Impact of FGF-2 and FGF-2 containing cell culture supernatants on the intracellular protein composition of ReNcell derived neuron

ABSTRACT: FGF-2 is an important protein involved in a vast number of molecular pathways and a number of cellular functions including glial cell differentiation and response to axon injury. This protein was found to be present significantly in supernatants of female derived murine oligodendrocyte progenitor cells (OPC) that have been maintained under hyperoxic conditions. Therefore, this protein might be important in protecting the brain against oxidative stress. Functional assays on neuronal cells in the presence of FGF-2 or OPC culture supernatants were performed and findings related to the impact of the treatment conditions on the intracellular protein response of ReNcell derived neurons were reported.

Cerebral oxygenation differences in the neonatal period of human preterm infants, along with sex-specific differences in combating oxidative stress, can lead to disruption of normal oligodendrocyte maturation and function, which in turn can differentially affect neuronal development and activity in the male and female brains. Secretory proteins and extracellular vesicles (EVs) are increasingly recognized as important mediators of intercellular communication and stress response in the brain. Our  ...[more]
