LncRNA NEAT1 promotes immunosuppression in gastric cancer by maintaining the m6A methylation of SEMA3A in CAFs via exosomes under endoplasmic reticulum stress

ABSTRACT: Endoplasmic reticulum stress (ERS) regulates the function of immune cells in the tumor microenvironment and suppresses the antitumor immune response. In order to formulate the TME of ERS-GC, we cocultured the human CAFs with the supernatant of human gastric cancer cells stimulated by TM. Mass spectrometry sequencing revealed that ERS state regulated the overexpression of 58 proteins and downregulation of the expression of 56 proteins in CAFs, including m6A-modified proteins and SEMA3A, an immunosuppression-related protein.

CDK12 regulates protein translation independent of kinase activity and its degradation sensitizes colorectal cancer to mTOR inhibition

Project description:Despite advances in treatment, colorectal cancer (CRC) remains a leading cause of cancer-related death worldwide, with limited therapeutic options for patients at advanced or metastatic stage. Synthetic lethality represents an emerging strategy that can expand options for patients unresponsive to existing therapies. However, characterizing effective synthetic lethal gene pairs remains a challenge. In this study, we used comprehensive bioinformatics approaches to identify that CDK12 and mTOR are synthetically lethal in CRC and propose a potential drug combination strategy. We demonstrate that the CDK12 degrader BSJ-4-116 and the mTOR inhibitor everolimus synergistically inhibit CRC cell proliferation and induce apoptosis, both in vitro and in vivo. Notably, this synergistic effect is independent of CDK12’s kinase activity and arises from the blockage of protein synthesis. Mechanistically, CDK12 promotes rRNA 2’-O-methylation, ribosome biogenesis, and protein translation by orchestrating the morphology and function of FBL and NPM1. The combined deficiency of CDK12 and mTOR inhibition synergistically remodels the translatome, affecting key oncogenic drivers such as KRAS and MDM2. Importantly, this combination therapy inhibited tumor growth in patient-derived xenografts, highlighting its potential clinical relevance. Our study uncovers a novel, kinase-independent function of CDK12 in protein translation and elucidates the synergistic benefits of CDK12 degraders and mTOR inhibitors, offering a promising new therapeutic strategy for colorectal cancer.

CRTAC1 Enhances the Chemosensitivity of Non-Small Cell Lung Cancer to Cisplatin by Eliciting RyR-mediated Calcium Release and Inhibiting Akt1 Expression

Project description:Sensitivity to platinum-based combination chemotherapy is associated with a favorable prognosis in the patients of non-small cell lung cancer (NSCLC). Here, our results obtained from analyses of the Gene Expression Omnibus database of NSCLC patients showed that cartilage acidic protein 1 (CRTAC1) plays a role in the response to platinum-based chemotherapy. Overexpression of CRTAC1 increased sensitivity to cisplatin in vitro, whereas knockdown of CRTAC1 decreased chemosensitivity of NSCLC cells. In vivo mouse experiments showed that CRTAC1 overexpression increased the antitumor effects of cisplatin. CRTAC1 overexpression promoted NFAT transcriptional activation by increasing intracellular Ca2+ levels, thereby inducing its regulated STUB1 mRNA transcription and protein expression, accelerating Akt1 protein degradation, and in turn enhancing cisplatin-induced apoptosis. Taken together, the present results indicate that CRTAC1 overexpression increases the chemosensitivity of NSCLC to cisplatin treatment by inducing Ca2+-dependent Akt1 degradation and apoptosis, suggesting the potential of CRTAC1 as a biomarker for predicting cisplatin chemosensitivity. Our results further reveal that modulating the expression of CRTAC1 could be a new strategy for increasing the efficacy of cisplatin in chemotherapy of NSCLC patients.

Identification of cytoplasmic proteins interacting with unliganded estrogen receptor α and β- in human breast cancer cells

Project description:Estrogen Receptor subtypes (ERα and ERβ) are transcription factors sharing similar structure, however, they often perform opposite roles in breast cancer’s cell proliferation and tumor progression. Besides the well-characterized genomic actions of ERs upon ligand binding, rapid non-genomic cytoplasmic changes together with the recently discovered ligand-free action of ERs are emerging as key regulators of tumorigenesis. The identification of cytoplasmic interaction partners of unliganded ERα and ERβ may help characterize the molecular basis of the extra-nuclear mechanism of action of these receptors, revealing novel mechanisms to explain their role in breast cancer response or resistance to endocrine therapy. To this aim, in this study, cytoplasmic extracts from stably expressing TAP-ERα and -ERβ MCF-7 cell clones were subjected to interaction proteomics in the absence of estrogen stimulation, leading to the identification of 84 and 142 proteins associated with unliganded ERα and ERβ, respectively. Functional analyses of ER subtype-specific interactomes revealed significant differences in the molecular pathways associated to each receptor in the cytoplasm. This work reports the first identification of the unliganded ERα and ERβ cytoplasmic interactomes in breast cancer cells, providing novel experimental evidence on the non-genomic effects of ERs in the absence of hormonal stimulus.

Quantitative proteomic analysis of human malignant ascites

Conversely, most proteomic studies on ascites have focused on ovarian cancer and provided insufficient depth for biomarker discovery. microRNA expressions in gastric cancer

Project description:The study was undertaken to identify microRNAs differently expressed by intestinal type of gastric cancer using miRNA microarray. The miRNA expression in the intestinal type of gastric cancer depending on H. pylori infection suggest that different gastric cancer pathogenesis could be exist between H. pylori-positive and -negative gastric cancer. Total RNA was extracted from cancerous region and non-cancerous regions in formalin fixed paraffin embedded tissues of intestinal type gastric cancer patients who were H. pylori-positive (n=8) or -negative (n=8).
