Identification of ELOB interacting proteins by LC-MS/MS

ABSTRACT: To identify proteins that interact with ELOB (TCEB2) in mouse multiple tissues, including brain, fat, heart, intestine, liver, lung, ovary, spleen, stomach, testis, and uterus. Immunocoprecipitation (IP) experiments were performed and subjected to LC-MS/MS analysis and Label-free quantification.

Whole genome transcriptional profile of human tissues and cells

Project description:We performed tiling array experiments to examine whole genome expression in human tissues. We investigated tissue specificity and association between evolutionary sequence conservation and transcription. A total of 22 tissues (bone marrow, cerebellum, colon, cortex, fetal brain, heart, kidney, liver, lung, pancreas, prostate, salivary gland, skeletal muscle, small intestine, spinal cord, spleen, stomach, testes, thymus, thyroid, trachea and uterus) and 2 cell lines (HeLa and SH-SY5Y) were examined in 36783546 regions of the Human genome in this study. Keywords: whole genome transcription, tiling array, non-coding transcripts, tissue specificity Total RNA isolated from Human tissues and cell cultures were labeled and hybridized to the GeneChip Human Tiling 1.0R Array set of 14 chips (24 samples x 14 = 336 chips total) according to the manufacturer's protocol.

Hepatocellular carcinomas (HCC) in mice transduced in utero with feline immunodeficiency virus-based vectors (expression)

Project description:Fetal mice (16 days gestation) were administered feline immunodeficiency virus (FIV)-based lentiviral viral particles containing the gene encoding GFP. Six liver tumors developed in three mice between the ages of 273 and 484 days, each mouse developed 2 tumors. These tumors and non-tumorous liver tissue from the same animals and animals that did not develop tumors and untransduced controls were harvested and microarrays were performed on total RNA extracted from these samples. We were interested in investigating the link between lentiviral integration and gene expression. Keywords: Comparison of HCC with non-tumorous liver tissue adjacent to tumor. Fourteen samples were analyzed in total: Two tumors from each of three mice (6 samples); surrounding non-tumorous liver tissue from each of these three mice (3 samples); two female mice and one male mouse that were transduced and did not develop tumors (3 samples); one female and one male untransduced mouse (2 samples). The samples were performed in two runs: one containing the samples from female mice and the second containing samples from male mice.

Gene expression profile of Human tissues and cells

Project description:We performed microarray experiments to examine gene expression in human tissues. This data was used for comparison with our humanized mouse study (GEO ID GSE33846) and threshold determination of our tiling array data (GEO ID GSE18490, public in the near future). A total of 22 tissues (bone marrow, cerebellum, colon, cortex, fetal brain, heart, kidney, liver, lung, pancreas, prostate, salivary gland, skeletal muscle, small intestine, spinal cord, spleen, stomach, testes, thymus, thyroid, trachea and uterus) and 2 cell lines (HeLa and SH-SY5Y) were examined. Total RNA isolated from Human tissues and cell cultures were labeled and hybridized to the GeneChip Human Genome U133 Plus 2.0 Array according to the manufacturer's protocol.
