triplicates of total cell extract from each cell extract (iPS, and iPS differentiated for 24 hours or 48hours along the PSM pathway). There are also triplicates of Negative, nascent and mature IPOND pull-down samples performed on each of the cell types. These samples were run over 4 TMT MS batches with each batch additionally contained a replicate standard sample for normalization.
