Project description:Crosstalk between cell death programs confers appropriate host anti-infection immune responses, but how pathogens remodel this network to facilitate their infections remain largely unclear. Here, we identify mammalian cell entry 3C (Mce3C) as a key regulator of host cell death from Mycobacterium tuberculosis (Mtb), which causes tuberculosis featured with lung inflammation and necrosis. Mce3C binds host lysosomal protease cathepsin B (CTSB), which acts as a decision-maker of cell death modalities, to inhibit the protease activity of CTSB towards BH3-interacting domain death agonist (BID) and receptor-interacting protein kinase 1 (RIPK1), thus preventing the production of pro-apoptotic truncated BID (tBID) while maintaining the abundance of pro-necroptotic RIPK1. Disrupting the Mce3C-CTSB interaction promotes host apoptosis and suppresses necroptosis with attenuated Mtb survival and mitigated lung inflammation in mice. These findings reveal a unique strategy by which the pathogen manipulates host lysosomal protease activity-dependent plasticity between cell death pathways to promote infection and pathogenicity.

Project description:human blood monocytes were isolated, activated and harvested at several timepoints In this study, we identified genes that were differentially expressed in human monocytes activated with eiter NOD2L and/or TLR2/1L. human blood monocytes were purified from healthy donors by Ficoll, Percoll and adherence. Monocytes were activated using NOD2L (MDP) and the TLR2/1L (19kD, triacylated peptide). Cells were harvested before activation (0h) and 6h and 24h after stimulation with ligands.

Project description:The BF-3 pocket of the androgen receptor (AR) has been identified as an allosteric modulator of the transactivation function of the AR. We now demonstrate that a duplicated GARRPR motif at the N-terminus of the cochaperone Bag-1L functions through this BF-3 domain. Amino acid exchanges in these two motifs impair binding of Bag-1L to the AR but increase the androgen-dependent activation of a subset of AR-target genes. We have therefore identified GARRPR as a novel BF-3 regulatory sequence important for fine-tuning the activity of the receptor. LNCaP cells stably expressing the empty vector construct (=control), or wild-type or N-terminal GARRPR mutant Bag-1L were cultured under hormone-starvation conditions for 72 h and then treated with vehicle or 10 nM DHT for 4 h. Biological triplicate samples were analyzed for each cell line.

The database utilized for this purpose was the Canadian deer protein database, which was downloaded from NCBI [Cervus_canadensis_GCF_019320065.1_protein.faa]. The software employed for searching was MaxQuant version 2.1.4.0. Gene Ontology and KEGG Pathway enrichment analysis were conducted using the EBI database and InterProScan software version 5.31-70.0.

Project description:Pathogenic mycobacteria, such as Mycobacterium tuberculosis, modulate the host immune system to evade clearance and promote long-term persistence, resulting in disease progression or latent infection. Understanding the mechanisms pathogenic mycobacteria use to evade the host immune system is critical to better understanding the pathogenesis of mycobacterial infection. Protein kinase G (PknG) in pathogenic mycobacteria has been shown to play an important role in avoiding clearance by macrophages through blocking phagosome-lysosome fusion; however, the exact mechanism is not completely understood. Here, to investigate the influence of mycobacterial PknG on the phosphoprotein signalling cascades triggered during early events prior to phagocytosis, RAW 264.7 macrophage cell lines were stimulated with M. bovis BCG wild-type and PknG knock-out mutant strains and harvested prior to uptake. After proteolysis, phosphopeptides were enriched via TiO2 columns and subjected to LC-MS/MS to identify differentially phosphorylated peptides between macrophages exposed to the wild-type and PknG knock-out mutant. A total of 1401 phosphosites on 914 unique host proteins were identified. Following phosphoproteome normalisation and differential expression analysis, a total of 149 phosphosites were differentially phosphorylated in the RAW 264.7 macrophages exposed to the wild-type versus the PknG knock-out mutant. A subset of 95 phosphosites was differentially up-regulated in the presence of PknG. In addition, a presence/absence analysis identified 34 phosphosites on 27 host proteins to be exclusively phosphorylated in the presence of PknG. Functional analysis of our data revealed that PknG kinase activity in mycobacteria primes an altered phosphorylation state in host macrophages prior to establishment of infection, through interfering with host spliceosomal and translational machinery, actin cytoskeletal organisation, pro-inflammatory cytokine induction, and programmed cell death, thus establishing a new role for PknG in directing the fate of mycobacteria in macrophage infections.

Project description:Metallosphaera sedula is an extremely thermoacidophilic archaeon that grows heterotrophically on peptides, and chemolithoautotrophically on hydrogen, sulfur, or reduced metals as energy sources. During autotrophic growth, carbon dioxide is incorporated into cellular carbon via the 3-hydroxypropionate /4-hydroxybutyrate cycle (3HP/4HB). To date, all of the steps in the pathway have been connected to enzymes encoded in specific ORFs, except for the one responsible for ligation of coenzyme A (CoA) to 4-hydroxybutyrate (4HB). While several candidates for this step have been identified through bioinformatic analysis of the M. sedula genome, none have been shown to catalyze this biotransformation. Transcriptomic analysis of cells grown under strict H2-CO2 autotrophy was used elucidate additional candidate genes involved in carbon fixation and identify the genes which encode for 4HB-CoA synthetase. Three slide loop for Mse cells includes 3 conditions tested in duplicate (biological repeats from tandem fermentors): autotrophic carbon limited (ACL), autotrophic carbon rich (ACR), and heterotrophic (HTR). Half of an RNA sample for one condition was labeled with Cy3 while the other half was labeled with Cy5. The two differently labeled samples were run on different slides. Each probe is spotted on each slide 5 times (5 replicates; spot intensities for all replicates on slide provided in associated raw data file).

Project description:Mycobacterium tuberculosis (Mtb) protein kinase G (PknG) is a critical protein which can inhibit the maturation of phagosome and its fusion with the lysosome, providing Mtb’s latency and survival in the host. However, its signaling pathways and functions in the human body are still unclear and needed to be further studied. In our research, we used the human protein microarray to study the proteins interacting with PknG and have found a list of proteins that are potentially related to PknG in the host, which may contribute to the infection of Mtb. Among these positive hits, we found human protein CypA has strong interaction with PknG. We further validated the interaction between PknG and CypA in vitro and in vivo and investigated its role in the infection of macrophage cells. Finally, we found that PknG can significantly down-regulate protein expression level of CypA through phosphorylation in site of T107 as well as its induction of inflammatory response through NF-κB and ERK1/2 pathways. Meanwhile, the survival of Mycobacterium smegmetis (Msm) in the macrophages with overexpressing PknG is also improved with lessen of cytokines expression. Our results provided a resourceful research base of PknG in regards to various target human proteins and offered an insightful understanding of PknG’s function in the host.
