Size-exclusion chromatography of soluble chloroplast proteins from Nicotiana tabacum (tobacco)

ABSTRACT: Maturase K (MatK) is the only splicing factor encoded in the chloroplast genomes of land plants. Here we used a transplastomic Nicotiana tabacum line, in which the plastidial MatK gene is C-terminally tagged with a 3x-HA tag, to conduct size-exclusion chromatography of soluble protein extracts enriched for chloroplast proteins. Fractions 5 and 6, in which MatK-HA was found most abundant by western blot analysis, were subjected to mass spectrometry analysis.

Project description:To confirm the localization of GCSC1 to chloroplast vesicle rather than in the stroma, we purified chloroplasts from WT plants and fractionated the proteins into the membrane and stroma fractions. Quantification of proteins by liquid chromatography-tandem mass spectrometry (LC-MS/MS) revealed that GCSC1 was present only in the membrane fractions, co-existing with several chloroplast membrane proteins.

Project description:In this study we analyzed the effects of CRY2 over-expression on chloroplast genome transcription of tomato, by developing and using a tiling array. This array containing about 90,000 overlapping probes (5-nt resolution) is a versatile tool for global functional studies of tomato cp genome. We profiled transcription in leaves of wild-type (WT) and CRY2-overexpressing (CRY2-OX) plants grown in a diurnal cycle, to generate a comprehensive map of plastid transcription and to monitor potential specific modulations of chloroplast transcriptome induced by the overexpression of CRY2.

Arabidopsis FTSH11 mutant

Project description:The membrane-integrated metallo-protease FtsH11 of Arabidopsis thaliana is proposed to be dual targeted to mitochondria and chloroplasts. A bleached phenotype was observed in ftsh11 grown at long days or continuous light, pointing to disturbances in the chloroplast. Under these growth conditions the morphology of the chloroplast became drastically altered, accompanied by a drop of NPQ, and the amount of PSI decreased relative to PSII. The abundance of plastidic proteins, especially of photosynthetic proteins, was altered in ftsh11 even during growth in normal conditions and short days. FtsH11 is crucial for chloroplast structure and function during growth in prolonged photoperiod . In chloroplasts FtsH11 was found to be located exclusively in the chloroplast envelope. Thanks to proteomic analyses, the Tic22-like protein and YGGT, both chloroplast proteins of unknown function, are proposed to be the substrates of FtsH11 .

Salt stress influence on cucumber chloroplast transcription

Project description:The experiment was made do to assess the influence of increased salt concentration on cucumber chloroplast transcription. We used 0,4M NaCl water solution and we checked its influence on plants after 0, 4, 8, 24, 48 hours. We have also measured changes in chloroplast gene expression after recovery of plants in water (48 hours of 0,4M NaCl solution treatment followed by 48 hours in clean water). Other growth conditions weren't changed comparing to those of standard plants growth in our experiments (check: versatile chamber growth protocol)

ECBiG PlasTi-microarray CSH validation

Project description:Experiment evaluating aplicability of PlasTi-microarray for cross-species hybridization studies. PlasTi-microarray is a tiling oligonucleotide microarray originally designed for cucumber plastome analysis.Chloroplast RNA from Arabidopsis, tomato and spinach leaves was extracted, labelled and hybridized to PlasTi-microarray with the cucumber samples labelled with the second dye as a control. For each species, one biological sample and one technical replicate (labelled in a dye-swap orientation) were analyzed, resulting in two microarray hybridizations per species and six microarrays (a-f) in total.
