Project description:Bacterial pathogens harbor specialized secretion systems that inject effector proteins into the host cell to establish infection and disease. While many bacterial effectors post-translationally modify proteins to influence host responses, the extent to which effectors modify host RNA is currently unknown. Here we performed RNA-interactome capture (RIC) to isolate effectors bound to host cellular messenger RNA (mRNA) during Legionella pneumophila infection. RIC identified an uncharacterized effector, FadA (Lpw16921), which localized to the host-cell nucleus and interacted with host mRNAs at uracil (U)-rich RNA motifs. FadA exhibited NADPH-oxidase activity that mediated 8-oxo-guanine (oxo8G) modifications of mRNA substrates, resulting in oxidative damage and inhibition of translation. Infection with L. pneumophila harboring wild-type FadA, but not a catalytically inactive mutant, increased oxo8G modifications, suppressed cytokine responses, and promoted bacterial persistence in vivo. Our findings demonstrate the potential for a secreted Q2 effector to post-transcriptionally modify host mRNA as a mechanism to promote bacterial virulence.

Project description:This study aimed to systematically identify host cellular proteins that interact with newly discovered Brucella abortus type IV secretion system (T4SS) effector proteins, designated BT4E3, BT4E4, BT4E19, BT4E32, BT4E33, BT4E37, and BT4E43. To achieve this, we employed an affinity purification–mass spectrometry (AP-MS) strategy in a human cell context. The coding sequences of individual BT4E effectors were cloned into the p3xFLAG-CMV-7.1 vector as C-terminal fusions with 3×Flag tag. These constructs, along with a p3xFLAG-CMV-7.1-control vector (mCherry-Strep only), were transiently transfected into HEK293T cells using Lipofectamine 3000. Twenty-four hours post-transfection, cells were lysed under mild non-denaturing conditions (100 mM Tris-HCl pH 8.0, 150 mM NaCl, 1 mM EDTA, 1% NP-40) supplemented with protease and phosphatase inhibitors. Flag-tagged effector complexes were affinity-purified using Flag Sepharose beads, followed by stringent washing and elution with 2.5 mM D-desthiobiotin. Each effector purification was performed in three independent biological replicates to ensure reproducibility. Eluted protein complexes were verified by Western blotting, then processed for liquid chromatography–tandem mass spectrometry (LC-MS/MS) analysis. LC-MS/MS was carried out on an Orbitrap Fusion Tribrid mass spectrometer (Thermo Fisher Scientific) coupled online to an EASY-nLC 1000/1200 nanoflow system. The resulting proteomics data enable the identification of high-confidence host binding partners for each BT4E effector, providing insights into the molecular mechanisms by which Brucella manipulates host cellular processes—including vesicular trafficking, innate immune signaling, autophagy, and cell death pathways—to establish intracellular infection. This dataset serves as a valuable resource for understanding Brucella pathogenesis and host–pathogen interactions mediated by T4SS effectors.

Project description:The oomycete pathogen Phytophthora sojae causes root rot of soybean. During infection, the pathogen is thought to deliver dozens, if not hundreds, of effector proteins into the host to manipulate intracellular systems. Although these pathogen proteins often exhibit similar N-terminal delivery domains, the remaining effector region is rarely homologous to known protein domains, making it difficult to predict its biochemical function during infection. As a complement to studies in the natural host, Saccharomyces cerevisiae has been successfully used as a model system to explore the biochemical function of individual pathogen effectors. The presumption is that many effectors target conserved eukaryotic pathways in the host and consequently the expression of effectors in yeast will confer corresponding phenotypes. Indeed, putative effector functions identified using yeast functional genomic approaches have subsequently been validated in the natural host. Over-expression of the Phytophthora sojae effector Avh172 (PsAvh172) inhibits the growth of Saccharomyces cerevisiae, suggesting that the effector targets a biological pathway conserved with plants. In this study, the transcriptomes of yeast expressing PsAvh172 or an empty vector were compared to examine the global transcriptional response, in hopes of discerning the effectors biochemical target.
