Screening of targets modified with poly(ADP-ribose) by LC-MSMS

ABSTRACT: We used mass spectrometry-based proteomic identification to screen poly(ADP-ribosyl)ation (PARylation) targets. Af1521 is a PAR/MAR (mono(ADP-ribose))-binding module with strong affinity towards ADP-ribose. We used Af1521 to perform proteome-wide screening to identify protein that has been modified with poly(ADP-ribose) in human primary ECs, human umbilical vein endothelial cells (HUVECs). The binding-defective mutant (GST-Af1521-Mut) was used as control.
