Lactoferrin-Osteopontin Complexes: Insights into Intestinal Organoid Bioavailability and Gut Microbiota Modulation

ABSTRACT: Lactoferrin (LF) and osteopontin (OPN) are key bioactive milk proteins with significant immune-modulatory, gut, and systemic health benefits. Using a microarrayed high-throughput 3D intestinal organoid platform, we evaluated the bioavailability and uptake of bovine LF-OPN soluble complexes (SC) and complex coacervates (CC) in vitro.

Project description:During the resolution of inflammation macrophages engulf apoptotic polymorphonuclear cells (PMN) and can accumulate large numbers of their corpses. Here we report that resolution-phase-macrophages acquire the neutrophil-derived glycoprotein lactoferrin (Lf) in vivo and ex vivo and process it to short fragments. During the onset and resolving phases of inflammation in murine peritonitis and bovine mastitis Lf fragments of 15 and 17 kDa occurred in various body fluids, and the murine fragmentation and release were mediated by macrophages. The 17 kDa fragment contained two bioactive tripeptides, FKD and FKE that promoted human macrophage reprogramming to a pro-resolving phenotype. At low concentrations (1-10 M) this was reflected by inhibition of LPS-induced TNFand IL-6 secretion and increased IL-10 levels. Both peptides inhibited ERK and cJun activation following macrophage exposure to LPS. In addition, FKD, and to a lesser extent FKE, promoted neutrophil-mediated resolution at high concentrations (30-100 M) by enhancing the formation of cytokine-scavenging aggregated NETs at low cellular density. Transcriptome and network analyses in Saccharomyces cerevisiae reveal that amphotericin B and lactoferrin synergy disrupt metal homeostasis and stress response

Project description:Invasive fungal infections (IFIs) are difficult to treat. Few effective antifungal drugs are available and many have problems with toxicity, efficacy and drug-resistance. To overcome these challenges, existing therapies may be enhanced using more than one agent acting in synergy. Previously, we have found amphotericin B (AMB) and the iron chelator, lactoferrin (LF), were synergistic against Cryptococcus neoformans and Saccharomyces cerevisiae. This study investigates the mechanism of AMB+LF synergy, using RNA-seq and network analyses. Genes involved in iron homeostasis showed increased expression upon treatment with AMB alone. Unexpectedly, AMB+LF treatment did not lead to increased expression of iron or zinc homeostasis genes however we observed decreased expression of oxidative stress response genes. Addition of iron or zinc to AMB+LF treated cells did not rescue the synergy, supporting the likelihood that the mechanism of synergy involves more than iron and zinc chelation. We clustered genes based on patterns of co-expression and found by network analysis that many genes involved in iron and zinc homeostasis, which have dysregulated expression upon AMB+LF treatment, are targets of transcription factors Aft1p and Zap1p. Hypothesizing that these might play a key role in the synergistic response, knock-out mutants of Aft1 and Zap1 were tested for increased sensitivity to AMB and oxidative stress. Both mutants showed hypersensitivity towards these treatments. Our results suggest the mechanism of AMB+LF synergy involves disruption to oxidative stress response, in addition to chelation of iron and zinc. Since Zap1 is conserved in C. neoformans and contains a putative drug binding domain, we suggest novel Zap1 binding molecules could be combined with existing antifungals to serve as synergistic antifungal treatments for this species.

The lncRNA expression profile along with mRNA expression profile of 7 control and 7 LF livers were analyzed by Arraystar Mouse LncRNA Microarray V3.0. One liver per array.

This study was carried out to identify and compare signaling networks provoked by the two LF isoforms. To do this, the two forms were overexpressed in HEK293 cells using the flp-in/tet-on system and genome-wide expression analysis of 18,367 genes was conducted.

An Oleuropein-based olive leaf extract enhances the muscle mitochondrial response to acute moderate – but not maximal – intensity exercise in humans

Project description:Sprint interval exercise (SIE) induces mitochondrial adaptations in skeletal muscle, comparable to or greater than moderate-intensity continuous exercise (MICE), despite lower training volume. These effects are partly due to enhanced mitochondrial bioenergetics, including increased Ca²⁺ uptake and pyruvate dehydrogenase (PDH) activation. Oleuropein (OLE), a compound from olive leaf extract, also promotes mitochondrial Ca²⁺ uptake and PDH activation in mice. The current study tested whether OLE could enhance these adaptations in humans during MICE or SIE. In a double-blind crossover design, healthy males performed either MICE or SIE with OLE supplementation. To elucidate the acute molecular response of skeletal muscle following exercise, we performed mass spectrometry-based proteomic analysis.

This is a pilot submission of mass-spectrometry analyses from 18 induced pluripotent stem cell lines generated by the HipSci project. This submission includes also data for two embryonic stem cell lines, and one reference sample comprising a mixture of 42 IPSC lines.
