MRNA Tethered Ribosome Interactome Profiling (mTRIP): A method for Identifying Trans-Acting Factors on Ribosome-Bound mRNA in Yeast
Ontology highlight
ABSTRACT: Translation initiation in eukaryotes is a tightly regulated process that ensures accurate recognition of the AUG start codon. However, certain mRNAs can utilize non-AUG codons such as UUG for initiation. In the Glycine tRNA Synthetase 1 (Grs1) mRNA both AUG and UUG serve as translation start sites. To investigate the translation-acting factors associated with Grs1 mRNA during initiation, and to compare these with canonical initiation events (e.g., Actin (Act1) mRNA), we developed a method to isolate mRNAs in the 40S ribosomal subunit–bound state. Proteomic analysis of these samples enabled the identification of the ribosome-bound interactome specific to Grs1 and Act1 mRNAs.
