Project description:Solid State Fermentation (SSF) processes have been explored for yeast growth and protein and metabolites production. However, most of these processes lack standardization. In this work, we present a polylactic acid (PLA) 3D printed matrix that dramatically enhances yeast growth when embedded in liquid media compared to equivalent static cultures, and changes yeast expression patterns at the proteome level. Moreover, differences in sugar assimilation and ethanol production, as the main product of alcoholic fermentation, are observed. Our results suggest that these matrixes may be useful for a vast range of biotechnological applications based on yeast fermentation.

Project description:Polylactic acid (PLA) is a promising biodegradable material used in various fields, such as mulching films and disposable packaging materials. Biological approaches for completely degrading biodegradable polymers can provide environmentally friendly solutions. However, to our knowledge, no studies have performed transcriptome profiling to analyze PLA-degrading genes of PLA-degrading bacteria. Therefore, this study reports for the first time an RNA sequence approach for tracing genes involved in PLA biodegradation in the PLA-degrading bacterium Brevibacillus brevis. In the interpretation results of the differentially expressed genes, the hydrolase genes mhqD and nap and the serine protease gene besA were up-regulated by a fold change of 7.97, 4.89, and 4.09, respectively. This result suggests that hydrolases play a key role in PLA biodegradation by B. brevis. In addition, Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses showed that genes implicated in biofilm formation were upregulated. The biodegradation of PLA starts with bacteria attaching to the surface of PLA and forming a biofilm. Therefore, it could be confirmed that the above genes were up-regulated for access to PLA and biodegradation. Our results provide transcriptome-based insights into PLA biodegradation, which pitch a better understanding of microbial biodegradation of plastics.

Project description:Plastics are one of the most preoccupying emerging pollutants. Macroplastics released in the environment degrade into microplastics and nanoplastics. Because of their small size, these micro and nano plastic particles can enter the food chain and, in addition to their ecotoxicological effects, contaminate humans with still poorly known biological effects. Plastics being particulate pollutants, they are handled in the human body by scavenger cells such as macrophages, which are important players in the immune system. Because of all the potential problems, it is advocated to replace fossil fuel-based plastics by bio-based and bio-degradable plastics, among which poly-hydroxyalkanoates are the most promising. However, the effects of these on mammalian cells are even less known than those of fossil fuel-based plastics. We therefore designed a study aiming at investigating the effects of polylactic acid (PLA) nanoparticles on macrophages. Indeed, being a plastic, PLA is known to fragment and liberate micro and nanoparticles, exactly as conventional plastics. These particles will be internalized by macrophages and may induce functional consequences on these cells. Proteomics showed important adaptive changes of the proteome in response to exposure to PLA, and several important pathways such as mitochondrion, lysosomes or endoplasmic reticulum were highlighted by the proteomic analysis. However, validation experiments showed that most of these changes were homeostatic and allowed the cells to keep these functions unaltered. When the inflammatory response was examined, no major increase in the secretion of tumor necrosis factor or interleukin 6 was observed. However, the secretion of these cytokines in response to lipopolysaccharide was altered after exposure to PLA. The production of interleukin 6 was decreased, while the production of tumor necrosis factor, showing a complex alteration of cellular responses after exposure to PLA nanoparticles. In conclusion, these results provide a better understanding of the responses of macrophages to exposure to the biodegradable PLA nanoparticles.

Project description:The freshwater mussel Dreissena bugensis was exposed for nine days to different microplastic particles, in detail, to three petroleum-based polymers (polyamide (PA), polyethylene terephthalate (PET), polystyrene (PS)), to one bio-based polymer (polylactic acid (PLA)) and to ground mussel shells (MS), serving as a natural particle control (size range: 20-75 µm;1000 p ml-1). Behavior endpoints were analyzed with hall sensor based real-time valvometry. Additionally, biochemical alterations of ROS detoxifying enzymes were analyzed, and a proteomic profiling on digestive gland tissue was performed.

Project description:Biodegradable plastics, such as polylactic acid (PLA), are widely considered as sustainable alternatives to fossil fuel-derived polymers; however, their degradation generates polylactic acid microplastics (PLA-MPs), which raises ecological concerns in freshwater ecosystems. In this study, we investigated the responses of Microcystis aeruginosa to PLA-MPs (1, 10, and 50 mg/L) across a 45-day co-culture experiment. We found the PLA-MPs initially inhibited algal growth (days 18, 27) through oxidative stress, as indicated by elevated MDA content (1.94 nmol/mg protein at 50 mg/L, 1.57 times higher than the control) and SOD activity (1.30 U/mg Protein, 2.45 times higher than the control). However, during stabilization (days 36 and 45), PLA-MPs promoted growth in a dose-dependent manner, with Chl-a content in the 1, 10, and 50 mg/L groups reaching 1.12, 1.36, and 1.44 times that of the control, respectively. Concurrent shifts in inorganic carbon speciation, EPS secretion, and cellular aggregation highlighted dynamic adaptation strategies. Finally, based on the proteomic analysis, it was found that PLA-MPs promoted the growth of M. aeruginosa by improving photosynthetic efficiency, enhancing the expression of the TCA cycle and oxidative phosphorylation, and promoting cell division and proliferation. Our results confirm that PLA-MPs have a long-term effect on the growth of M. aeruginosa, providing fundamental theoretical guidance for investigating the ecological and environmental impacts of PLA-MPs in aquatic ecosystems.

Project description:<p>The phycosphere is an important ecological niche for bacteria and antibiotic resistance genes (ARGs). However, whether and how the interaction between microalgae and bacteria changed, and its further effect on the transmission of ARGs under pollutant stress remains enigmatic. Here, Auxenochlorella pyrenoidosa was co-cultured with bacteria screened from lake water to explore the algal-bacteria interaction and ARGs transmission in the presence of florfenicol (FF) and polylactic acid microplastics (PLA MPs). Our study demonstrated that the growth and metabolism of A. pyrenoidosa were promoted under FF treatment or co-treatment with PLA MPs, validated by phenotypic, transcriptome, and metabolome analyses. In contrast, the abundance of phycospheric bacteria was decreased as a result of niche competition. Nonetheless, the transmission of ARGs in the phycosphere was promoted due to the enrichment of antibiotic-resistant bacteria, especially Pseudomonas, rather than horizontal gene transfer. Algal-bacteria co-culture experiment further suggested that vitamin B6 secreted by Pseudomonas sp. likely contributes to underpinning A. pyrenoidosa survival under FF and PLA MPs stress. These findings underscore the dynamic interplay and co-evolution between algae and bacteria under pollutant exposure, and reveal a potential mechanism of vitamin B6-mediated mutualism. This study provides new insights into the assembly of phycospheric bacterial communities and the adaptive strategies of microalgae in contaminated aquatic environments.</p>

Project description:Nanoplastics were generated from common consumer plastics (PET, HDPE, PS, PVC) and exposed to simulated marine weathering for up to 10 weeks. Fourier-Transform Infrared (FT-IR) spectroscopy and ζ-potentials measurements revealed continuous changes in the composition of the nanoplastics consistent with oxidation. Although the chemical composition and oxidation of the nanoplastics influenced their ability to sorb Polycyclic Aromatic Hydrocarbons (PAHs), for all investigated conditions, sorption of PAH to nanoplastics achieved effective PAH concentrations that were orders of magnitude higher than the solubility limit in water. In an intestinal membrane model, PAH-loaded nanoplastics enhanced the overall PAH transport into and across the membrane, with HDPE achieving the highest intracellular PAH concentration. RNA sequencing of cell membranes exposed to nanoplastics revealed significant transcriptional changes, including upregulation of oxidative stress and detoxification pathways (NQO1, CYP1A1, CYP1B), especially in response to PAH-loaded nanoplastics, while genes associated with basic cell functions, such as cellular migration (NCKAP5, MACROD2) and division (KIF20A) were downregulated. These findings suggest that nanoplastics can increase bioaccessibility and bioavailability of hydrophobic carcinogens and enhance cellular stress, raising concerns about the potential environmental and health impacts associated with nanoplastics as carriers of hydrophobic environmental toxins.
