LC-MS/MS analysis of murine endothelial cell lines: normal brain microvascular cell line vs tumor endothelial cell line (b.End5)

ABSTRACT: The aim of the project was to compare the secretomes of normal endothelial cells (NECs) and tumor endothelial cells (TECs). Cells were grown in full growth media up to 80-90% confluence. After washing with PBS, cells were incubated for 24 hours in serum free medium to collect conditioned medium, which was filtered and processed for LC-MS/MS analysis.

Background
Metastasis is a major contributor of cancer-related mortality and involves complex crosstalk between cancer cells and stromal cells modulated by various cytokines and growth factors. While the endothelium is essential for supplying nutrients and oxygen, little is known about its role in metastasis initiation. Determining the effect of endothelial-derived angiocrine factors on cancer cells may explain the mechanisms regulating metastasis initiation.
Results
In this study ...[more]
