Project description:Gametophytic self-incompatibility is the primary cause of low fruit set in almond. The mechanism of recognition that determines whether the gametophyte is successfully fertilized between pollen tube SCF (F-box-SSK1-Cul1-Rbx1) protein and pistil S-RNase protein during fertilization is unclear. In this study, the pistils of two almond cultivars 'Wanfeng' and 'Nonpareil' were used as the experimental materials after selfing- and nonselfing/cross-pollination, and pistils from the stamen-removed flowers were used as controls. We used fluorescence microscopy to observe the development of pollen tubes after pollination and 4-dimensional label-free quantitation (4D-LFQ) to detect the protein expression profiles of 'Wanfeng' and 'Nonpareil' pistils and in controls. The results showed that it took 24-36 h for the development of the pollen tube to 1/3 of the pistil, and a total of 7684 differentially accumulated proteins (DAPs) were identified in the pistil after pollinating 36 h, of which 7022 were quantifiable. The up- and down-regulation of the 12 differential protein expressions identified by parallel reaction monitoring (PRM) was the same as that identified by 4D-LFQ, with an average fold-change difference of 14.34%, a maximum of 31.37% and a minimum of 3.68%. Bioinformatics analysis based on the function of the differential proteins, including classification, enrichment, and clustering, identified RNA polymerases (4 DAPs), autophagy (3 DAPs), oxidative phosphorylation (3 DAPs), and homologous recombination (2 DAPs) pathways associated with the self-incompatibility process. The interaction between the serine/threonine kinase (MARK2) protein E3 ubiquitin ligase and the microtubule protein component microtubule-associated protein tau (MAPT/ACT) was found using the STRING database, which demonstrated the involvement of the MARK2 protein in the reaction of pollen tube recognition the nonself- and the self-S-RNase protein. It provides a new way to reveal the mechanism by which almond pollen tubes recognize the self and nonself S-RNase enzyme protein.

Project description:Coiled-coil domain-containing 68 (CCDC68) plays different roles in cancer and is predicted as a tumor suppressor in human colorectal cancer (CRC). However, the specific role of CCDC68 in CRC and the underlying mechanisms remain unknown. Here, we showed that CCDC68 expression was lower in CRC than in corresponding normal tissues, and CCDC68 level was positively correlated with disease-free survival. Ectopic expression of CCDC68 decreased CRC cell proliferation in vitro and suppressed the growth of CRC xenograft tumors in vivo. CCDC68 caused G0/G1 cell cycle arrest, downregulated CDK4, and upregulated ITCH, the E3 ubiquitin ligase responsible for CDK4 protein degradation. This increased CDK4 degradation, which decreased CDK4 protein levels and inhibited CRC tumor growth. Collectively, the present results identify a novel CDK4 regulatory axis consisting of CCDC68 and ITCH, which suggest that CCDC68 is a promising target for the treatment of CRC.

Project description:This study aims to analyze and compare the post-translational modification differences between human plasma- and genetic recombination-derived human serum albumin (HSA) in clinical treatments in China. 6 different post-translational modifications, including acetylation, succinylation, crotonylation, phosphorylation, beta-hydroxybutyrylation, and lactylation, were detected using pan-specific antibodies through Western blot. The samples, consisting of human plasma-derived HSA (pHSA) from six different manufacturers and recombinant HSA (rHSA) expressed in yeast and oryza sativa, underwent detection of types of post-translational modifications. 4D label-free PTMs quantitative proteomic analysis was conducted to detect N-glycosylation and the above-mentioned post-translational modifications in pHSA and rHSA samples, and analyze the differences in modification sites and levels. Through Western blot analysis, all six pHSA and two rHSA samples were positive in the band of albumin (66.5 kDa) for the six post-translational modifications. Further analysis using 4D label-free PTMs quantitative proteomics revealed 25 (29) acetylated, 30 (32) succinylated, 41 (50) malonylated, 15 (23) phosphorylated, 36 (30) beta-hydroxybutyrylated, and 27 (34) lactylated modification sites in pHSA and rHSA samples and no N-glycosylation modification sites. The analysis results identified 1 acetylation (ALB_K160), 2 beta-hydroxybutyrylation (ALB_K569, ALB_K426), 3 crotonylation (ALB_K264, ALB_K581, ALB_K560), and 15 phosphorylation specific modification sites in pHSA, as well as 3 crotonylation (ALB_K560, ALB_K562, ALB_K75), 1 succinylation (ALB_K490), and 23 phosphorylation specific modification sites in rHSA. In pHSA (rHSA), 2 (6) acetylation, 10 (12) succinylation, 0 (9) crotonylation, 1 (9) phosphorylation, 6 (0) beta-hydroxybutyrylation, and 0 (7) lactylation specific modification sites were found. Additionally, in the common modification sites of pHSA and rHSA, pHSA showed up-regulation of amberylation (16:1) and beta-hydroxybutyrylation (12:2) in more sites, and up-regulation of acetylation (7:11), crotonylation (2:11), phosphorylation (1:8), and lactylation (1:14) in less sites compared to rHSA. In clinical treatment, the used pHSA and rHSA in China generally exhibit acetylation, succinylation, crotonylation, phosphorylation, beta-hydroxybutyrylation, and lactylation, and there are differences in the site characteristics and modification levels of these modifications between pHSA and rHSA. Further experimental investigations are needed to explore the impact of post-translational modification differences on the biological function, efficacy, and safety of pHSA and rHSA.

Project description:Oyster plasma contains various soluble factors secreted by hemocytes and other cells, which were separated from the haemolymph and collected for antibacterial activity as-says. Plasma was collected after V. parahaemolyticus injection, with PBS injection being set as a control. Plasma of the V. parahaemolyticus challenged group was markedly more bac-tericidal than that of the PBS treated control。Peptidomics was herein employed to identify new bioactive peptides with antibacterial activity from plasma based on liquid chromatography and tandem mass spectrometry. To apply large-scale peptidomics to oyster plasma proteins, oyster plasma was extracted from the V. parahaemolyticus challenged group (P4, P5, P6) and PBS treated control (P1, P2, P3).

Project description:ANCA associated vasculitis(AAV) is a systemic vasculitis of small vessels, characterzied by injury of vascular endothelial cells induced by abnormally activated neutrophils. Microscopic polyangiitis (MPA) is a member of AAV with a high risk of kidney involvement. Blood lipid disorder promotes endothelial injury. We aim to investigate the correlation between blood lipid levels and renal prognosis in MPA patients. Firstly, we retrospectively included 110 patients diagnosed with MPA and the primary endpoint was the occurrence of ESRD. The association between blood lipids and renal outcome was evaluated with logistic regression analysis and survival analysis. During a median follow-up period of 23 months, 44 out of 110 patients (40%) developed ESRD. High serum triglycerides (TG) and VLDL at diagnosis were associated with ESRD development after adjusting for several confounding factors. MPA patients with TG >1.45 mmol/L or VLDL > 0.66mmol/L had significantly higher risk of ESRD development than those with TG ≤1.45 mmol/L or VLDL ≤0.66 mmol/L. Secondly, we explored the underlying mechanism of poor renal prognosis in patients with high TG levels using data independent acquisition (DIA) quantitative proteomics. DIA quantitative proteomics analysis suggested that patients in high TG group had up-regulated profibrotic pathway, inflammatory signaling pathways and complement and coagulation cascades compared with those in low TG group. In conclusion, high baseline TG or VLDL is associated with an increased risk of ESRD development in MPA patients. The potential mechanisms may be the upregulation of pro-fibrotic and inflammatory signaling pathways, and the activation of complement and coagulation cascades.

Project description:Neurobrucellosis is a serious complication of brucellosis, which poses a significant threat to human health and quality of life. The origin of this phenomenon is that the patients are more vulnerable to the development of chronic Brucella infection, which is also an important clinical problem. The regulation of Brucella on microglia apoptosis is a primary contributor to chronic neurobrucellosis, nevertheless, the precise molecular mechanism remains incompletely understood. In our research, human microglial clone 3 (HMC3) cells were infected with Brucella suis vaccine strain S2 (B. suis S2) at different MOI for different times to evaluate the influences of B. suis S2 on the IRE1/Caspase-12/Caspase-3 pathway. Subsequently, under the conditions of IRE1/Caspase-12/Caspase-3 pathway was suppressed by B. suis S2, CALR protein was screened by ubiquitin-modified proteomics. Eventually, we infected CALR-overexpression and -knockdown HMC3 cell lines with B. suis S2 to elucidate the molecular mechanism by which the B. suis S2 inhibited HMC3 cells apoptosis. In summary, we confirm that B. suis S2 inhibits apoptosis in HMC3 cells mediated by the IRE1/Caspase-12/Caspase-3 pathway through modulation of CALR ubiquitination. Our study establishes the theoretical foundation for investigating the mechanisms underlying neurobrucellosis and provides a guidance for the clinical treatment of neurobrucellosis.
