U2OS cells EL133 and Bardoxolone

ABSTRACT: U2OS cells were incubated with DMSO at 0.1% or with compound for 20 hours in quadruplicates. Following incubation, the cells were lysed in a mixed detergent buffer (Cells were lysed using detergent based buffer containing the following: (50mM HEPES pH 8.0, 1% SDS, 1% Triton X-100, 1% NP-40, 1% Tween 20, 1% sodium deoxycholate, 50mM NaCl, 5mM EDTA, 1% (w/v) glycerol). Samples were then reduced using DTT, followed by reaction with iodoacetamide. The labeled proteins were then precipitated on a dispersion of glass beads, followed by overnight trypsin digestion and purification of peptides
