Wide-ranging analysis of microRNA profiles in sporadic amyotrophic lateral sclerosis using next-generation sequencing

Project description:miRNAs has an important role in the diagnosis and treatment of amyotrophic lateral sclerosis. we aimed to profile dysregulation of miRNAs in ALS blood and neuromuscular junction as well as healthy blood control by Next Generation Sequencing (NGS). The expression of three up-regulated miRNAs, as miR-338-3p, miR-223-3p and miR-326, in the ALS samples compared to healthy controls, has been validated by qRT-PCR in a cohort of 45 samples collected previously. Bioinformatics tools were used to perform ALS microRNAs target analysis and to predict novel miRNAs secondary structure. The analysis of the NGS data identified 696 and 44 novel miRNAs which were differentially expressed in ALS tissues.

Network analysis of the CSF proteome characterises convergent pathways of cellular dysfunction in ALS

Project description:Amyotrophic lateral sclerosis is a clinical syndrome with complex biological determinants, but which in most cases is characterised by TDP-43 pathology. The identification in CSF of a protein signature of TDP-43 network dysfunction would have the potential to inform the identification of new biomarkers and therapeutic targets. We compared CSF proteomic data from patients with ALS (n=41), Parkinson's disease (n=19) and healthy control participants (n=20). Weighted correlation network analysis was used to identify modules within the CSF protein network and combined with gene ontology enrichment analysis to functionally annotate module proteins. Analysis of module eigenproteins and differential correlation analysis of the CSF protein network was used to compare ALS and Parkinson's disease protein co-correlation with healthy controls. In order to monitor temporal changes in the CSF proteome, we performed longitudinal analysis of the CSF proteome in a subset of ALS patients. Alterations in the co-correlation network in CSF samples identified a set of pathways known to be associated with TDP-43 dysfunction in the pathogenesis of ALS, with important implications for therapeutic targeting and biomarker development.

Comprehensive and Unbiased Plasma Proteomics with Proteograph

Project description:Mild Cognitive Impairment (MCI) is a precursor to Alzheimer's disease (AD), necessitating early identification through reliable markers. We employed ProteographTM, an automated platform utilizing nanoparticles coupled with a liquid chromatography-tandem mass spectrometry (LC-MS/MS) workflow, enabling comprehensive, unbiased detection of low-abundance proteins (LAPs) in plasma. We analyzed the plasma proteome from 42 MCI patients and 13 controls. A total of 35 differentially expressed proteins (DEPs) were identified, with significant correlations to Mini-Mental State Examination (MMSE) scores. Composite panels of candidate proteins exhibited potential for distinguishing MCI and its subtypes, providing insight into underlying pathophysiological mechanisms. Our findings identify novel blood protein candidates with potential to enhance early MCI diagnosis and provide insight into the progression of AD.
