Project description:Plant productivity is adversely affected by exposure to abiotic stresses including salinity. Leaf tissue undergoes accelerated senescence with exposure to salt stress conditions, culminating in lowered efficiency of photosynthetic machinery. The N6-adenine derivative phytohormone Cytokinin (CK) is well known to delay natural leaf senescence by maintaining functioning photosynthesis processes. How various forms of CK function to control this activity and alter physiological and molecular responses under salt-induced leaf senescence is still not known. Here the CKs, trans-Zeatin (tZ) and its N-glucosylated (tZNGs) forms; tZ7G and tZ9G, were tested for their ability to delay leaf senescence in Arabidopsis thaliana. A modified dark-induced leaf senescence CK bioassay was used to study the physiology of photosynthesis performance as well as transcriptome and proteome level changes in detached leaves, across different phases of salt-induced leaf senescence from 2h-72h. Treatments with different CK forms (tZ, tZ7G, and tZ9G) showed that tZNGs can improve PSII efficiency (Fv/Fm) and increase chlorophyll levels compared to salt only treatments, indicating that these CK forms function as active CK forms in delaying senescence. At the transcript level, tZ, tZ7G, and tZ9G revealed differentially expressed genes (DEGs) across both developmental and salt-treated senescence, with tZNGs predominantly altering DEG in the terminal timepoint of salt-treated leaf senescence. Additionally, we found that tZNGs uniquely regulate crucial biological processes (BPs) including CK signaling compared to tZ. Proteome analysis showed that tZNGs and tZ showed similarities in number of differentially abundant proteins (DAPs) trends across senescence timepoints although each of the three forms showing uniquely abundant proteins. GO term analysis for tZNG regulated genes and proteins revel the enrichment of senescence- and chloroplast-related BPs. This study highlights the role of tZNGs as active cytokinin forms towards delaying salt-treated leaf senescence in Arabidopsis at the physiology, transcriptome, and proteome level.

Project description:Pedunculate oak (Quercus robur) is a foundation tree species in European forests and reforestation programs, and forest nurseries represent key phytosanitary bottlenecks where asymptomatic planting stock can disseminate cryptic pathogens. Cadophora luteo-olivacea is frequently associated with grapevine trunk diseases and has been recovered from diverse woody hosts, yet its pathogenic potential on oak and its interactions with antagonistic fungi remain unresolved. Here, we combine pathogenicity testing, dual-culture confrontation assays and time-resolved, spatially resolved contact-zone proteomics and metabolomics to connect detection to causality and mechanism in an oak associated Cadophora–Trichoderma system. We fulfill Koch’s postulates for C. luteo-olivacea on Q. robur seedlings and quantify inhibition of Cadophora by a natural isolate of Trichoderma atroviride. Contact-zone proteomics at 4 and 8 days post-contact reveals a staged, interface-localized antagonistic program dominated by fungal cell-wall targeting hydrolases (including chitinases and β-1,3-glucanases), secreted proteases, oxidoreductases, transporters and small secreted proteins, alongside factors consistent with adhesion and self-protection. Together, these complementary assays provide the first experimental evidence that C. luteo-olivacea is pathogenic on Q. robur and delineate mechanistic signatures of T. atroviride mycoparasitism at the interaction front. This framework links nursery-relevant pathogen screening to mechanistic readouts and informs evaluation of biocontrol potential in a foundation forest species.

Project description:The transition of skotomorphogenesis to photomorphogenesis is induced by the perception of light and is characterized by the inhibition of hypocotyl elongation and opening of cotyledons. Although it is known that the plant hormone cytokinin, when applied in high concentrations, inhibits hypocotyl elongation in the dark-grown Arabidopsis plants, it is unclear to what extent this response is the result of cytokinin alone or cytokinin-induced ethylene production. We show that treatment of etiolated seedlings in presence of ethylene inhibitors (eg. AgNO3) or treatment of the ethylene-resistant mutant ein2, resulted in a significant inhibition of hypocotyl elongation. This indicates that cytokinin induced de-etiolation is largely independent of ethylene and suggests a close connection between the cytokinin two component system and the light singalling networks. We show that this cytokinin signal is mainly mediated through the cytokinin receptor ARIBIDOPSIS HISTIDIN KINASE 3 (AHK3) and the ARABIDOPSIS RESPONSE REGULATORS 1 (ARR1) in combination with ARR12. Interestingly, mutation of COP1, DET1 and CIN4/COP10 renders plants insensitive to cytokinin and these factors are indispensable for the transcriptional response during cytokinin induced de-etiolation which indicates that a functional light signaling pathway is essential for this cytokinin response. In addition, the cytokinin effect on hypocotyl elongation is highly dependent on the ambient light conditions where higher light intensities causes a switch in the response to CK from an inhibitor to a promoter of hypocotyl elongation.

Project description:The phytohormone cytokinin has diverse functions in plants, most of which have been primarily elucidated in dicots. In order to further characterize the role of cytokinin in a monocot, we used CRISPR-Cas9 to generate rice lines with frameshift alleles in the four putative cytokinin receptors (Histidine Kinases 3,4,5,6) in all single and multiple combinations. The transcriptome of the hk3,4,5,6 mutant is completely insensitive to cytokinin, suggesting that all cytokinin signaling regulating transcription acts through these four HKs, despite the presence of a CHASE-containing Serine-Threonine kinase in rice.

Project description:Cytokinin is a phytohormone involved in the regulation of diverse developmental and physiological processes in plants. Its potential for biotechnology and development of high-yield and more resilient plants has been recognized, but the molecular mechanisms behind its action are far from understood. In this report, the roots of barley seedling were explored as a new tool to reveal as yet unknown cytokinin-responsive proteins. Significant differences were reproducibly observed for 176 proteins, and at least some of the revealed cytokinin-responsive pathways were confirmed in metabolome analysis, including alterations in phenylpropanoid pathway, amino acid biosynthesis or ROS metabolism. Bioinformatics analysis indicated a significant overlap between cytokinin response and response to abiotic stress. This was confirmed by comparing proteome and metabolome profiles in response to drought, salinity or a period of temperature stress. The results illustrate complex abiotic stress response in the early development of model crop plant and confirm an extensive crosstalk between plant hormone cytokinin and response to temperature stimuli, water availability or salinity.

Project description:According to the well-documented scenario with regard to the cytokinin-mediated phosphorelay signal transduction in Arabidopsis thaliana, certain members of the type-B ARR family are crucially implicated in the regulatory networks that are primarily propagated by the cytokinin-receptors (AHKs) in response to cytokinin. Nevertheless, clarification of the biological impact of these type-B ARR transcription factors is at a very early stage. Here we focused on a pair of highly homologous ARR10 and ARR12 genes by constructing an arr10 and arr12 double-null mutant. The mutant alleles used in this study were arr10-5 and arr12-1. arr10-5 is the SALK_098604 T-DNA insertion line, whose mutation was determined to be located in the fifth exon of the ARR10 coding sequence. arr12-1 is the SALK_054752 T-DNA insertion line, whose mutation was determined to be located in the third exon of the ARR12 coding sequence. The resulting mutant showed remarkable phenotypes with special reference to the cytokinin-action in roots (e.g., inhibition of root elongation, green callus formation from explants). Furthermore, we demonstrated that ARR10 and ARR12 are involved in the AHK-dependent signaling pathway that modulates the differentiation of root-vascular tissues (i.e., protoxylem-specification), suggesting that ARR10 and ARR12 are the prominent players that act redundantly in the AHK-dependent cytokinin signaling in roots. Keeping this in mind, we then collected the root-specific and combinatorial DNA microarray datasets with regard to the cytokinin-responsible genes by employing both the wild-type and arr10 arr12 double-mutant plants. In this study, wild-type and the arr10 arr12 mutant grown vertically on MS agar plates for 2 weeks were treated with 20 microM of the cytokinin trans-zeatin (TZ) or 0.02% DMSO (solvent for trans-zeatin solution) for 1h. These treated plant samples were divided into three portions, from which RNA samples were prepared separately from roots of seedlings with use of RNeasy Plant Mini Kit (Qiagen, Valencia, CA, U.S.A.). The quality of RNAs prepared was analyzed by Bioanalyzer 2100 (Agilent Technologies). These RNA samples were processed as recommended by the Affymetrix instruction (Affymetrix GeneChip Expression Analysis Technical Manual, Affymetrix). These datasets will provide us with bases for understanding the early response to cytokinin on roots of seedlings in Arabidopsis thaliana.

Project description:The plant hormone cytokinin regulates diverse aspects of plant growth and development, likely through context-dependent transcriptional regulation that relies on a dynamic interplay between regulatory proteins and chromatin. We employed the Assay for Transposase Accessible Chromatin (ATAC-Seq) to profile changes in the chromatin landscape of Arabidopsis roots and shoots in response to cytokinin. Our results reveal differentially accessible chromatin regions indicative of dynamic regulation in response to cytokinin. These changes in chromatin occur preferentially upstream of genes whose expression changes in response to cytokinin and largely overlap with binding sites for the type-B ARRs, transcription factors that mediate the primary response to cytokinin. Further, the type-B ARRs were found to be necessary for the changes in chromatin state in response to cytokinin. Lastly, we find context-dependent responses by comparing root and shoot profiles. These datasets provide new insights into the transcriptional regulatory mechanisms involved in the cytokinin response and how cytokinin mediates its pleiotropic effects.
