Identification of drought stress related genes in NatA depleted mutants

Project description:N-terminal protein acetylation is one of the most common protein modifications in eucaryotes and is catalyzed co-translationally by N-terminal aceytltransferases (Nats). Regarding the number of predictaed substrates, NatA is the main Nat complex in human and yeast and is composed of the catalytic subunit NAA10 and the auxilliary subunit NAA15. The down-regulation of NAA10 and NAA15 results in Arabidopsis in drought resistant plants. This study focus on the identification of altered gene expression leading to the drought resistant phenotype. Microarray analysis was used to identify misregulated genes in the NatA depleted mutants responsible for the drought resistant phenotype Arabidopsis thaliana plants were grown on soil for 6 weeks under short-day conditions (8h light / 16h dark cycles) with normal watering and subsequently challenged with drought for 10 days. After 10 days of drought Arabidopsis leaf material was harvested and used for RNA extraction and hybridization on Affymetrix microarrays. Four biological replicates from each genotype and condition were analyzed.

Analysis of lacking Nα-terminal acetylation of proteins by NatA and NatB in Saccharomyces cerevisiae on mRNA levels and protein synthesis

Project description:N-terminal (Nt)-acetylation is a highly prevalent co-translational protein modification in eukaryotes, catalyzed by at least five Nt-acetyltransferases (Nat) with differing specificities. Nt-acetylation has been implicated in protein quality control but its broad biological significance remains elusive. We investigated the roles of the two major Nats of S. cerevisiae, NatA and NatB, by performing transcriptome and translatome profiling by using mRNA sequencing and ribosome profiling. The results are combined with global proteome, aggregome and stabilome datasets. To analyze previously proposed physiolocal roles of NatA and NatB in yeast cells on protein stability and interaction specific growth conditions such as different growth media and heat stress are used in addition to physiological growth.

AtNAA50 behaviour in the core NatA complex

Project description:In eukaryotes, the N-terminal acetylation (NTA) is one of the most frequent protein modifications. In many organisms, and especially in plants, its biological function remains a mystery. In Arabidopsis thaliana, a large part of the proteome acetylation is catalyzed co-translationally by the action of the core NatA complex, which consitst of NAA10 and NAA15, respectively the catalytic and ribosome-anchoring subunits. This complex interacts with the NAA50 protein (NatE), which also has an N-acetyltransferase in organisms such as human and fruit fly. This project focuses on the effect of AtNAA50 knockouts on the activity of the essential NatA complex.

Project description:We used immunoprecipitation (IP) and mass spectrometry (MS) to identify proteins that interact with MFSD7C. Because an anti-MFSD7C monoclonal antibody is not available, we used MFSD7C tagged with both Myc and FLAG epitopes in a MFSD7C-expressing murine alveolar macrophage cell line MH-S (Supplementary Fig. 2a-c and see Methods for details). A total of 58 proteins were identified excluding MFSD7C and common contaminants of IP experiments such as keratin, actin, and ribosomal proteins (Table 1). Twenty-six of the 58 proteins are annotated as mitochondrial proteins and 4 (three Fc receptors and one integrin) are known to reside in the cytoplasmic membrane. The 58 proteins were classified into 10 different functional categories based on gene ontology.

The Nα-acetyltransferase NAA50 mediates plant immunity independent of the NatA complex

Project description:Nα-terminal acetylation (NTA) ranges among the most abundant protein modifications in higher eukaryotes. Over 40 % of the human and plant proteome are co-translationally acetylated by a single Nα-acetyltransferase (Nat) termed NatA. The core NatA complex consists of the catalytic subunit NAA10 and the ribosome-binding subunit NAA15. In humans, the regulatory subunit HYPK and the acetyltransferase NAA50 join the complex. Even though both are conserved in Arabidopsis thaliana, only AtHYPK is known to interact with AtNatA. Here we analyse the interactome of AtNAA50 and provide evidence for the association of the enzyme with AtNatA. Moreover we demonstrate that AtNAA50 interacts with ribosome-associated proteins involved in protein translation, folding and translocation. A recent study shows that acetylation protects NatA substrates from proteasomal degradation. In consequence, NatA depletion results in an accelerated protein turnover. We report that this is not true for the depletion of AtNAA50, suggesting that the enzyme is not required for NatA activity. In line with this, novel amiNAA50 knockdown lines do not display the characteristic drought resistance of NatA mutants. Instead, complementary transcriptome and proteome analyses suggest that AtNAA50 is a negative regulator of plant immunity. Pathogen challenges confirm that amiNAA50 plants are resistant to oomycetes and bacteria.

Overlap of NatA and IAP substrates implicates N-terminal acetyltion in protein stabilization

Project description:SMAC/DIABLO and HTRA2 are mitochondrial proteins whose N-termina sequences, known as inhibitor of apoptosis binding motifs (IBMs), bind and activate ubiquitin ligases knwon as inhibitor of apoptosis proteins (IAPs), unleashing a cell´s poteintial.IBMs comprise a 4-residue, loose consensus sequence, and binding to IAPs requires an unmodified N-terminus. Closely related, IBM-like N-termini, are present in approximately 5% of human proteins. We show that suppression of the N-alpha-acetytransferase NatA truns these cryptic IBM-like sequences into very efficient IAP binders in cell lysates and in vitro, and ultimately also triggers apoptosis. Thus, N-terminal acetylation of IBM-like motifs in NatA substrates shields them from IAPs. This previously unrecognized relatonship suggests that N-terminal acetylation is generally protective against protein degradation in human cells. It also identifies IAPs as agents of a general quality control mechanism targeting unacetylated rogues in metazoans
