Proteomics characterization of plastic degrading purple bacteria

ABSTRACT: The aim of this research was to isolate purple bacteria from waste that degrade plastics. Using metagenomic techniques, bacteria living in plastic debris were identified, and subsequently the metabolic pathways and proteins involved in them were studied using metaproteomics.

Functional characterization of microbial communities from glacial ecosystems

Project description:Glaciers are populated by a large number of microorganisms including bacteria, archaea and microeukaryotes. Several factors such as solar radiation, nutrient availability and water content greatly determine the diversity and abundance of these microbial populations, the type of metabolism and the biogeochemical cycles. In order to study their metabolic potentials, samples of glacial ice were taken from several glacial ecosystems. Microorganisms were analyzed by a polyphasic approach that combines a set of -omic techniques: 16S rRNA sequencing, culturomics and metaproteomics. This combination provides key information about diversity and functions of microbial populations, especially in rare habitats. Several whole essential proteins and enzymes related to metabolism and energy production, recombination and translation were found that demonstrate the existence of cellular activity at subzero temperatures.

Project description:Glaciers are populated by a large number of microorganisms including bacteria, archaea and microeukaryotes. From an ecological point of view, three ecosystems can be differentiated in glaciers: the supraglacial ecosystem, the subglacial ecosystem and the englacial ecosystem. Several factors such as solar radiation, nutrient availability and water content greatly determine the diversity and abundance of microbial populations, the type of metabolism and the biogeochemical cycles. This huge diversity is threatened by climate change. This sampling place, Sarrios ice cave (Monte Perdido massif, Spanish Pyrenees) host one of the last permafrost deposits. Permafrost can provide us very worthy information about past climate and environmental conditions, therefore it should be studied as soon as possible due to the huge retreat that is already taking place. In order to study their metabolic potentials, samples of ice and permafrost were taken from an Antarctic glacier. The cells were harvested and their proteins were extracted and analyzed by matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI/TOF/TOF). Several proteins and enzymes were found that demonstrate the existence of cellular activity at subzero temperatures. In this way it is shown that the englacial microorganisms are not quiescent, but that they maintain an active metabolism and play an important role in the glacial microbial community.

However, these unknown englacial microorganisms establish a true trophic chain and appear to have an active metabolism. In order to study their metabolic potentials, samples of englacial ice were taken from an Antarctic glacier. Project description:Purple phototrophic bacteria (PPB) naturally accept CO2 into their metabolism as a primary redox sink system in photo-heterotrophy. Dedicated use of this feature for developing sustainable processes (e.g., through negative-emissions photo-bioelectrosynthesis) requires a deep knowledge of the inherent metabolic mechanisms. Here we provide evidence of the tuning of the PPB metabolic mechanisms upon redox stressing through negative polarization (-0.4 and -0.8 V vs. Ag/AgCl) in photo-bioelectrochemical devices. Using metaproteomic analysis at both reactor ans species level, we showed that a mixed PPB-culture up-regulates its ability to capture CO2 from organics oxidation through the Calvin-Besson-Bassam cycle and anaplerotic pathways, and the redox imbalance is promoted to polyhydroxyalkanoates production. The ecological relationship of PPB with mutualist bacteria stabilizes the system and opens the door for future development of photo-bioelectrochemical devices focused on CO2 up-cycling.
