Ku80 interaction proteins under enhanced mitophagy

ABSTRACT: We reported Ku80 as a critical mitophagy protein and closely related to chemotherapy efficacy in liver cancer. The project detects the Ku80-interacted proteins under the condition of mitophagy activator or exogenously overexpression of Ku80 gene. After immunoprecipitation pulldown via anti-Ku80 or anti-flag antibody, LC MS/MS was performed to determine Ku80-interacted mitophagy-related proteins.

Project description:The spatial co-presence of aberrant long non-coding RNAs (lncRNAs) and abnormal coding genes contributes to malignancy development in various tumors. However, precise coordinated mechanisms underlying this phenomenon in tumorigenesis remains incompletely understood. Here, we show that Prohibitin 2 (PHB2) orchestrates the transcription of an oncogenic CASC15-New-Isoform 2 (CANT2) lncRNA and the coding tumor-suppressor gene CCBE1, thereby accelerating melanoma tumorigenesis. In melanoma cells, to further capture the binding proteins of CANT2, ChIRP-MS was performed.

Project description:Ticks are hematophagous vectors of great medical and veterinary importance because they transmit numerous pathogenic microorganisms to humans and animals. The argasid Ornithodoros lahorensis is the vector of tickborne Anaplasma ovis,Crimean-Congo haemorrhagic fever virus and other pathogenic. Tick salivary gland express bioactive molecules that perform key functions in blood feeding and pathogen transmission. To explore new strategies for tick control, in this work we have obtained and compared the salivary gland proteomics of O.lahorensis before and after a blood meal and identified the DEPs regulated after feeding. The proteins were annotated, functionally and their expression levels compared between both physiological conditions (unfed females/males and fed females/males at 24h post-engorgement).

LC-MS of interacting partners Toxoplasma gondii GRA57

Project description:We identified GRA57 as a Toxoplasma gondii secreted effector required for parasite survival of interferon gamma in HFFs. To identify interaction partners of GRA57 during infection of HFFs, which could indicate how GRA57 functions to protect the parasite, we performed co-immunoprecipitation (IP) experiments with an endogenously tagged RHGRA57-HA strain.To enable detection of protein-protein interactions specific to activated cells, HFFs were pre-stimulated before infection with 2.5 U/ml IFN for 6 h, which we found was sufficient to induce IFN responses while retaining host cell viability. HFFs were infected for 24 h with either RH∆Ku80 or RHGRA57-HA parasites prior to lysis. GRA57-HA was immunoprecipitated from lysates, then co-immunoprecipitated proteins were identified by liquid chromatography (LC)-tandem mass spectrometry (MS/MS).

AP2X-1 is a negative regulator of Toxoplasma gondii sexual commitment

Project description:Toxoplasma gondii is a widespread protozoan with a complex life cycle, characterized by transitions between various hosts and developmental stages, each tailored to a specific niche within its host. However, the regulatory mechanisms governing these life cycle transitions are not well understood. In this study, we investigated the AP2 factor AP2X-1, which is expressed during the tachyzoite and bradyzoite stages but decreases in the mature merozoite stage. Knockout of ap2X-1 significantly impaired tachyzoite invasion and replication while increasing the frequency of bradyzoite differentiation. As a component associated with the HDAC3/MORC complex, loss of ap2X-1 led to the upregulation of bradyzoite- and sexual stage-specific genes. Single-cell sequencing revealed that ap2X-1 knockout strain exhibited a mixed population of tachyzoite-, bradyzoite-, merozoite-, and sporozoite-like parasites. CUT&Tag analysis revealed a substantial overlap between AP2X-1 and HDAC3/MORC complex at the promoters of bradyzoite- and sexual stage-specific genes. Additionally, ATAC-seq analysis demonstrated that AP2X-1 influences chromatin compaction and accessibility, suggesting that AP2X-1 may modulate the function of the HDAC3/MORC complex to facilitate the repression of bradyzoite differentiation and sexual commitment. Loss of ap2X-1 resulted in a significant attenuation of T. gondii virulence and decreased brain cyst formation in vivo. These findings identify AP2X-1 as a critical negative regulator of T. gondii sexual development.

MS-based proteomic characterization of Toxoplasma gondii GSK3 proxisome

Project description:In an effort to identify novel drug candidates against T. gondii parasites, an interesting compound was identified, allowing to inhibit tachyzoite growth without impacting the HFF monolayer. A forward genetic screen identifies TgGSK3 as the enzyme targeted by this molecule. To investigate the biological function of TgGSK3 and its regulatory mechanisms, we performed in vivo biotinylation using the proximity labeling approach employing BioID2 with TgGSK3 as the bait protein. To identify candidate interacting partners or substrates of TgGSK3, biotinylated proteins were enriched from lysates using streptavidin affinity purification. As a control, we conducted a mock experiment with parasites that do not express BioID2 fusion protein (RH ku80).

Proteasome beta2 and beta5 subunit inhibition by carfilzomib causes acute contractility impairment due to dysbalance of calcium homeostasis

Project description:Treatment of multiple myeloma (MM) with the second-generation proteasome inhibitor (PI) carfilzomib (CFZ) is associated with higher incidence of cardiovascular adverse events compared to first-generation PI bortezomib (BTZ). CFZ and BTZ inhibit at lower doses only the β5 proteasome subunit, whereas they differ in co-inhibition of other proteasome subunits at the higher doses: BTZ co-inhibits β1 subunit, whereas CFZ co- inhibits β2 subunit. This different pattern of inhibition affects differently the degree of functional proteasome inhibition with β5+β2 showing more effective functional proteasome inhibition. It is unclear, whether CFZ-induced cardiotoxicity is the result of proteasome inhibition, or an off-target effect. With an unbiased multi-omics approach in conjunction with proteasome subunit-specific inhibitors, we show that CFZ-type β5+β2 proteasome subunit inhibition, in contrast to the BTZ-type β5+β1 inhibition, directly interferes with cardiomyocyte contractility in vitro and in vivo. For the in vivo analysis, we used 6-8 weeks old Balb/c mice that were exposed to the drugs for 1h and subsequently euthanized. The hearts were processed for LC/MS-MS analysis. The hearts from CFZ-treated mice for 1h show showed impaired accumulation of proteins related to calcium handling (Atp2a2) and retinoid pathway (Aldh1a1), which is prevented by atRA (1 mg/kg) co-treatment. Our data suggests a novel mechanism for PI-induced cardiotoxicity that reflects clinical findings in which CFZ-type β5+β2 proteasome inhibition, in contrast to β5+β1 co-inhibition of BTZ, directly interferes with the contractile activity of cardiomyocytes.

MS-based proteomic characterization of Toxoplasma gondii PRP4K and PRP8 interactomes

Project description:The apicomplexa comprise a large phylum of single-celled, obligate intracellular protozoa that infect humans and animals and cause severe parasitic diseases. Available therapeutics against these devastating diseases are limited by suboptimal efficacy and frequent side effects, as well as the emergence and spread of resistance. Here, we use a drug repositioning strategy and identify altiratinib, a compound originally developed to treat glioblastoma, as a promising drug candidate with broad spectrum activity against apicomplexans. Altiratinib is parasiticidal and blocks the development of intracellular zoites in the nanomolar range and with a high selectivity index. We have identified TgPRP4K of T. gondii as the primary target of altiratinib by genetic target deconvolution. TgPRP4K is phylogenetically related to the cyclin-dependent-like kinase family (CLK) and its closest ancestor in humans is the splicing factor kinase PRP4 kinase (PRP4K or PRPF4B) and in P. falciparum is PfCLK3 (PF3D7_1114700), a kinase that has been identified as a multistage cross-species antimalarial drug target. We found an altiratinib-resistant parasite line has a wild-type (WT) allele of TgPRP4K and a mutation E1325K in TgPRP8, a protein located in the catalytic core of the spliceosome that has been shown to interact with PRP4K in Schizosaccharomyces pombe to facilitate spliceosome activation. This reinforces the possibility that the PRP4K-PRP8 complex is at the basis for the anti-Toxoplasma activity of altiratinib. To gain insight about the role of both proteins in T. gondii, we characterized their interactomes.
