Proteomics analysis of TRiC interactome from Stra8-Pdcd5-/- mouse elongating spermatids

ABSTRACT: In order to investigate the interaction proteins of TRiC in Stra8-Pdcd5-/- elongating spermatids, TRiC immunoprecipitation from Pdcd5F/F and Stra8-Pdcd5-/- elongating spermatids were collected for LC-MS/MS analysis. A total of 4419 proteins were quantified, and 375 proteins (p < 0.05, Foldchange > 1.5) were differentially expressed. Among these differential proteins, 83 proteins were downregulated (22%) and 292 proteins were upregulated (78%) in Stra8-Pdcd5-/- elongating spermatids.

Quantitative proteomics analysis of Stra8-Pdcd5-/- mouse elongating spermatids

Project description:In order to investigate the molecular mechanism of PDCD5 in the regulation of spermatids development, elongating spermatids from Pdcd5F/F and Stra8-Pdcd5-/- mice were collected for quantitative proteomics analysis using TMT-based liquid chromatography-tandem mass spectrometry (LC-MS/MS) approach. A total of 8730 proteins were quantified, and 512 proteins (p < 0.05, Foldchange > 1.5) were differentially expressed. Among these differential proteins, 58 proteins were downregulated (11%) and 454 proteins were upregulated (89%) in Stra8-Pdcd5-/- elongating spermatids.

Project description:Histone hyperacetylation is thought to drive the replacement of histones by transition proteins that occurs in elongating spermatids after a general shut-down of transcription. The molecular machineries underlying this histone hyperacetylation remain still undefined. Here we focused our attention on the role of Cbp and p300 in histone hyperacetylation and in the preceding late gene transcriptional activity in elongating spermatids. A strategy was designed to partially deplete Cbp and p300 in elongating spermatids. These cells progressed normally through spermiogenesis and showed normal histone hyperacetylation and removal. However, a genome-wide transcriptomic analysis, performed in the round and elongating spermatids, revealed the existence of a gene regulatory circuit encompassing genes presenting high expression levels in pre-meiotic cells, undergoing a repressed state in spermatocytes and early post-meiotic cells, but becoming reactivated in elongating spermatids, just prior to the global shut-down of transcription. Interestingly, this group of genes was over-represented within the genes affected by Cbp/p300 knock-down and were all involved in metabolic remodelling. This study revealed the occurrence of a tightly regulated Cbp/p300-dependent gene expression program that drives a specific metabolic state both in progenitor spermatogenic cells and in late transcriptionally active spermatids and confirmed a special link between Cpb/p300 and cell metabolism programming previously shown in somatic cells. Total RNA were obtained from post meiotic male germ cells (round spermatids and condensing spermatids) of adult mice either wild-type or with a double knock down of Cbp/P300 in post-meiotic cells. In each experiments, 6 replicates of each genotype and cell type were used.

Project description:We reasoned that the pi-RISC, by virtue of the enormous sequence portfolio of piRNAs, might mediate elimination of a large variety of mRNAs in late stages of spermiogenesis. Both Miwi-null and Caf1-null mice showed early spermiogenic arrest, preventing us from determining the role of MIWI and CAF1 in elongating spermatids using the existing genetic models. We therefore used microarrays to detail the global effect of MIWI or CAF1 on mRNA levels in mouse elongating spermatids. Using GFP+ elongating spermatids sorted from mouse testes transduced with shMiwi:GFP, shCaf1:GFP, or control pSilencer:GFP, we performed transcriptome profiling on Affymetrix mouse arrays.

Cells from 6 WT mice or 10 Stra8-cre/Hdac3fl/- mice were pooled for one biological replicate. RNA-seq libraries were prepared in biological triplicates.

Project description:Spermatogenesis culminates in a dramatic morphological transformation, including a tight compaction of the chromatin and nuclear re-shaping that largely silences transcription. Due to transcriptional silencing, the production of the sperm-specific proteins needed for the morphological transformation requires active storage and translational regulation of mRNAs transcribed in earlier cell types. The germline-specific RNP granule, the chromatoid body (CB), accumulates RNAs and has a role in RNA storage in early haploid cells (round spermatids). In late haploid cells (elongating spermatids), the CB is transformed to so-called late-CB, whose function in RNA regulation has remained elusive. Here we characterized the function of the late-CB by identifying proteins and RNAs interacting with the known late-CB marker, Testis-Specific Serine Kinase 2 (TSSK2). We showed that TSSK2 interacts with eukaryotic translation initiation factors and the late-CB associates with active, localized protein synthesis. Furthermore, we revealed an association of TSSK2 with a specific set of mRNAs that are enriched in polysome fractions in elongating spermatids, supporting the role of the late-CB in the temporally regulated translation. These results link the function of the late-CB to the RNA regulation during late spermatogenesis for the first time, providing important novel information about the RNA regulatory processes required for spermatogenesis and male fertility.
