A Microneedle Device for Rapid Dermal Interstitial Fluid Sampling

ABSTRACT: Dermal interstitial fluid (ISF) shows promise for novel skin diagnostics and as an alternative to invasive blood tests. This dataset includes ISF sampled via a microneedle device from two ex vivo human abdominal skin specimens and the forearms of ten healthy volunteers. Paired blood samples via finger prick were also collected from five volunteers. We studied the ISF proteome in relation to collection time and pressure ex vivo. We also created a reference ISF proteome from the healthy volunteers, compared it to the plasma proteome, and quantified errors from blood contamination.

Identification of candidate circulating glucocorticoid response biomarkers in humans using a proteomic approach

Project description:Glucocorticoids used in pharmacological doses for the treatment of a variety of medical conditions, and endogenous glucocorticoid excess – Cushing’s syndrome, may result in several adverse effects, but currently there is no clinically useful biomarker of glucocorticoid activity. A three-phase protein biomarker discovery strategy was used. Proteomic biomarker discovery and qualification was conducted on the secretome of ex vivo-stimulated peripheral blood mononuclear cells (PBMC) isolated from 6 volunteers, incubated ± dexamethasone 100 ng/mL for 4h and 24h. Untargeted proteomics with label-free quantification (LFQ) was conducted to discover candidate proteins which were quantified using targeted proteomics by a custom multiple reaction monitoring mass spectrometry (MRM-MS) assay. Five proteins were selected for serum measurement by immunoassay in 20 healthy volunteers, with blood drawn at baseline and 12h after 4 mg oral dexamethasone. Paired analysis of the discovery proteomics data (576 and 280 proteins for the 4h and 24h secretomes, respectively) generated a shortlist of candidates which were qualified using MRM-MS to obtain protein level intensity data for 39 proteins. In the validation cohort, 3/5 proteins were dexamethasone-responsive, two significantly decreased: lysozyme C (mean±SEM) – 101±5.5 vs 67±4.4 ng/mL, (P<0.0001); nucleophosmin-1 (median (interquartile range)) – 16.6 (14.4-18.4) vs 14.2 (11.1-17.4) ng/mL, (P<0.01), while high mobility group box 2 (mean±SEM) significantly increased – 819±34 vs 984±60 pg/mL (P<0.01). Using an ex vivo proteomic approach in PBMC, we have identified and verified circulating glucocorticoid-responsive proteins which may have potential as serum biomarkers.

Synthetic double stranded RNA reliably induces innate immunity similar to a live viral vaccine in humans

Project description:Adjuvants are critical for the success of vaccines, and agonists for microbial pattern recognition receptors are promising new candidates. A mechanism for the immune enhancing role of adjuvants is to stimulate innate immunity. We studied the innate immune response in humans to synthetic double stranded RNA (poly ICLC), a ligand for TLR3 and MDA-5 cytosolic RNA helicase. Transcriptional analysis of blood samples from eight volunteers, after subcutaneous administration of poly ICLC showed upregulation of genes involved in multiple innate immune pathways in all subjects, including interferon and inflammasome signaling. Blocking of type I interferon receptor ex vivo significantly dampened the response to poly IC. Comparative transcriptional analysis showed that several innate pathways were similarly induced in volunteers immunized with the highly efficacious Yellow Fever Vaccine. Therefore a chemically defined microbial agonist like poly ICLC can be a reliable and authentic microbial mimic for inducing innate immunity, here for a live attenuated viral vaccine in humans. Project description:Baseline gene expression in circulating peripheral blood mononuclear cells isolated from 18 apparently healthy volunteers (10 females and 8 males). The volunteers fasted overnight for 10 hours prior to collection of blood samples (but they were allowed water). 5 blood samples were collected from each volunteer at consecutive 8-day intervals. Volunteers were included in this study provided they met the following selection criteria, that they should be ?Aged between 20 and 45 years of age; ?A non-smoker (or an ex-smoker); ?Not pregnant or been pregnant in the last 12 months ?Not diagnosed with any long-term medical condition; ?Or have not (in the previous 4 months) donated blood, received any inoculations and are not taking regular prescribed medicine (including HRT and oral contraception) Prior to inclusion in this study volunteers were screened for general health and had a blood sample taken for routine screening of Hb content etc. Only those volunteers who were in good general health were allowed to participate in the study. The age of all volunteers has been reported here as being 20-45 years in order to preserve the anonymity of the data, and so that the individuals involved may not be able to track down their own specific data. Lineage relationship in human dendritic cells

Project description:The lineage relationships and fate of human blood and tissue dendritic cells (DC) has significance for a number of diseases including HIV where both blood and tissue DC may be infected. We used gene expression profiling of monocyte and DC sub-populations sorted directly from blood and skin and compared this to monocyte derived DC (MDDC) and MUTZ3 Langerhans cells (LCs) to define the lineage relationships. Hierarchical clustering analysis showed that plasmacytoid DCs formed the most discrete cluster. The ex vivo derived myeloid cells formed two separate clusters of cells derived from blood, and skin. Separate and specific DC populations could be determined within the sub-clusters. During overnight culture CD14+ dermal DCs (DDC) converted to CD1a+ expressing cells in situ consistent with origin of the CD1a+ DDC from a local precursor rather than from circulating blood DC or monocyte precursors. The in vitro derived MDDC and MUTZ3 populations grouped within the skin DC cluster and MDDCs clustered most closely to CD14+ DDC consistent with the proposed similarity between these two cell types. We identified differential expression of novel genes in particular DC subsets including genes related to DC surface receptors (including C-type lectin receptors, toll-like receptors and galectins). Total RNA was extracted and hybridised to 24 bead arrays. Enhanced expression of Ficolin 1 is a specific gene marker of Takayasu's vasculitis

Project description:Takayasu's arteritis (TA) Vasculitis (angiitis) is a chronic inflammatory disease involving large blood vessels. We here identify the genes whose mRNA levels are commonly augmented in the peripheral blood mononuclear cells (PBMCs) from TA patients regardless of the patient's symptom, active or inactive phase, compared with those of normal volunteers. Among them, ficolin 1 (FCN1) was notable because its mRNA level was more than 4.0 fold in all 8 patients examined. A total of 8 two color arrays were performed and each individual array included a target sample and a control sample.
