Proteomic analysis of Fosinopril-induced effects in A549 human lung adenocarcinoma cells

ABSTRACT: This project contains the quantitative proteomics data from a study investigating the effects of fosinopril on human lung adenocarcinoma cells. A549 cell lines were treated with 90 μM fosinopril or vehicle control for 24 days. Proteins were digested and analyzed by LC-MS/MS to identify and quantify differentially expressed proteins, aiming to reveal the drug's mechanisms of action and potential therapeutic targets.

Repurposing existing drugs offers a promising approach to cancer therapy. Fosinopril, an angiotensin converting enzyme inhibitor (ACEI) approved for hypertension, has demonstrated antitumor effects in hepatocellular carcinoma. GeLC-FAIMS-MS Workflow for In-Depth Middle-Down Proteomics

Project description:Middle-down proteomics is an analytical approach whereby protein samples are digested with proteases such as Glu-C to generate large peptides (>3 kDa) that are then analyzed by mass spectrometry. This method is useful for characterizing high-molecular-weight proteins that are difficult to detect by top-down proteomics, in which intact proteins are analyzed by mass spectrometry. In this study, we applied GeLC-FAIMS-MS, a multidimensional separation workflow that combines gel-based prefractionation with LC-FAIMS Orbitrap mass spectrometry, for deep middle-down proteomics. Middle-down peptides obtained under the condition of optimized limited Glu-C digestion were first size-fractionated by polyacrylamide gel electrophoresis, followed by C4 reversed-phase liquid chromatography separation and additional ion mobility fractionation, resulting in a significant increase in peptide length detectable by mass spectrometry. In addition to global analysis, the GeLC-FAIMS concept can also be applied to targeted middle-down proteomics, where only proteins in the desired molecular weight range are gel-fractionated and their Glu-C digestion products are analyzed, as demonstrated by targeted analysis of integrins in exosomes. In-depth middle-down proteomics achieved by global and targeted GeLC-FAIMS-MS allows the exploration of proteoform information not covered by conventional top-down proteomics via increase in the number of detectable proteoforms and improvement in sequence coverage.

Project description:Preparation of AMDs. 125 μM Ac4ManNAz were incubated with Raw 264.7 (~ 107 cells/dish) for 24, 48, and 72 h, followed by gentle washing with PBS or complete medium to produce Raw 264.7-Ac4ManNAz. Subsequently, Raw 264.7-Ac4ManNAz (~ 107 cells) were resuspended in PBS containing Alkynyl-Ce6(0.1 μM), ascorbic acid (0.2 μM) and CuSO4 (0.2 μM) and incubated for 1h at 37 °C. Then centrifuged (1000 rpm, 3 min) and washed 3 times with PBS to obtain precursor of AMDs. precursor of AMDs (~ 107 cells/tube, 1.0 ml) was immersed in liquid nitrogen, 24 h later, the frozen tubes were thawed at 37°C, centrifuged at 1500 rpm for 3 min, washed and resuspended by PBS to generate the AMDs (5 x 106 units/tube, 1.0 ml).

Recurrent disruption of HIP1 identified in patients with epilepsy, learning difficulties, intellectual disability, and neurobehavioral abnormalities

Project description:We report 12 individuals from ten unrelated families with epilepsy, learning difficulties, intellectual disability, and neurobehavioral abnormalities, who segregated a microdeletion distally adjacent to the Williams-Beuren syndrome region. In six families, a recurrent ~ 1.1 Mb deletion likely resulted from nonallelic homologous recombination between flanking large complex low-copy repeats. Three smaller sized microdeletions (~ 180-500 kb) enabled us to narrow the critical region to one gene, HIP1, encoding Huntington interacting protein 1. genomic DNA from blood was used to analyse 12 samples with control sample using custom made NimbleGen 12x135K microarrays for chr7 region.

Low P stress transcriptome of control rice variety (Dongjin-byeo) and NtPT1-trasngenic rice (PT 1-7-8)

Project description:To understand expression of candidate gene located on QTLs for phosphate sensitivity traits under low P using NtPT1-transgenic rice with increased Pi uptake efficiency and gene expression profile at the V3-stage seedling through the 60K Rice Whole Genome Microarray Uniformly germinated seedlings were transferred to a hydroponic system filled with aerated standard solution (normal P, 320 M Pi) and grown for 2 weeks in a glasshouse at 25°C/18°C (day/night) under a natural photoperiod (about 16 h). The composition and concentration of each element in the standard solution were as follows: N (1.43 mM), P (0.32 mM), K (0.51 mM), Ca (0.75 mM), Mg (1.64 mM), Fe (59.37 μM), B (18.92 μM), Mn (9.50 μM), Mo (0.10 μM), Zn (0.15 μM), Cu (0.16 μM), and citric acid (70.72 μM) (Yoshida et al., 1976). Some of the two-week-old seedlings were transferred to the low P solution (the same nutrient solution containing 32 M Pi) over a period of 5 days to expose them to P-deficiency condition. Control plants were maintained in the standard solution for the entire period.
