Project description:GST-KRT32 and its interacting proteins in primary keratinocytes cells were enriched by pull-down using glutathione-Sepharose beads. Then, KRT32-interacting proteins were identified by mass spectrum. Proteins obtained in Pulldown were subjected to SDS-PAGE gel electrophoresis and silver staining. The SDS-PAGE gel was sent to the mass spectrometry platform of the Jingjie PTM Biolab for subsequent processing and mass spectrometry detection.

Project description:Human lung cancer cell line 95D cells were injected subcutaneously into right flank of Balb/c nude mice (n=8). 7 days later, the plasmid of p-T-miR-7 (100mg) or p-Cont (100mg) was remote given by subcutaneous injection into the left flank of nude mice five times every three days. 3 days after last injection, tumor mass were collected. We sent the tumor tissue to Shanghai Kang Cheng company to detecte the cDNA chip in order to detect the gene expression in p-T-miR-7 injection group.

Project description:To confirm the downstream genes of LINC00958, we employed RNA sequencing (RNA-seq) to identify the differentially expressed genes when LINC00958 was silenced in Ishikawa cells. The sample of LINC00958-silenced Ishikawa cells (sh-LINC00958, n=3) and negative control cells (sh-NC, n=2) were sent to Shanghai Sangon Biotech of China for library construction, sequencing, data preprocessing and gene mapping. Sequencing was performed on an DNBSEQ-T7 platform for the generation of raw data. Genes were considered significantly differentially expressed if the P value < 0.05 and |log2FC| > 1.The results showed that a total of 305 mRNAs were regulated after LINC00958 knockdown (fold change>2, P<0.05)

Project description:The C57BL/6J mice were randomly divided into three groups: control group (n=3), CuO NPs group (n=3), and CuO NPs + TTM group (n=3). A suspension of 2 mg/mL CuO NPs in 100 μl sterile saline was directly once administered by intratracheal instillation in CuO NPs treatment group. Mice in control group received 100 μl sterile saline. In the CuO NPs + TTM intervention group, 0.02 mg/mL TTM was added to the daily drinking water. All mice were sacrificed on day 7. Total RNA of the mouse lung tissues were extracted using tissue RNA isolation kit. All samples were sent to Majorbio Biotech (Shanghai, China) for transcriptome sequencing.
