Proteomics of PLGA-based nanoparticles protein corona after mild isolation and its impact on macrophage uptake (PLGA100).


ABSTRACT: Recognition of the protein corona, a layer of adsorbed protein around nanosystems, is necessary for a better understanding of bio-nano interaction. This study first compares different experimental methods for PLGA nanoparticles protein corona isolation, then investigated the biological identity and fingerprint profile of adsorbed proteins on variously pegylated PLGA-based nanoparticles using microfiltration as an ideal isolation method coupled with proteomics. The total proteins identified in the corona of nanoparticles with different compositions were classified based on their abundance and gene-ontology. Lastly, measurements of nanoparticles absorption by macrophages showed a strong association with the characteristics of the protein corona.
