TMT-based quantitative proteomics to compare the proteomes of wild-type and unc-45(m94) animals during the recovery phase in C. elegans

ABSTRACT: The wild-type (WT) and unc-45(m94) C. elegans were cultured at 15 °C under normal conditions and shifted to 25 °C for 16–18 h at the L4 stage, followed by recovery at 15 °C. In WT animals, muscle integrity and protein homeostasis are efficiently restored during recovery, whereas in unc-45(m94) mutants, defective chaperone activity is expected to impair this process, leading to persistent proteostatic stress. This TMT-based proteomic dataset reveals the proteins that are differentially regulated in unc-45(m94) animals compared to WT during this temperature shift, particularly during recovery.
