Proteomic Profiling of Plasma in Type-1 Diabetes Mellitus
Ontology highlight
ABSTRACT: Plasma samples were obtained from a total of sixty individuals, comprising forty participants with type-1 diabetes (T1D) and twenty non-diabetic, healthy control participants. Among the T1D cohort, insulin resistance was stratified according to estimated glucose disposal rate (eGDR): twenty participants met the criterion for insulin resistance (eGDR ≤6 mg/kg/min), whereas twenty were classified as insulin-sensitive (eGDR ≥8 mg/kg/min). Venous blood was collected into EDTA anticoagulant tubes and processed promptly to minimise pre-analytical variation. Plasma separation was initiated within thirty minutes of phlebotomy. Samples were centrifuged at 2400×g for 20 min at 4°C to remove cells and debris. The resulting supernatant plasma was carefully aspirated, aliquoted into cryovials to avoid repeated freeze-thaw cycles, and stored at -80°C until further analysis.
