Characterization of MS white matter derived extracellular vesicles

ABSTRACT: This dataset contains proteomic profiles of extracellular vesicles (EVs) isolated from human multiple sclerosis (MS) white matter tissue, alongside the corresponding tissue proteome. The aim of this study is to characterize the protein cargo of MS-derived EVs compared to bulk tissue and to identify molecular features linked to MS pathology. Proteins were labeled using TMTpro 18-plex reagents, fractionated by basic pH reversed-phase LC, and analyzed by Orbitrap Fusion Lumos mass spectrometry.

Extracellular vesicles (EVs) are increasingly recognized as mediators of central nervous system (CNS) function and pathologies, including multiple sclerosis (MS). While plasma-derived EVs have been explored as biomarkers in MS, little is known about EVs in CNS tissue. Here, we characterize EVs from postmortem normal-appearing white matter (NAWM) of MS and control brains. EVs were separated by differential centrifugation followed by size exclusion chromatography and characterized using nanoflow c  ...[more]
