Targeting the UFL1-AKT Cascade Suppresses Triple-Negative Breast Cancer Progression

Project description:Triple-negative breast cancer (TNBC) is an aggressive and highly lethal disease with limited therapeutic options. While UFL1-mediated UFMylation has been implicated in various diseases, its role in TNBC remains unexplored. In this study, we demonstrate that AKT1 directly interacts with UFL1 and undergoes UFMylation at Lys189, Lys276, and Lys297. This modification enhances AKT phosphorylation and activation, promoting tumor growth and chemoresistance in TNBC. In turn, AKT phosphorylates UFL1 at Thr426, establishing a positive feedback loop that sustains high activity of both pro-oncogenic regulators in TNBC. Disrupting the UFL1-AKT interaction using the specific peptide PDAU-TAT significantly inhibits TNBC progression both in vitro and in vivo. Clinically, elevated pT426 UFL1 expression significantly correlates with increased pAKT levels in TNBC specimens. These findings uncover a crucial positive feedback loop between UFL1 and AKT that drives TNBC progression and suggest that targeting the UFL1-AKT axis could offer a promising therapeutic strategy for TNBC and potentially other aggressive cancers characterized by upregulated UFL1 and AKT activation.

LC-MS/MS analysis of ribosome UFMylation in vitro

Project description:UFMylation, is a Ubiquitin-like modification occurring on RPL26, a ribosomal subunit, upon ribosome stalling at the ER membrane. This modification is achieved by a pool of enzymes commonly referred to as E1-activating, E2-conjugating and E3-ligase enzymes. The UFM1 E3 ligase comprises of three proteins: UFL1, UFBP1 and CDK5RAP3, referred to as UFM1 Ribosome E3 Ligase (UREL) complex. While UFL1 and UFBP1 are sufficient for E3 ligase activity in vitro, CDK5RAP3 is indispensable for ribosome UFMylation in vivo. In this work, we characterize in vitro UFMylation reaction of purified 60S ribosomes in the presence and absence of CDK5RAP3 using LC-MS/MS. Specifically, we analyse sites of UFMylation, linkage type and quantify monoUFMylation and diUFMylation on RPL26. Interestingly, we find UFMylation to occur on a specific lysine residue, K134, in the presence of UFL1/UFBP1 alone and in complex with CDK5RAP3. In addition, we also provide quantitative information on relative abundances of RPL26 mono- and di-UFMylation under these conditions.

Mechanism of 60S ribosomal subunit recognition and UFMylation by the UFM1 E3 ligase complex

Project description:During co-translational translocation at the endoplasmic reticulum (ER), ribosomes can stall and become covalently modified with the ubiquitin-like protein UFM1 on the 60S ribosomal subunit RPL26 (uL24). This process, known as UFMylation, is mediated by the UFM1 Ribosome E3 Ligase (UREL) complex, comprised of UFL1, UFBP1, and CDK5RAP3. However, the functional consequences of UFMylation and catalytic mechanisms of UREL are unknown. Here, we present crosslinking-mass spectrometry (XL-MS) data of UREL bound to 60S ribosomes.

Project description:DUSP22 (also named JKAP) is a dual-specificity phosphatase that inhibits T cell activation. Here we identified the E3 ubiquitin ligase UBR2 as an upstream activator of Lck during T-cell activation. JKAP dephosphorylated UBR2 at two residues, leading to ubiquitin-mediated UBR2 degradation. The SCF (SKP1-CUL1-βTrCP) complex induced UBR2 Lys48-linked ubiquitination at three lysine residues. Moreover, single-cell RNA sequencing analysis and UBR2 knockout showed that UBR2 increased proinflammatory cytokines. Remarkably, UBR2 induced Lys63-linked ubiquitination of Lck at two lysine residues and subsequent Lck Tyr394phosphorylation/activation in TCR signaling. Conversely, TCR-induced Lck activation and JKAP knockout-enhanced inflammatory phenotypes were attenuated by UBR2 knockout. Consistently, the UBR2-Lck interaction and Lck Lys63-linked ubiquitination were induced in peripheral blood T cells of human SLE patients. Collectively, UBR2 protein stability and UBR2-induced Lck ubiquitination/activation are inhibited by JKAP, leading to attenuation of T-cell activation and T-cell-mediated inflammation.

Ufl1 deficiency causes skin pigmentation by u-pregulation of Endothelin-1

Project description:Ufmylation (UFM1 modification) is a newly identified ubiquitin-like modification system involved in numerous cellular processes. However, the regulatory mechanisms and biological functions of this modification remain mostly unknown. We have recently reported that Ufmylation family genes have frequent somatic copy number alterations in human cancer including melanoma, suggesting involvement of Ufmylation in skin function and disease. UFL1 is the only known Ufmylation E3-like ligase. In this study, we generated the skin-specific Ufl1 knockout mice and show that ablation of Ufl1 caused epidermal thickening, pigmentation and shortened life span. RNA-Seq analysis indicated that Ufl1 deletion resulted in upregulation of the genes involved in melanin biosynthesis. Mechanistically, we found that Endothelin-1 (ET-1) is a novel substrate of Ufmylation and this modification regulates ET-1 stability, and thereby deletion of Ufl1 upregulates the expression and secretion of ET-1, which in turn results in up-regulation of genes in melanin biosynthesis and skin pigmentation. Our findings establish the role of Ufl1 in skin pigmentation through Ufmylation modification of ET-1 and provide opportunities for therapeutic intervention of skin diseases.

DNA microarray studies of tumors grown in nude mice from breast cancer cells MDA-MB-231 overexpressing or not the angiotensin II AT2 receptor, and treated or not with AT2 agonist compound C21

Project description:This experiment aims at investigating the effect of AT2 overexpression and/or activation in breast cancer. Human breast cancer cells MDA-MB-231 were transfected with lentiviral construct containing flagged human AT2 receptor sequence (or empty vector) and were subcutaneously injected into immuno-deficient mice (2. 10e6 cells). When the tumors reached 5 mm3 volume, mice were treated daily with compound C21 (0.3 mg/kg) or vehicle for 43 days. Total RNA was extracted from tumors and submitted to DNA array analysis.

Stable Cas9 Expression on Cell Growth Regulation Through mTORC2 Activation Facilitation

Project description:CRISPR, widely used for gene editing, relies on bacterial endonucleases like Cas9 to study gene functions and develop therapies. However, its potential effects on mammalian cellular responses and behavior remain unclear. Here, we systematically profiled effects of stable Cas9 expression on growth of 30 cell lines spanning 10 cancer types, observing growth alterations in 12 of them. To uncover the underlying mechanisms, we established the SpCas9 interactome in DU145 and MDA-MB-231 cells, both of which exhibited Cas9-increased growth phenotypes, and identified ribosomal proteins as the top shared interactors. Mechanistic studies revealed that ribosomal proteins such as RPL26 and RPL23a bind Sin1, a key mTORC2 component, relieving Sin1-PH-mediated suppression of the mTOR kinase domain leading to mTORC2 activation. Notably, SpCas9 interacts with both RPL26/RPL23a and Sin1, acting as a "glue" to stabilize their association and enhances mTORC2 activation, even in the absence of growth factors. Our study systematically characterizes Cas9’s effects on cell growth regulation and uncovers a novel Cas9-ribosome-mTORC2 signaling axis that promotes cell growth. These findings underscore the need to consider unintended cellular effects in CRISPR applications and highlight the importance of engineering safer Cas9 variants for biomedical research and clinical therapies.
