PSME3 Regulates Migration and Differentiation of Myoblasts

ABSTRACT: This is the second of two experiments in this project. It sought to measure the changes in the proteome following depletion of PSME3. C2C12 cells were depleted of PSME3 using specific siRNA. Controls received siRNA with a scrambled sequence. Cells were grown to confluency (Day 0 of differentiation), harvested, and the whole cell lysate was analyzed by mass spectrometry.

Project description:This is the first of 2 proteomics experiments in the same project (see https://www.biorxiv.org/content/10.1101/2025.01.08.632048v1.full). This one is a co-Immunoprecipitation experiment aimed at determining the interaction partners of proteasome regulator PSME3. PSME3 was captured from C2C12 cell lysates using a PSME3-targeting antibody or an IgG control at several timepoints of serum starvation-induced differentiation (Cycling, Day 0, and Day 1). The immunoprecipitate was analyzed by mass spectrometry.

Shared behavioural impairments in visual perception and place avoidance across different autism models are driven by periaqueductal grey hypoexcitability in Setd5 haploinsufficient mice.

Project description:Despite the diverse genetic origins of autism spectrum disorders (ASDs), affected individuals share strikingly similar and correlated behavioural traits that include perceptual and sensory processing challenges. Notably, the severity of these sensory symptoms is often predictive of the expression of other autistic traits. However, the origin of these perceptual deficits remains largely elusive. Here, we show a recurrent impairment in visual threat perception that is similarly impaired in three independent models of ASD with different molecular aetiologies. Interestingly, this deficit is associated with reduced avoidance of threatening environments - a non-perceptual trait. Focusing on a common cause of ASDs, the Setd5 gene mutation, we define the molecular mechanism. We show that the perceptual impairment is caused by a potassium channel (Kv1) mediated hypoexcitability in a subcortical node essential for the initiation of escape responses, the dorsal periaqueductal grey (dPAG). Proteomic profiling of tissue samples across different brain regions didn't show a perturbation in Kv1 levels in mutant mice. Targeted pharmacological Kv1 blockade rescued both perceptual and place avoidance deficits, causally linking seemingly unrelated trait deficits to the dPAG. Furthermore, we show that different molecular mechanisms converge on similar behavioural phenotypes by demonstrating that the autism models Cul3 and Ptchd1, despite having similar behavioural phenotypes, differ in their functional and molecular alteration. Our findings reveal a link between rapid perception controlled by subcortical pathways and appropriate learned interactions with the environment, and define a non-developmental source of such deficits in ASD.

Project description:RNA-seq analysis for C2C12 myoblasts knocked down for either only-BRG1 or only-BRM or both BRG1 and BRM. Cells were treated with an siRNA SMARTpools of four oligos each targeting either BRG1 (siGENOME mouse Smarca4 pool #M-041135-01-0020) or BRM (siGENOME mouse Smarca2 pool #M-056591-00-0020) or both. Control samples were treated with non-targeting scrambled oligos (siRNA SMARTpool ON-TARGETplus scrambled non-targeting pool # D-001810-10-20). siRNA pools were purchased from Dharmacon, Horizon Discovery Ltd., USA.

Carnitine O-octanoyltransferase is a novel inducer of vascular calcification via promoting fatty acid metabolism and mitochondrial dysfunction

Project description:Objective – Vascular calcification is a critical pathology associated with increased cardiovascular event risk, but there are no FDA-approved anti-calcific therapies. We hypothesized and validated that an unbiased screening approach would identify novel mediators of human vascular calcification. Approach and Results – We performed an unbiased quantitative proteomics and pathway network analysis that identified increased carnitine O-octanoyltransferase (CROT) in calcifying primary human coronary artery smooth muscle cells (SMCs). Additionally, human carotid artery atherosclerotic plaques contained increased immunoreactive CROT near calcified regions. CROT siRNA reduced fibrocalcific response in calcifying SMCs. In agreement, histidine 327 to alanine point mutation inactivated human CROT fatty acid metabolism enzymatic activity and suppressed SMC calcification. CROT siRNA restored mitochondrial proteome alterations and suppressed mitochondrial fragmentation in calcifying SMCs. Lipidomics analysis of SMCs incubated with CROT siRNA revealed increased eicosapentaenoic acid, a vascular calcification inhibitor. CRISPR/Cas9-mediated Crot deficiency in low-density lipoprotein receptor-deficient mice reduced aortic and carotid artery calcification without altering bone density, or liver and plasma cholesterol and triglyceride concentrations. Conclusions – CROT is a novel inducer of vascular calcification via promoting fatty acid metabolism and mitochondrial dysfunction, as such CROT inhibition has strong potential as an anti-fibrocalcific therapy.
