Project description:Left-right asymmetry is a basic character of aging brain; however, the molecular foundation of the left-right asymmetry remains unclear. The morphology, physiology and behavior of rhesus aging are obviously similar to human aging, but the aging-rate of rhesus is roughly three times as fast as human, in which the underlying mechanism needs further investigation. By using of 6-plex tandem mass tag (TMT) labeling, we presented a high throughput quantitative proteomics analysis to 6 group hippocampal samples including left and right hippocampus from 3 years, 6 years and 20 years old rhesus. Our data identified 3391 high-confidence proteins. After screening, we found 340 aging-related proteins of left hippocampus and 334 aging-related proteins of right hippocampus, in which there were 114 overlap proteins. Furthermore, the aging-related proteome of left rhesus hippocampus aging was compared with human aging-related proteome of left hippocampus that was reported by our lab previously. As the results show, we discovered 446 aging-related proteins in rhesus and 830 aging-related proteins in human with an overlap of 106 proteins.

Project description:Streptococcus pneumoniae (S.pneumoniae) is a Gram-positive bacterial pathogen that colonizes on the mucosal surfaces of the host's nasopharynx and upper respiratory tract and results in pneumonia. In order to survive and infect the host, S.pneumoniae must have obtain essential nutrients, such as transition metal ions . Our previous study had shown that the mRNA and protein levels of SPD-0090 are significantly upregulated in the ΔpiuA/ΔpiaA/ΔpitA triple mutant (three major iron transports), but its detailed biological function is unknown. In this study, we found that the knockout spd-0090 gene induced a delayed growth in the medium with different sugar sources. F urther iTRAQ quantitative proteomics studies revealed that SPD-0090 affects galactose metabolism and iron ion transport system. Then RT-qPCR and intracellular galactose content assays showed that SPD-0090 affects galactose uptake, leads to reduced galactose utilization. In addition, in vitro biochemical assays showed that SPD-0090 is a hemin transporter and affects iron uptake. Notably the knockout of spd-0090 resulted in an enhanced infection ability of S.pneumonaie to Our study reveals that the dual function of SPD-0090 plays an important role in the virulence of S.pneumoniae.

Project description:Nowadays, high-throughput quantitative proteomic and transcriptomic approaches have been widely used for exploring molecular mechanisms and acquiring biomarkers for cancers. Our study aims to illuminate the multi-dimensional molecular mechanisms underlying renal cell carcinoma via investigating the quantitative global proteome and the profile of phosphorylation. A total of 5428 proteins and 8632 phosphorylation sites were quantified in RCC tissues, with 709 proteins and 649 phosphorylation sites changing in expression compared with matched adjacent non-tumor tissues. These differentially expressed proteins were mainly involved in the metabolic process terms involving glycolysis pathway, oxidative phosphorylation and fatty acid metabolism which had been considered to be a potential mechanism of RCC progression. Moreover, phosphorylation analysis indicated that these up-regulated phosphorylated proteins were implicated in glucagon signaling pathway and cholesterol metabolism, while the down-regulated phosphorylation proteins were predominantly involved in glycolysis, pentose phosphate pathway, carbon metabolism and biosynthesis of amino acids. In addition, we also found several new candidate proteins such as CD14, MPO, NCF2, SOD2, PARP1 up-regulated and MUT, ACADM, PCK1 down-regulated in RCC, which might be recognized as new biomarkers for RCC. These findings could broaden our insight into the underlying molecular mechanisms of RCC and acquire biomarker candidates for the treatment of RCC.

Project description:Anthocyanins and flavonols are natural compounds that accumulate preferentially in grapevine flowers and fruits. They are among the most abundant flavonoids and play a very important role in grape and wine quality. In particular, they confer and stabilize colour and contribute to other organoleptic characteristics of the final product. Their complex profile in terms of concentration and relative abundance varies among cultivars and makes wine typicity. The synthesis of these compounds is mainly regulated at transcriptional level. Flavonoids accumulate at specific stages and in specific tissues during flower and berry development, as a consequence of the timely expression of the genes necessary for their synthesis. Although the general flavonoid pathway has been genetically and biochemically elucidated and the main determinants of colour have been identified, the molecular reasons of the fine variation among grape cultivars are still not completely understood. To shed light on this issue, extreme genotypes of a segregating population derived from the cross Syrah x Pinot Noir were characterized at transcriptional level. The transcriptome analysis along three berry developmental stages of these genotypes has allowed the identification of a large set of transcripts differentially modulated between the two groups. A population derived from M-bM-^@M-^XSyrahM-bM-^@M-^Y x M-bM-^@M-^XPinot NoirM-bM-^@M-^Y consisting of 170 F1 individuals plus the parental lines was characterized for the content of flavonols and anthocyanins in the skins of mature berries (38E-L; 18M-BM-0Brix). Based on the biochemical data, 4 high- and 4 low-flavonol producers (HFPs and LFPs: F1 16, F1 56, F1 63, F1 223 and F1 64, F1 256, F1 260, PN), two of which having also either very high or very low anthocyanin content (HAPs and LAPs: F1 63, F1 223 and F1 256, F1 260), were selected for gene expression analysis. A representative sample for each individual (one biological replicate) was collected at three berry developmental stages (hard green berry (33E-L, PV), veraison (35E-L, 50% coloured berries, VER) and maturity (38E-L, 18M-BM-0Brix, MAT)) during the 2007 season.

Project description:Mandatory potency testing of Leptospira vaccine batches relies partially on in vivo procedures, requiring large numbers of laboratory animals. Cell-based assays could replace in vivo tests if biomarkers indicative of Leptospira vaccine potency are identified. We investigated innate immune responsiveness induced by inactivated L. interrogans serogroups Canicola and Icterohaemorrhagiae, and two bivalent, non-adjuvanted canine Leptospira vaccines containing the same serogroups. First, the transcriptome and proteome analysis of canine 030-D cells stimulated with Leptospira strains, and the corresponding vaccine revealed more than 900 DEGs and 23 DEPs in common to these three stimuli. Second, comparison of responses induced by this Leptospira vaccine and a vaccine from another manufacturer revealed a large overlap in DEGs and DEPs as well, suggesting potential to identify biomarkers of Leptospira vaccine activity. Because not many common DEPs were identified, we selected seven molecules from the identified DEGs, associated with pathways related to innate immunity, of which CXCL-10, IL-1β, SAA, and complement C3 showed increased secretion upon stimulation with both Leptospira vaccines. These molecules could be interesting targets for development of biomarker-based assays in the future. Additionally, this study contributes to the understanding of the mechanisms by which Leptospira vaccines induce innate immune responses in the dog.

Project description:Background: Microalgae are promising feedstocks for production of renewable biofuels and value-added bioproducts. Temperature and nitrogen supply are important environmental and nutritional factors affecting the growth and metabolism of microalgae, respectively. In this study, the growth and lipid accumulation of filamentous microalga Xanthonema hormidioides under different temperatures (5, 7, 10, 15, 20, 25, 27 and 30℃) and initial nitrogen concentrations (3, 9, 18 mM) were investigated, and its adaptive mechanisms of tolerance to low temperature and nitrogen stress were analysis by proteomics. Results: The optimum temperature range for the growth of X. hormidioides was between 15℃ and 20℃, and the algal cells had slow growth rate at 5℃ and could not survive at 30℃. The maximum biomass concentration was 11.73 g L-1 under the temperature of 20℃, and the highest total lipid content was 56.63% of dry weight. Low temperature did not change the fatty acids profiles but promoted the accumulation of unsaturated fatty acids of X. hormidioides. The maximum contents of palmitoleic acid, eicosapentaenoic acid and total fatty acid were 23.64%, 2.49% and 41.14% of dry weight, respectively. Proteomics was performed under three temperature (7、15、25℃), two nitrogen concentrations (3 and 18 mM) and two cultivation times (day3 and 12). A total of 6503 proteins were identified. In the low temperature, photosynthesis related proteins were down-regulation to protect the photosynthetic apparatus. The up-regulation of key enzymes DGAT and PDAT demonstrated the accumulation of TAGs under low nitrogen treatment. The proteins related to ribosome, phosphatidylinositol signaling system, antioxidant system and cold shock proteins (CSPs) in X. hormidioides were co-up-regulate under the treatment of low temperature, which can alleviate the damages induced by temperature stress and maintain the normal growth and metabolism of algal cells. Conclusions: X. hormidioides is a psychrotolerant microalga. It is an oleaginous filamentous microalga containing hyper palmitoleic acid and a certain amount of eicosapentaenoic acid with great potential for biofuel development, as well as for applications in nutritional health products and other industries.
