Violacein as a multitarget agent in bladder cancer cells

ABSTRACT: Violacein is a bacterial purple pigment with antimicrobial, antiparasitic, antifungal, antiviral, and anticancer properties. We previously reported that violacein has antiproliferative activity in cervical and bladder cancer-derived cells, and also sensitizes cells to cisplatin. Currently, we report the results obtained on T24 bladder cancer cells by using quantitative differential proteomics.

Proteomics to identify putative genes underlying fruit ripening and related to long shelf life

Project description:Our aim was to gather additional evidence of tomato fruit ripening by assessing two long shelf life near isogenic lines (NILs). These two NILs, named NIL115 and NIL080, carried wild introgression provided by S. pimpinellifolium accession LA0722 in the genetic background of Caimanta (CAI) from S. lycopersicum. Pericarp tissue was collected from the four genotypes (NIL115, NIL080, CAI and LA0722) at two ripening stages: mature green (MG) and red ripe (RR). A label-free quantitation (LFQ) proteomic analysis was carried out to identify differentially abundant proteins between MG and RR.

Stress response and detoxification mechanisms involved in non-target site herbicide resistance in sunflower

Project description:The objective of this study was to evaluate the role of cytochrome P450 monooxygenases (P450s) and other detoxification-related proteins in Imisun sunflower resistance. Two sunflower inbred lines were used: HA 425 and HA 89, imidazolinone (IMI)-resistant and susceptible, respectively. Growth response to imazethapyr herbicide in combination with P450s inhibitors 1-aminobenzotriazole (ABT) and piperonyl butoxide (PBO) was evaluated in 15-day-old sunflower plantlets. Roots were collected and label-free quantitation (LFQ) proteomic analysis were carried out in order to characterize plant response to IMI. Roots of 15-day-old plants treated with 0 and 3.3 µM imazethapyr were frozen in liquid nitrogen. Protein extraction was performed as described in Wu et al. (2014) from 1 g root tissue per genotype/treatment combination.

Amyloids as protein traps in bladder cancer

Project description:Despite the current diagnostic and therapeutic approaches to bladder cancer being widely accepted, there have been few significant advancements in this field over the past decades. This underscores the necessity for a paradigm shift in the approach to bladder cancer. The role of amyloids in cancer remains unclear despite their identification in several other pathologies. In this study, we present evidence of amyloids in bladder cancer, both in vitro and in vivo. In a murine model of bladder cancer, a positive correlation was observed between amyloids and tumor stage, indicating an association between amyloids and bladder cancer progression. Subsequently, the amyloid proteome of the RT4 non-invasive and HT1197 invasive bladder cancer cell lines was identified and included oncogenes, tumor suppressors, and highly expressed cancer-related proteins. It is proposed that amyloids function as structures that sequester key proteins. Therefore, amyloids should be considered in the study and diagnosis of bladder cancer.

Transcription profiling by array of human bladder cancer cell lines with or without TP53 mutations

Project description:The established cell lines RT4, 5637 and T24 from human bladder TCC were obtained from the Cell Bank of the Federal University of Rio de Janeiro, Brazil. The 5637 cells harbor two TP53 mutations, at codon 72 (Arg . Pro) and codon 280 (Arg . Thr).T24 cells contain a TP53 allele encoding an in-frame deletion of tyrosine 126. Both cell lines were established from high-grade bladder tumors. No specific mutations were detected in RT4 cells, which had been established from a low-grade papillary bladder tumor. The RT4 and T24 cell lines were maintained in DulbeccoM-^Rs modified EagleM-^Rs medium (Sigma-Aldrich, Inc, St Louis, MO, USA), and the 5637 cells were kept in Roswell Park Memorial Institute medium (Sigma-Aldrich). Both media were supplemented with 10% fetal bovine serum (Cultilab Ltd, Campinas, Brazil), 100 U/mL penicillin G (Sigma-Aldrich), 100 U/mL streptomycin (Sigma-Aldrich) and 1% kanamycin sulfate (Amresco, Branded Products Group, Solon, OH, USA); cells were cultured at 37M-:C in an atmosphere of 5% CO2. A pooled reference design was chosen. More specifically, each array was hybridized with the same reference sample labeled with Cy5, while the experimental samples (control or treated) were labeled with Cy3.

PROTEOME IMPACT ON MAIZE SILKS UNDER PRIMING STATE INDUCED BY TRICHODERMA ROOT COLONIZATION

Project description:Trichoderma species can stimulate local and distant immune responses in colonized plant tissues to prevent future pathogenic attacks. Priming of plant defenses is characterized by changes on transcriptional, metabolic, and epigenetic states after stimulus perception. We have previously investigated the transcriptional reprogramming in silk tissues from maize plants inoculated with Trichoderma atroviride and challenged with Fusarium verticillioides (Agostini et al 2019). To better understand about the molecular changes induced by T. atroviride in maize, a proteomic approach was conducted in this instance. Several proteins belonging to different metabolic categories were detected as priming involved proteins. However, we detected a very low correlation with those priming-modulated transcripts suggesting the importance of regulatory events posteriori of transcriptional stage to accomplish the final goal of blocking the pathogen entrance. Specifically, we focused on phenylpropanoid pathway; since we detected several proteins that are upregulated in priming state might explain the cell wall reinforcement and, the increase in the content of flavonoids and lignin in silks of maize plants after induced systemic resistance activation.

H2A.Z Mapping During G0/G1 and Mitosis

Project description:We report genome wide mapping of the histone variant H2A.Z during G0/G1 and mitosis in T24 bladder cancer cells. The results show that the broad enrichment pattern of H2A.Z near transcription start sites of active genes is maintained during mitosis. Furthermore, using H2A.Z localization to visualize nucleosome positioning near the start site, we see that the +1 nucleosome of active genes shifts upstream to occupy the transcription start sites during mitosis and the nucleosome depleted region is shortened. H2A.Z is also maintained on the -2 nucleosome which also shifts towrds the transcription start site during mitosis, further contributing to the shorteneing of the nucleosome depleted region.
