Global proteome analysis of sperm from tropically-adapted rams with contrasting parameters of frozen-thawed semen
Ontology highlight
ABSTRACT: This study integrated functional assessments of ram sperm (motility, viability, membrane and acrosome integrity, chromatin stability) with shotgun proteomics (LC-MS/MS, label-free) to identify molecular signatures associated with cryotolerance. Hair rams were classified into high- and low-freezability groups based on post-thaw motility reduction. Proteomic profiling revealed both exclusive and differentially abundant proteins between groups, highlighting extracellular/immunoprotective modules in high-freezability rams and mitochondrial/axonemal pathways in low-freezability rams. These findings provide candidate biomarkers and mechanistic insights into sperm cryoresistance.
