Proteomics on mouse obese and non-obese visceral adipose tissue derived extracellular vesicles

Project description:Visceral adipose tissue (VAT) was harvested from wild-type mice fed high-fat diet to induce obesity or matched diet as non-obese controls. VAT was ex vivo cultured for 48 hrs and its conditioned media was collected. Extracellular vesicles were isolated from conditioned media by ultracentrifugation. The aim of the project was to characterize extracellular vesicle protein cargo from obese and non-obese VAT.

A long-term high fat diet induces differential gene expression changes in spatially distinct adipose tissues.

Project description:The accumulation of visceral adipose tissue (VAT) is strongly associated with cardiovascular disease and diabetes. In contrast, individuals with increased subcutaneous adipose tissue (SAT) without corresponding increases in VAT are associated with a metabolic healthy obese phenotype. These observations implicate dysfunctional VAT as a driver of disease processes, warranting investigation into obesity-induced alterations of distinct adipose depots. To determine the effects of obesity on adipose gene expression, mice were fed either a high fat or normal laboratory diet for 12-14 months. Mesenteric VAT and hindlimb SAT were isolated from four lean controls and four obese mice for bulk RNA- sequencing. AT from lean controls served as a reference to obesity-induced changes. The long-term high fat diet induced the expression of 169 and 814 unique genes in SAT and VAT, respectively. SAT from obese mice exhibited a total of 308 differentially expressed genes (164 upregulated, 144 downregulated). VAT from obese mice exhibited 690 differentially expressed genes (262 genes upregulated, 428 downregulated). KEGG pathway and GO analyses revealed that metabolic pathways were upregulated in SAT vs. downregulated in VAT while inflammatory signaling was upregulated in VAT. We next determined common genes that were differentially regulated between SAT and VAT in response to obesity and identified four genes that exhibited this profile: elovl6 and kcnj15 were upregulated in SAT/downregulated in VAT while trdn and hspb7 were downregulated in SAT/ upregulated in VAT. We propose that these genes in particular should be further pursued to determine their roles in SAT vs. VAT with obesity.

Project description:Obesity is correlated with multitype of cancer development, and pancreatic ductal adenocarcinoma (PDAC) in obese patients often shows dismal prognosis and resistance to immune checkpoint blockade (ICB) therapy. The molecular mechanism for these phenomena is largely unknown. Here we show that obese visceral adipose tissues (VAT) can talk to distant PDAC by delivering their extracellular vesicle (EV) carrying signal molecules. We reveal that PDAC cells can take in VAT-EVs to their lysosomes where EVs-delivered Cathepsin A (Ctsa) stabilize ribonuclease Rnaset2b to produce extra pseudouridines from RNA cleavage. Pseudouridines released from cancer cells activate mast cells which inhibit CD8+ T cell activity, forming an immunosuppressive tumor microenvironment which enhances cancer progression and resistance to ICB. We also demonstrate the harmful effects of VAT-EV CTSA-pseudouridine-mast cell axis for PDAC in obese patients. Animal experiments indicate that Ctsa knockdown effectively enhances the ICB efficacy on PDAC. Our study uncovers a mechanism connecting obesity and cancer which holds promise for developing new therapeutic strategy for obesity related cancers.

Project description:The protein Tsg101 plays an integral role extracellular vesicle formation and secretion from cells. We developed a mouse model with Tsg101 knockdown (Tsg101dAd). Visceral adipose tissue (VAT) was harvested from Tsg101dAD and Tsg101flfl mice fed high-fat diet to induce obesity or matched diet as non-obese controls. VAT was ex vivo cultured for 48 hrs and its conditioned media was collected. Extracellular vesicles were isolated from conditioned media by ultracentrifugation. The aim of the project was to characterize extracellular vesicle protein cargo from obese and non-obese VAT.

We also demonstrate harmful effects of VAT-EV CTSA-pseudouridine-mast cell axis for obesity-related PDAC. Animal experiments indicate that Ctsa knockdown effectively enhances ICB efficacy on PDAC. Our study uncovers a mechanism connecting obesity and cancer which holds promise for developing new therapeutic strategy for obesity-related cancers.

Project description:The molecular background of mitochondrial dysfunction in adipose tissue of morbidly obese individuals and bariatric surgery-induced changes in adipose mitochondrial function remain incompletely understood. To evaluate the mechanisms behind the surgery-induced changes and differences between morbidly obese subjects and nonobese controls, we performed a LC-MS/MS proteomics analysis of abdominal subcutaneous (SAT) and visceral adipose tissue samples (VAT) collected from the bariatric surgery, SAT samples collected 6 months after surgery, and control SAT and VAT samples collected from baseline.
