Project description:This project mainly aims to characterize the complex toxic components present in the venom of Indian cobra (Naja naja) from the Western Ghats of India. Naja naja (NN) is native to the Indian subcontinent and is also found in Pakistan, Sri Lanka, Bangladesh and Southern Nepal. It is a highly venomous snake species of genus Naja of the Elapidae family. They are seen in wide habitats like plains, dense or open forests, rocky terrains, wetlands, agricultural lands, and outskirts of villages and even in highly populated urban areas. This species has been included in the ‘Big 4’ category of venomous snake species that accounts for majority of morbidity and mortality cases in India. Therefore, exploring the venom proteome of Naja naja is decisive to develop and design new antivenom and therapeutics against its envenomation. The venom proteome of Naja naja was characterized through various orthogonal separation strategies and identification strategies. In order to achieve this the crude venom components were resolved on a 12% SDS page. Also, the venom was decomplexed through reversed-phase HPLC followed by SDS analysis. Further each of the bands were subjected to in-gel digestion using trypsin, chymotrypsin and V8 proteases. All the digested peptides were then subjected to Q-TOF LC-MS/MS analysis.

Project description:This project mainly aims to characterize the complex toxic components present in the venom of Trimeresurus malabaricus (Malabar pit viper). Since Trimeresurus malabaricus (Malabar pit viper) species are mainly inhabited to plantation crop areas, its envenomation is a serious threat to the human population thriving in these zones, especially to the plantation workers. Therefore, exploring the venom proteome of Malabar pit viper is decisive to develop and design new antivenom and therapeutics against its envenomation. As described in this study, applying various orthogonal separation strategies helped in dissecting venom constituents of Trimeresurus malabaricus and is the first comprehensive attempt in revealing the complex venom profile of Malabar pit viper through proteomics approaches incorporating multiple database searches. In order to achieve this the crude venom components were resolved on a 12% SDS page. Further each of the bands were subjected to in-gel trypsin digestion. The crude venom was also subjected to ion-exchange chromatography separation. The obtained fractions were subjected to in-solution trypsin digestion. All the digested peptides were then subjected to Q-TOF LC-MS/MS analysis.

Project description:Background: Helicobacter pylori is an important human pathogen that infects the human gastric mucosa leading to chronic inflammation which is responsible for severe gastroduodenal diseases. The Outer Inflammatory Protein A (OipA) of H. pylori is an important virulence factor. However, the role of OipA in gastric cancer and ulcer development is still not well-established. Phase variation within a CT dinucleotide repeat motif of oipA switches its expression “on” and “off”. This study aims to elucidate the role of OipA in H. pylori infection using oipA “on” and “off” clinical strains and oipA deletion mutant. Methods: Proteomics analysis was performed on AGS human gastric epithelial cell line post-infection with oipA “on” and “off” H. pylori using liquid chromatography / mass spectrometry (LC/MS). In addition, AGS apoptosis and cell cycle assays were performed. To confirm the findings, oipA deletion mutant (ΔoipA) was generated and expression of VacA was detected using Western blotting. Result: Expression of AGS proteins involve in human disease was suppressed / down-regulated post-infection with oipA “off” strains, as well as oipA mutant, compared to oipA “on” strains. Furthermore, oipA “off” strains and oipA mutant resulted in a higher level of apoptosis and G0/G1 cell-cycle arrest in AGS cells than oipA “on” strains. Interestingly, deletion of oipA was found to increase VacA production by the bacterium. Discussion: The capability of oipA “off” strains to induce apoptosis and suppress proteins having roles in human disease may suggest that these H. pylori may be less virulent or may even be protective against carcinogenesis compared to its oipA “on” counterparts. This may potentially explain the higher incidence of gastric cancer among East Asian which oipA “on” strains predominates. Conclusions: Data from this study strengthened our understanding of the role of H. pylori OipA as a virulence factor in chronic inflammation, metastasis and carcinogenesis.

Project description:Sesbania grandiflora, a fast-growing shrub from the Fabaceae family, is extensively researched for its therapeutic properties. However, the seeds of Sesbania grandiflora have been largely overlooked in scientific studies. Despite its highly valued medicinal properties, there has been no proteomic investigation and structural characterization of the seed proteins of this plant. Our study aims to address this gap by exploring the proteomic profile of Sesbania grandiflora seeds through bottom-up proteomic analysis. The extracted seed proteins, fractionated by ammonium sulfate into three different saturations viz. 30%, 60%, and 90% were separated on an SDS-PAGE gel. Protein containing bands were carbamidomethylated and trypsin digested followed by high resolution mass spectrometric data collection in a data-dependent mode. Proteins were then identified by searching the mass spectrometry data against Fabaceae database through Mascot search engine. To augment the reliability and accuracy of the identifications, Scaffold software was employed for validation by setting the minimum protein and peptide probability filter at 99% and 95%, respectively. Our comprehensive proteomic analysis identified 731 proteins from Sesbania grandiflora seeds, including seed storage proteins, proteases, protease inhibitors, ribonucleoproteins, transferases, isomerases, and hydrolases. This study provides a fundamental understanding of the seed proteome of Sesbania grandiflora, offering insights that could facilitate further research into the plant's therapeutic properties and potential applications.

Project description:The nose-horned viper, its nominotypical subspecies Vipera ammodytes ammodytes (Vaa) in particular is, medically, the most relevant snake in Europe. The local and systemic clinical manifestations of poisoning by the venom of this snake are the result of the pathophysiological effects inflicted by enzymatic and non-enzymatic venom components acting, most prominently, on blood, cardiovascular and nerve systems. This venom comprises the most complex mixture of pharmacologically active proteins and peptides of all European snakes. To help improve the current antivenom therapy towards higher specificity and efficiency, and to assist drug discovery, we have constructed, by combining transcriptomic and proteomic analyses, the most comprehensive library yet of the Vaa venom proteins and peptides. At the protein level, 57 venom proteins belonging to 16 different protein families have been identified and, with SVSPs, sPLA2s, snaclecs and SVMPs, comprise about 80% of all venom proteins.

Project description:Sirex noctilio F., a Eurasian horntail woodwasp recently introduced into North America, oviposits in pines and other conifers and in the process spreads a phytopathogenic fungus that serves as a food source for its larvae. During oviposition the woodwasp also deposits a mucus produced in its acid (venom) gland that alters pine defense responses and facilitates infection by the fungus. A 26,496-feature loblolly pine cDNA microarray was used to survey gene expression of pine tissue responding to S. noctilio venom. Six genes were selected for further assessment by qRT-PCR, including one that encoded an apparent PR-4 protein and another that encoded a thaumatin-like protein. Expression of both was strongly induced in response to venom, while expression of an apparent actin gene (ACT1) was stable in response to the venom. The pattern of gene response was similar in Pinus taeda L. and P. radiata D. Don, but the magnitude of response in P. radiata was significantly stronger for each of the induced genes. The magnitude of biomarker gene response to venom also varied according to genotype within these two species. The qRT-PCR assay was used to demonstrate that the primary bioactive component in S. noctilio venom is a polypeptide. Reference design. Two condition experiment, two time points each compared to a common reference. Two biological replicates, two technical replicates, 12 slides total, duplicate/re-scanned images submitted for each slide.

Project description:Mucuna pruriens extract MPE pretreatment may have a direct protective effect on heart (other than immunological neutralization of the venom neurotoxin and phospholipase A2 by the anti-MPE antibodies) that renders the heart more resistant to the toxic action of the venom The direct protective effect probably involves functional changes to the cardiac tissue that enable the heart to resist the reduction of contractility and rate induced by the cobra venom.To explore the possibility of the direct action of MPE pretreatment on heart and to understand the molecular events involved in the protection of MPE pretreatment against the lethal action of Naja sputatrix venom, gene expression studies were carried out using microarray analysis. Rats were divided into four groups (n=6): negative control (abbreviated as ‘negative’ group), MPE pretreated group (abbreviated as ‘MPE’ group), N. sputatrix venom-challenge group (abbreviated as ‘NS’ group) and N. sputatrix venom-challenge to MPE pretreated animals group (abbreviated as ‘MPE-NS’ group). In the ‘MPE’ group, rats were injected with MPE at a dose of 21 mg/kg (i.p.), on day 0, 7 and 14, and sacrificed on day 21. In the ‘negative’ group (the untreated, control group), rats were injected with saline of the same volume and sacrificed also on day 21. Hearts were then harvested immediately. In the N. sputatrix venom-challenge group (‘NS’ group), untreated rats were challenged with 1.5 LD50 (1.25 ?g/g) of N. sputatrix venom whereas in the venom challenge to MPE pretreated animals group (the ‘MPE-NS’ group), MPE pretreated rats were challenged with 1.5 LD50 (1.25 ?g/g) of N. sputatrix venom, both on day 21. For the ‘NS’ and ‘MPE-NS’ group, the rats were observed for 24 h after venom challenged and hearts were harvested as soon as death occurred or 24 h after the venom injection, whichever occurred first.

Project description:While safety of fasting therapy is debated in humans, extended fasting occurs routinely and safely in wild animals. To do so, food deprived animals like breeding penguins anticipate the critical limit of fasting by resuming feeding. To date, however, no molecular indices of the physiological state that links spontaneous refeeding behaviour with fasting limits had been identified. Blood proteomics and physiological data reveal here that fasting-induced body protein depletion is not unsafe “per se”. Indeed, incubating penguins only abandon their chick/egg to refeed when this state is associated with metabolic defects in glucose homeostasis/fatty acid utilization, insulin production and action, and possible renal dysfunctions. Our data illustrate how the field investigation of “exotic” models can be a unique source of information, with possible biomedical interest.
