Lactiplantibacillus plantarum, LC-MSMS

Project description:This project uses TMT labeling quantitative proteomics technology to carry out research, and a total of 898 proteins have been identified. Differentially expressed proteins were screened according to the criteria of expression fold change of more than 1.5-fold (up-regulation more than 1.5-fold or down-regulation less than 0.67) and P value<0.05. Among them, taking the comparison group Control VS H2O2 as an example, there were 31 up-regulated differentially expressed proteins and 81 down-regulated differentially expressed proteins. Through GO enrichment and KEGG pathway analysis, it was found that these differentially expressed proteins are mainly involved in important biological processes such as single-organism metabolic process, small molecule metabolic process, organophosphate metabolic process, organophosphate biosynthetic process and carbohydrate derivative biosynthetic process, and are mainly involved in the regulation of Metabolic pathways, Fructose and mannose metabolism, Oxidative phosphorylation, Tyrosine and Degradation of aromatic compounds and other important KEGG metabolic pathways.

A c-di-AMP receptor mediates epithelial adhesion of Lactiplantibacillus plantarum

Project description:Cyclic di-adenosine monophosphate (c-di-AMP) is a crucial second messenger used by bacteria to mediate bacterial-host interactions. However, its roles in Lactiplantibacillus plantarum remain poorly understood. Here, we identify RwpL, which consists solely of an HTH domain, as a c-di-AMP receptor in L. plantarum WCFS1. RwpL mediates epithelial adhesion of L. plantarum WCFS1 and functions as both a transcription inhibitor and activator, with transcriptional output determined by the position of its binding site relative to the transcription start site (TSS) of target genes. Specifically, RwpL inhibits wxlA transcription by binding downstream of its TSS, thus reducing WxlA interactions with fibronectin, a matrix protein on the surface of intestinal epithelia. In contrast, RwpL activates transcription of the matrix-degrading metalloprotease gene mmpL by recruiting the sigma factor RpoA upstream of its TSS. Notably, elevated c-di-AMP levels diminish RwpL binding affinity for both DNA and RpoA, thereby reversing its regulatory functions and enhancing L. plantarum adhesion to host epithelial cells. Together, our findings uncover a dual and reversible regulatory mechanism through which c-di-AMP modulates bacterial-host interactions in L. plantarum.

SWATH-MS-based proteome analysis of PBA-enriched glycated/ glycosylated proteins from knee synovial fluid in osteoarthritis (OA) and osteoarthritis with type 2 diabetes mellitus (OADM)

Project description:Osteoarthritis (OA) severity is often exacerbated in individuals with type 2 diabetes mellitus (T2DM), giving rise to a co-morbid condition (OADM) characterized by enhanced metabolic and degenerative alterations. However, the molecular mechanisms underlying this association remain insufficiently understood. In this study, we performed a SWATH-MS-based global proteomic analysis of phenylboronic acid (PBA)-enriched cis-diol–containing glycated/ glycosylated proteins from knee synovial fluid samples of OA and OADM patients. The dataset captures differentially abundant protein signatures associated with metabolic dysregulation, extracellular matrix remodeling, and inflammatory processes in OADM. From a translational perspective, these proteins may have potential relevance as candidate biomarkers for distinguishing OADM from OA and for supporting future investigations into disease stratification and targeted therapeutic approaches.

Maize and Arabidopsis Golgi proteome

Project description:Comparative proteomics of ER-Golgi membranes from etiolated coleoptiles of maize (Zea mays) and young leaves of Arabidopsis (Arabidopsis thaliana) isolated by flotation centrifugation demonstrate differences in the complement of synthases and glycosyl transferases unique to the type of wall made. Additionally, maize Golgi membranes enriched by flotation were separated further by free-flow electrophoresis (FFE), yielding 3009 proteins, of which 229 were known to function in cell wall synthesis or metabolism. A subset of proteins identified after flotation centrifugation were identified in Golgi membranes but failed to enrich in them. Individual classes of proteins associated with cell wall synthesis were asymmetrically distributed across the fractions of Golgi membranes separated by FFE.

Food-associated Lactiplantibacillus plantarum C9O4 reverts inflammation and apoptosis in intestinal epithelial cells: a proteomic insight

Project description:Nowadays, Western diets and lifestyle lead to an increasing occurrence of chronic gut inflammation, that represents an emerging health concern with still a lack of successful therapies. Fermented foods, and their associated Lactic Acid Bacteria, have recently regained popularity for their probiotic potential including the maintenance of gut homeostasis by modulating the immune and inflammatory response. Our study aims to investigate the cross-talk between the food-borne strain Lactiplantibacillus plantarum C9O4 and intestinal epithelial cells in an in vitro inflammation model. Cytokines profile shows the ability of C9O4 to significantly reduce levels of IL-2, IL-5, IL-6, and IFN-γ. Proteomic functional analysis reveals an active host-microbe interaction that highlights an immunoregulatory role of C9O4, able to revert both the detrimental effects of IFN-γ through the JAK/STAT pathway and the apoptosis process in inflamed cells. These results suggest a promising therapeutic role of fermented food-associated microbes for the management of gastrointestinal inflammatory diseases.

Interaction between Lactiplantibacillus plantarum RX-8 and Wickerhamomyces anomalus Y-5 under co-culture by proteomics analysis

Project description:In present study, it was found that the co-culture of Wickerhamomyces anomalus Y-5 and L. plantarum RX-8 could enhance bacteriocin production. To analyze the interaction between W. anomalus Y-5 and L. plantarum RX-8, a quantitative proteomic approach was used to analyze and compare the proteome in L. plantarum RX-8 and W. anomalus Y-5 under mono-culture and co-culture. In total, 339 differently expressed proteins (DEPs) were screened in comparison of L. plantarum RX-8 under mono-culture and co-culture, 645 proteins of W. anomalus Y-5 changed in mono-culture and co-culture. The Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis indicated that DEPs participated in various metabolic pathways such as PTS system, glycolysis, galactose metabolism, glutamate, aspartate, arginine and cysteine metabolism etc. These pathways were related to inducing mechanism on improving bacteriocin production by co-culture. Quantitative proteomic analysis-based strategies can therefore provide further evidence for new regulated targets to improve the production of bacteriocins.

Fermentation of soy juice by Lactiplantibacillus plantarum confers a yogurt-like texture, flavours and health-promoting metabolites

Project description:The aim of the study was to decipher metabolisms responsible (i) for the peculiar adaptation of L. plantarum during soy juice fermentation and (ii) for the release of aroma compounds, amino and short-chain fatty acid, and metabolites with health-promoting properties in soy yogurt. The strategy was the sequencing and annotation of a strain (L. plantarum CIRM-BIA777, PRJEB77707) able to degrade galacto- oligosaccharides, the sampling of soy yogurt, RNA-seq to identify differentially expressed genes of L. plantarum and corresponding metabolisms throughout the kinetics of fermentation. Acids and volatile compounds were also quantified.
