Animal blood African arworks LC-MSMS

Project description:Multiple analytical techniques were used to characterize materials from the surfaces of two African sculptures in the collection of the Art Institute of Chicago: a Bamana power object (boli), and a Yoruba wooden sculpture of a female figure. Surface accretions on objects such as these have received relatively little scientific attention to elucidate their composition and function, in part because they are made with complex mixtures of natural materials, which are often unfamiliar and poorly represented in the scientific literature on artists’ materials. Mass spectrometry-based proteomics provided new insights on proteinaceous components, allowing us to identify the presence of sacrificial blood, and more specifically, blood from chicken, goat, sheep and dog. This new scientific evidence supports and supplements knowledge derived from curatorial and field work studies, and opens new doors to understanding the objects’ significance and history of use.

Multiple regulatory intrinsically disordered motifs control FOXO4 transcription factor binding and function

Project description:Transcription factors harbour defined intrinsically disordered regulatory regions, which raises the question of how they mediate binding to structured co-regulators and how this regulates activity. Here, we present a detailed molecular regulatory mechanism of Forkhead box O4 (FOXO4) by the structured transcriptional co-regulator β-catenin. We find that the largely disordered FOXO4 C-terminal region, which contains its transactivation domain binds β-catenin through two defined interaction sites, and this is regulated by combined PKB/AKT- and CK1-mediated phosphorylation. Binding of β-catenin competes with the auto-inhibitory interaction of the FOXO4 disordered region with its DNA-binding forkhead domain, and thereby enhances FOXO4 transcriptional activity. Furthermore, we show that binding of the β-catenin inhibitor protein ICAT is compatible with FOXO4 binding to β-catenin, suggesting that ICAT acts as a molecular switch between anti-proliferative FOXO and pro-proliferative Wnt/TCF/LEF signalling. Together these data illustrate how the interplay of intrinsically disordered regions, post-translational modifications and co-factor binding contribute to transcription factor function. Identification of beta-catenin binding regions in SW480 cells

Project description:Deregulation of canonical Wnt/beta-catenin pathway is one of the earliest events in the pathogenesis of colon cancer. Mutations in APC or CTNNB1 (beta-catenin gene) are highly frequent in colon cancer and cause aberrant stabilization of b-catenin, which activates the transcription of Wnt target genes by binding to chromatin via the TCF/LEF transcription factors. Here we report an integrative analysis of genome-wide chromatin occupancy of b-catenin by chromatin immunoprecipitation coupled with high-throughput sequencing (ChIP-seq) and gene expression profiling by microarray analysis upon RNAi-mediated knockdown of beta-catenin in colon cancer cells (GSE53656). Immunoprecipitated samples from human colon cancer SW480 cells with antibodies against beta-catenin and control IgG respectively were used for ChIP-seq experiments.

Black, red and white: characterization of painting materials on a group of Bwa masks from Burkina Faso, proteomics

Project description:The distinctive and visually striking wooden masks associated with the Bwa culture in Burkina Faso, West Africa, are carved from a soft wood into different shapes and display various geometrical patterns and symbols according to the purpose. One of their characteristic features is the use of the colors black, red and white, which evoke the three major rivers crossing the country: the Black, Red and White Voltas. According to published accounts of scholars who have worked directly with the artists, the materials used to obtain these colors include reptile excrement for the white, iron-rich stones powdered and mixed with egg or plant gums for the red, and boiled Acacia seed pods for the black, as well as modern materials such as enamel paint in some cases. A group of four Bwa masks in the Arts of Africa collection of the Art Institute of Chicago was investigated using a complement of analytical techniques including Fourier transform infrared spectroscopy, pyrolysis gas chromatography mass spectrometry, and mass spectrometry based proteomics to characterize their painting materials. The results obtained corroborate the published accounts, while also providing new insights into the nature of the coloring materials and the selection and substitution of pigments and binders. These findings highlight the complementary value of scientific research, in combination with field work and artists’ accounts, to generate a fuller understanding and appreciation of this traditional artistic practice.

A transphyletic study of metazoan β-catenin protein complexes

Project description:β-catenin plays a vital role in various biological processes, such as body axis determination and cell differentiation, during embryonic development in metazoans. These β-catenin functions are thought to be exerted through complexes formed with various types of proteins. Although β-catenin complex proteins have been identified in several bilaterian models, little is known about the structural and functional properties of β-catenin complexes in the early metazoan evolutionary phases. In this study, we performed a comparative analysis of β-catenin sequences in nonbilaterian lineages that branched off early in metazoan evolution. We aslo carried out a transphyletic function experiments of β-catenin from non-bilaterian metazoans using developing Xenopus embryos, which included secondary axis induction in embryos and proteomic analysis of the β-catenin protein complex. Comparative functional analysis of nonbilaterian β-catenins also demonstrated sequence characteristics important for β-catenin function, and the deep origin and evolutionary conservation of the cadherin-catenin complex. Proteins coimmunoprecipitated with β-catenin included several proteins conserved across metazoans. These data provide a new insight into the conserved repertoire of β-catenin complexes.
