A lipoprotein partner for the Escherichia coli outer membrane protein TolC

ABSTRACT: TolC is an outer-membrane protein that serves as an activated exit duct in multi-drug efflux pumps and protein secretion machinery. We have discovered that the E. coli lipoprotein YbjP interacts extensively with the periplasmic surface of TolC and its N-terminal lipid moiety is embedded in the membrane, mimicking the intramolecular and modification-membrane interactions seen in TolC homologs. Our results suggest that YbjP may contribute to stabilising the orientation and distribution of TolC in the outer membrane as well as the expression of transporters. The accompanying data corresponds to DIA mass spectra from tolC, ybjP, tolC-ybjP mutants, as well as WT E. coli as a reference.

Transcriptomal changes of Escherichia coli TolC grown with the biofilm inhibitor Carolacton

Project description:Carolacton is a novel biofilm inhibitor that kills biofilm cells of Streptococcus mutans and inhibits growth of the clinically relevant and human pathogenic bacterium Streptococcus pneumoniae TIGR4 in nanomolar concentrations. Interestingly, Carolacton also inhibits Escherichia coli with a defective outer membrane protein TolC . The cellular target of Carolacton is still unknown. Here, we adressed the differential transcription of cellular RNAs when E.coli TolC was grown in the presence of Carolacton. This was done to identify transcriptional regulatory networks that are directly affected by treatment of the strain with Carolacton. In order to gain insights into the primary transcriptional response, early time-points were chosen for sampling, which should not reflect secondary responses (e.g. due to differences in growth phase, nutrient depletion etc.).

A TetR-family regulator of an RND efflux system that directs artemisinin resistance in Vibrio cholerae

Project description:Artemisinin (ARS) displayed bactericidal activity against Vibrio cholerae. To assess the mechanistic details of its antibacterial action, we have isolated V. cholerae mutants with enhanced ARS resistance and identified a gene (VCA0767), whose loss-of-function resulted in the ARS resistance phenotypes. This gene (atrR) encodes a TetR family transcriptional regulator, and its deletion mutant displayed the reduction in ARS-induced ROS formation and DNA damage. Transcriptomic analysis revealed that the genes encoding an RND efflux pump operon (vexRAB) and the outer membrane component (tolC) were highly upregulated in the artR mutant, suggesting that AtrR might act as a negative regulator of this operon and tolC. Gene deletion of vexR, vexB or tolC abrogated the ARS resistance of the atrR mutant and, more importantly, the ectopic expression of VexAB-TolC was sufficient for the ARS resistance, indicating that the increased expression of the VexAB-TolC efflux system is necessary and sufficient for the ARS resistance of the atrR mutant. The cytoplasmic accumulation of ARS was compromised in the vexBtolC mutant, suggesting that the VexAB-TolC might be the primary efflux system exporting ARS to reduce its toxicity inside of the bacterial cells. The atrR mutant displayed resistance to erythromycin as well in a VexR-dependent manner. This result suggests that AtrR may act as a global regulator responsible for preventing intracellular accumulation of toxic chemicals by enhancing the RND efflux system.

Porin-mediated passage of negamycin across the outer membrane of Escherichia coli

Project description:The rise of antimicrobial resistant pathogens calls for new antibacterial treatments, but potent new compounds are scarce. Development of new antibiotics is difficult, especially against Gram-negative bacteria, as here uptake is strongly hindered by the additional outer membrane. Most antimicrobial agents against Gram-negatives use the porin mediated pathway to cross the outer membrane, which limits the choice of an antibiotic, as it has to fit by size, charge and hydrophilicity. In E. coli, the major porins OmpF and OmpC are associated with antibiotic translocation and therefore also with unspecific antibiotic cross-resistance. In this regard, alternative uptake routes are of interest. We were interested in the uptake opportunities of the small, natural product antibiotic negamycin and thereby found new uptake pathways across the outer membrane of E. coli. Besides OmpF and OmpC, we investigated the role of the minor porins OmpN and ChiP in negamycin translocation. We detected an effect of OmpN and ChiP on negamycin susceptibility and confirmed passage by electrophysiological assays. The structure of OmpN was resolved in order to analyze the negamycin translocation mechanism by computational simulations. As abundancy of these minor porins was low in E. coli, their transcript levels were analyzed by RNA-Seq. Increased transcripts levels of ompN and chiP were observed upon negamycin treatment, hinting at a role in antibiotic uptake. These new, additional uptake pathways across the outer membrane of E. coli highlight the antibiotic potential of negamycin, especially as resistance development is low due to availability of multiple uptake routes at both the outer and inner membranes

Gram negative promiscuous lipoproteins keep surface topology when transplanted from one species to another and can deliver foreign polypeptides to the bacterial surface

Project description:In Gram negative bacteria outer membrane-associated lipoproteins caneither face the periplasm orprotrude out of the bacterial surface. The mechanisms involved in lipoprotein transport through the outer membrane are not fully elucidated. A group of lipoproteins reach the surface by using species-specific transport machineries. By contrast, a still poorly characterized group, we referred to as “promiscuous lipoproteins”, appears to cross the outer membrane using transport systems conserved among diderms. To provide further evidence of the existence of promiscuous lipoproteins,we tested the expression and compartmentalization in E. coli of three surface-exposed lipoproteins, twofrom Neisseria meningitidis (Nm-fHbp and NHBA) andone fromAggregatibacteractinomycetemcomitans (Aa-fHbp). We found that the three lipoproteins werelipidated and compartmentalized in E. coliouter membrane and in Outer Membrane Vesicles (OMVs). Furthermore, FACS analysis,proteolytic surface shaving, and confocal microscopy revealed that the three proteins were also exposed on the external side of the outer membrane. Removal or substitution of the first four amino acids following the lipidated Cysteine residue and extensive deletions of the C-terminal regions in Nm-fHbp did not prevent the protein from reaching the external side of the outer membrane. Heterologous polypeptides fused to the C termini of the proteins are efficiently transported to the E. coli surface and efficiently compartmentalized in OMVs. These data indicate that promiscuous lipoproteins travel from the cytoplasm to the cell surface using conserved mechanisms and when transplanted from one species to another they maintain their natural topology without the need of ancillary transport machineries. Furthermore such proteins can be exploited in biotechnological applications requiring the decoration of Gram negative bacterial surface with foreign polypeptides.

A genetic platform to investigate the functions of bacterial drug efflux pumps

Project description:Efflux pumps are a significant challenge for the development of new antibacterial agents. Overcoming efflux requires an in-depth understanding of efflux pump functions, substrate specificities, and the development of inhibitors. However, the complexities of drug efflux networks have limited such studies. To address these challenges, we report the generation of Efflux KnockOut-35 (EKO-35), a highly susceptible Escherichia coli strain lacking 35 efflux pumps. We demonstrate the utility of this strain by constructing an efflux platform consisting of strains individually expressing genes encoding efflux pumps forming tripartite complexes with the outer membrane channel TolC. This platform was profiled against a curated diverse compound collection, which enabled us to define physicochemical properties that contribute to transport. We also show the E. coli drug efflux network is conditionally essential for growth, and that the platform can be used to investigate efflux pump inhibitor specificities and also efflux pump interplay. We believe EKO-35 and the efflux platform will have widespread application for the study of drug efflux.

Determinants of heat resistance in Escherichia coli AW1.7

Project description:Microarray hybridization of cDNA libraries obtained from exponentially growing or heat-shocked AW1.7 or GGG10 cultures was performed to compare gene expression of these two strains. Expression of selected genes from different functional groups was quantified by quantitative PCR (q-PCR). DnaK, 30S and 50S risobomal subunits were overexpressed in E. coli GGG10 relative to E. coli AW1.7 upon heat shock at 50°C, indicating improved ribosome stability. The outer membrane porin NmpC and several transport proteins were overexpressed in exponentially growing E. coli AW1.7.
