Gut microbial postbiotics as potential therapeutics for lymphoma: Proteomics Insights of the synergistic effects of Nisin and Urolithin B against human lymphoma cells

Project description:Lymphoma remains a substantial global health concern, necessitating innovative therapeutic strategies. The emerging role of gut microbiota-derived metabolites, known as postbiotics, offers promising avenues for cancer treatment. This study evaluated the antiproliferative activities of various postbiotics, including Nisin (N) and urolithin B (UB), and their combinations against the human lymphoma cell line HKB-11. Systematically, the antiproliferative effects and underlying mechanisms were characterised using Alamar Blue assays, combination index (CI) analyses, ROS measurements, flow cytometry, and bottom-up proteomics analyses of combined and mono-treatments. Nisin and UB demonstrated significant antiproliferative activity with IC50 values of 1467 µM and 87.56 µM, respectively. Among other ratios, their combination at a ratio of 4:6 exhibited potent synergy (CI =0.09 at IC95), notably enhancing apoptosis (p < 0.0001) and modulating ROS levels. Proteomics analyses identified key protein alterations involved in lipid metabolism, mitochondrial function, cell cycle progression, and apoptosis.

Unveiling Proteomic Responses to Combined Nisin and Urolithin B Treatment (7:3) in Lymphoma Cells

Project description:This study investigates the antiproliferative and pro-apoptotic effects of the postbiotics nisin and urolithin B (UB) on lymphoma cells, individually and in combination. Human Burkitt lymphoma (HKB-11) and Hs 313.T cell lines and normal stromal HS-5 cells were treated with nisin, UB, and nine different ratios of their combinations. A 7:3 ratio (nisin 5600 μM + UB 150 μM) demonstrated synergistic anticancer activity with minimal cytotoxicity to normal cells. Flow cytometry confirmed that UB alone was the most potent inducer of apoptosis (p < 0.0001), followed by the 7:3 combination, significantly improving nisin’s apoptotic effect. Reactive oxygen species (ROS) assays demonstrated that nisin triggered concentration-dependent ROS production, whereas UB modulated this effect when combined, thereby reducing oxidative stress. Label-free proteomic analysis revealed distinct expression profiles. Nisin treatment upregulated oxidative stress-related proteins (HBA1, CYC1) and suppressed cell cycle proteins (CDK1, CCNB1), indicating mitotic inhibition. UB altered mitochondrial and epigenetic regulators (CYCS, DNMT1), and the combination therapy significantly downregulated ribosomal proteins (RPS14, RPL11, RPS27) and TP53, suggesting that ribosomal stress mediates antiproliferative effects. Synergy analysis confirmed combination index (CI) values <1 at 4:6 and 7:3 ratios, validating synergistic interactions.

Single-cell-type Spatial Proteomics Unveils Precision Medicine Insights in Composite Small Lymphocytic and Classical Hodgkin Lymphoma

Project description:Deep Visual Proteomics (DVP) is an innovative technique that enables spatially resolved sing-cell-type proteome analysis. The coexistence of classical Hodgkin lymphoma (cHL) and small lymphocytic lymphoma (SLL) in a single patient presents a challenging scenario for clinical decision-making. In this study, we investigate the potential of DVP to provide insights into precision medicine strategies. We use DVP to comprehensively profile the proteomic landscapes and signaling pathways within cHL and SLL compartments in a 71-year-old woman with composite lymphoma. Our analysis reveals distinct proteome profiles in cHL and SLL populations, highlighting their clonal unrelatedness. Beyond ABVD chemotherapy, we identify subtype-specific therapeutic targets: Minichromosome Maintenance protein inhibitors and proteasome inhibitors for cHL, and H3K27 methylation inhibitors and receptor tyrosine kinase inhibitors for SLL. Additionally, we explore interleukin-4 inhibition as a potential strategy to manage chemo-resistance. DVP provides insights into spatial single-cell proteomics, guiding personalized treatments for composite lymphoma patients.

Urolithin A maintains primordial follicle dormancy and prevents cyclophosphamide-induced follicle loss by inhibiting the PI3K/Akt signaling

Project description:Urolithin A, a natural gut microbial metabolite, exerts multiple beneficial effects upon supplementation, including prolonging lifespan, mitigating diseases, restoring the quality of aged oocytes and alleviating drug toxicity. The study aims to investigate the ovarian protective role of Urolithin A, focusing on its mechanisms for inhibiting primordial follicle activation and mitigating cyclophosphamide (CY)-induced follicle apoptosis. The results showed that Urolithin A significantly decreased the number of growing follicles, downregulated the expression of oocyte growth-related genes (Gdf9 and Zp3) and protein (DDX4), as well as Ki-67 and BrdU-positive signals. Further studies revealed that Urolithin A downregulated the levels of phosphorylated Akt and FOXO3a and decreased the percentage of oocytes with FOXO3a nuclear export. Molecular docking showed a strong binding ability between Urolithin A and its downregulated gene Pik3cg (PI3Kγ). Moreover, Urolithin A attenuated CY- and 4-HC-induced increases in Cleaved caspase-3 and cleaved PARP1 positive signals. Meanwhile, RNA-seq analysis indicated that Urolithin A downregulated CY-induced expression of DNA damage-related genes (Trp73 and Trim29). In short, Urolithin A inhibits primordial follicle activation by reducing PI3K/Akt pathway reactivity. Furthermore, Urolithin A prevents CY-induced follicle apoptosis. The study provides valuable insights into Urolithin A treatment for chemotherapy-induced infertility.

Urolithin A mitigates behavioral and synaptic deficits associated with epilepsy through VDAC-1 inhibition: A novel treatment approach

Project description:Supplementation of Urolithin-A, a catabolite of ellagic acid-containing precursors, has been shown to benefit the central nervous system (CNS). This study focused on deciphering the potential anti-epileptic effects of Urolithin-A, a small molecule of natural origin. To understand the primary underlying mediators of UA’s anti-epileptic efficacy, RNA sequencing was performed in the sub-acute epileptic model of Drosophila (4 hours of treatment duration with epileptic inducer-picrotoxin (PTX)) to abstain from the interference of secondary pathways. Four treatment groups were included in the study: 1) Vehicle (0.5% DMSO), 2) 1mM PTX, 3) 1mM PTX + Urolithin-A (100µM), and 4) Urolithin-A (100µM). Administration of Urolithin-A restores the PTX-induced misregulated genes of brain hemolymph and metabolic dysfunctions.

Untargeted proteomics on Enterocloster bolteae DSM 15670 Treated with urolithin C

Project description:Urolithin A is a polyphenol derived from the multi-step metabolism of dietary ellagitannins by the human gut microbiota which can affect host health. Most, but not all, individuals harbor a microbiota capable of urolithin A production; however, the enzymes that dehydroxylate its dietary precursor, urolithin C, are unknown. Here, we used a combination of transcriptomics and proteomics to reveal a urolithin C dehydroxylase (ucd) operon that dehydroxylates 9-hydroxy urolithin compounds in Enterocloster spp. Using comparative genomics, we identified Lachnoclostridium pacaense as a novel urolithin C metabolizer. Biochemical characterization and structure predictions of proteins in the Ucd complex demonstrated that dehydroxylation was both NADH- and molybdopterin-dependent and used urolithin C as a terminal electron acceptor. A meta-analysis publicly available metagenomic data revealed that both bacteria and ucd operon genes are widely distributed in gut metagenomes and likely comprise keystone species in the metabolism of urolithins by the human gut microbiota.

Untargeted proteomics of Enterocloster asparagiformis, E. citroniae, E. pacaense treated with DMSO or urolithin M6

Project description:Urolithins are a class of bioactive metabolites derived from the metabolism of dietary ellagitannins by the human gut microbiota. In the gut, urolithins are dehydroxylated regioselectively based on microbiota composition and activity. A single 9-hydroxy urolithin dehydroxylase (ucd) operon in gut resident Enterocloster species has been described to date; however, most enzymes in the urolithin metabolic pathway remain uncharacterized. Here, we investigate urolithin cross-feeding between members of the gut microbiota and discover a novel urolithin dehydroxylase in a subset of Enterocloster species. We show that urolithin intermediates, released by gut resident Gordonibacter species during ellagic acid metabolism, are dehydroxylated at both the 9- and 10-positions by E. asparagiformis, E. citroniae, and E. pacaense, but not E. bolteae. Using untargeted proteomics, we uncover a 10-hydroxy urolithin dehydroxylase operon, termed uxd, responsible for these species-specific differences in urolithin metabolism. By inducing uxd expression with diverse urolithins, we show that 9-hydroxy urolithins are required for uxd transcription and 10-position dehydroxylation. Collectively, this study reveals some of the genes, proteins, and substrate features underlying differences in urolithin metabolism by the human gut microbiota.

It includes human peripheral blood monocyte (MONO), human peripheral blood monocyte derived macrophages treated with M-CSF (MACRO)and primary alveolar macrophages (BAL). All the experiments are performed comparing two of the three cell types from the same person (total 4 persons). Totally we got three set of microarray data, MONO/MACRO, MONO/BAL and MACRO/BAL with 4 biological replicates.
