Proteome profiling of IEC-6 cells after treatment with Akkermansia muciniphila (AM), Bifidobacterium longum (BL), or AM-BL combination

ABSTRACT: To profile the proteome of intestinal epithelial IEC-6 cells in response to treatment with Akkermansia muciniphila, Bifidobacterium longum, or Akkermansia muciniphila + Bifidobacterium longum co-culture. In addition, we also add arabinoxylan (AX) to Akkermansia muciniphila + Bifidobacterium longum combination since we previously found that this glycan enhance their synergistic effect.

Project description:To understand how glycans influence the growth of probiotic strains in monocultures and communities, we characterized the growth and perform proteome profiling of 3 major probiotics, A. muciniphila (AM) BAA-835, B. breve (BB) JCM1192, and B. longum (BL) JCM1217 in monocultures and co-cultures grown in media with monosaccharides (Glucose & GlcNAc), Mucin, or Arabinoxylan. For the proteomic file annotations, A refers to AM monoculture, B refers to BB monoculture, and L referes to BL monoculture, AL refers to AM+BL co-culture, AB refers to AM+BB co-culture. The letter following the underline specify the media: g refers to Glc+GlcNAc, m refers to mucin, ax refers to arabinoxylan. If there is no underline following the species annotations, the cultures were grown in the base media only. Numbers following the letters specify biological replicates.

Transcriptional response to protease treatments (kallikrein, papain and chymotrypsin) of Bifidobacterium breve 210B

Project description:Members of the serpin (serine protease inhibitor) superfamily have been identified in higher, multicellular eukaryotes, as well as in bacteria, although surveillance of available genome sequences indicates that bacterial serpin-encoding (ser) homologs are not widely distributed. In members of the genus Bifidobacterium this gene appears to be present in at least five, and perhaps up to nine, out of 30 species tested. Moreover, phylogenetic analysis using available bacterial and eukaryotic serpin sequences revealed that bifidobacteria specify serpins that form a separate clade. We characterized the ser210B locus of Bifidobacterium breve 210B, which consists of a number of genes, whose deduced protein products display significant similarity to proteins encoded by corresponding loci found in several other bifidobacteria. Northern hybridization, primer extension, micro array analysis, RT-PCR and Quantitative Real Time (qRT) - PCR analysis revealed that a 3.5 kb polycistronic mRNA, encompassing the ser210B operon with a single transcriptional start site, is strongly induced following treatment of B. breve 210B cultures with particular proteases. Project description:Bifidobacterium longum subsp. infantis is a bacterial commensal that colonizes the breast-fed infant gut where it utilizes indigestible components delivered in human milk. Accordingly, human milk contains several non-protein nitrogenous molecules, including urea at high abundance. This project investigates the degree to which urea is utilized as a primary nitrogen source by Bifidobacterium longum subsp. infantis and incorporation of hydrolysis products into the expressed proteome.

Akkermansia muciniphila outer membrane protein Amuc_0904 promotes goblet cell differentiation

Project description:Akkermansia muciniphila is recognized as a promising probiotic that improves the symptoms of a variety of diseases. However, the role and mechanism of A. muciniphila in regulating intestinal homeostasis remain to be explored. Here, we discovered that A. muciniphila was dramatically increased during colitis recovery, and its colonization greatly increased goblet cells to protect the intestinal barrier in mice. Amuc_0904, a previously uncharacterized A. muciniphila outer membrane protein, was identified to induce goblet cell differentiation.
