Project description:High-throughput comparative proteomic analysis based on TMT labeling combined with LC-MS/MS were performed on an in vivo Ang II infusion mice model to obtain a comprehensive view of the protein ensembles associated with thoracic PVAT (tPVAT) dysfunction induced by Ang II. In total, 5,037 proteins were confidently identified, of which 4,984 proteins were quantified. Compared with saline group, 145 proteins were upregulated and 146 proteins were downregulated during Ang II-induced tPVAT pathogenesis. Bioinformatics analyses revealed that the most enriched GO terms were annotated as gene silencing, monosaccharide binding, and extracellular matrix. In addition, some novel proteins potentially associated with Ang II infusion were identified, such as acyl-CoA carboxylase α, very long-chain acyl-CoA synthetase (ACSVL), uncoupling protein 1 (UCP1), perilipin, RAS protein-specific guanine nucleotide-releasing factor 2 (RasGRF2), and hypoxia inducible factor 1α (HIF-1α). Ang II could directly participate in the regulation of lipid metabolism, transportation, and adipocyte differentiation by affecting UCP1 and perilipin. Importantly, the key KEGG pathways were involved in fatty acid biosynthesis, FABP3-PPARα/γ, RasGRF2-ERK-HIF-1α, RasGRF2-PKC-HIF-1α, and STAT3-HIF-1α axis.

For each day, the number of differentially expressed genes (DEGs) each showing the same GO terms were summed up. In order to correlate these changes with the number of all genes with the same GO term in Euglena gracilis, data from sequencing projects were employed [1,2]. Genes with the same GO terms were counted and correlated with the number of observed DEGs in Euglena gracilis. In the first table, GO terms of DEGs were assigned to corresponding days of culturing and correlated with the data of [1, 2] (total numbers and ratios). Project description:Euglena gracilis is a unicellular freshwater flagellate, which uses the gravitational vector for orientation in the water column in the dark. This allows the cell to reach areas in the water column for reproduction and growth. How exactly the gravitational vector is perceived, and which intracellular pathways are involved in the signaling is not very well understood so far. In the past, parabolic flight campaigns were used to study the swimming behavior of Euglena gracilis under altered gravitational accelerations. It was shown that cells adapt their swimming direction very fast: in the dark under 1xg cell show negative gravitaxis, i.e. they move upwards in the water column of the experiment hardware and against the gravitational vector. With onset of the first hypergravity period of 1.8xg the precision of upward swimming increases slightly. This first hyper gravity lasts for only 20 seconds and is followed by 22 sec of microgravity. During this period no gravitational vector is perceived by the cells, therefore they lack a cue for orientation and move randomly in all direction. In the subsequent hyper gravity period of 1.8xg, which also lasts for 20 sec, cells direct their movement again and swim upwards. Over different parabolic flight campaigns and other experiments it was shown that this gravitactic behavior is linked to changes in membrane potential, calcium and cAMP concentration. However, due to the lack of genomic and transcriptomic data, it was so far not possible to link the differential movement to the abundance of distinct mRNA transcripts. In contrast, other model organisms, such as Arabidopsis thaliana, have been analyzed by means of gene expression with respect to the effects of altered gravitational accelerations. Also various human cell lines have shown to adapt their gene expression in dependence of the prevailing acceleration. With the recently published Euglena gracilis transcriptome, we now aimed at analyzing effects of altered acceleration on the gene expression in the flagellate. Therefore, Euglena gracilis samples were taken in the course of the 29th DLR parabolic flight campaign during parabola 1 and 31 (time difference of 2 hours and 30 additional parabolas). During both parabolas samples were fixed with TRIzol at 1xg just before onset of the first hyper gravity period, 20 sec into hyper gravity (1.8xg), 20 sec into microgravity (µg) and 20 sec into the last hypergravity period.

Project description:Mitogen-activated protein kinases usually affect biological processes by phosphorylating proteins. To further investigate the consequences of MAPK11 interacting with SnRK1 in controlling tomato seed dormancy, we performed a phosphorylation label-free quantitative proteomic assay in vivo with dry seeds of TS-9and MAPK11-OE lines. Data analysis showed that a total of 8261 phosphosites and 6026 phosphopeptides derived from 2711 phosphoproteins were identified. The phosphopeptides with an average fold-change >2 or <0.5 and a P value <0.05 were selected as being significantly different. Among all identified phosphopeptides, 87 phosphopeptides were significantly up-regulated and 83 phosphopeptides were significantly down-regulated in seeds of MAPK11-OE line. In addition, we also found that 348 phosphopeptides were just abundant in MAPK11-OE-13 line, and 221 phosphopeptides merely in TS-9, respectively. To explore potential biological function of MAPK11, we conducted GO function annotation and enrichment, KEGG passway annotation and enrichment, and protein protein interaction network (PPI) analyses, and detected that MAPK11 might have effect on binding, catalytic activity, transporter activity, molecular function regulator and signal transducer; and involved in cellular process, metabolic process, biological regulation, regulation of biological process and response to stimulus

Project description:Little is known about cell survival in the presence of reducing reagents, likewise changing the redox state of living organisms and, therefore potentially conferring a negative effect on respiration and major metabolic pathways. We employed Escherichia coli as a model organism to analyze its responses to sublethal concentrations of sodium sulfite, one of the common food preservatives used for inhibition of microbial growth. We found that partial inhibition of E. coli growth by sodium sulfite was accompanied by profound changes in gene expression. Besides affecting major metabolic pathways, these changes apparently led to reduced production of flagellar proteins, thus potentially impacting cell motility. Concomitantly, E. coli upregulated a number of genes involved in repair of macromolecules and antisense control.
