Shell composition of the marine planktonic ciliate Schmidingerella (Alveolata, Ciliophora, Oligotrichea)

ABSTRACT: About 1,700 shells of the tintinnid ciliate Schmidingerella (SPMC176) were collected from monoclonal culture material. The shells were enzymatically digested to determine their composition by tandem-mass spectrometry.

Deceptive Ceropegia sandersonii uses an arabinogalactan for trapping its fly pollinators

Project description:Many plant species have evolved surfaces designed to reduce insect attachment. Among such plants are deceptive trap flowers of Ceropegia. Their gliding zones consist of epidermal cells each topped by a papilla secreting a single small liquid droplet on its tip. So far, the molecular and physical mechanisms controlling the function of these droplets are unknown. We analyzed the droplets of C. sandersonii flowers by microscopic approaches, studied how they behave when getting in contact with the feet of fly pollinators, and analyzed their chemical composition. The droplets contaminate the insect feet, on which they solidify. As its main component a negatively charged polysaccharide containing a β1,3-galactan backbone and Rhaa1,4GlcAβ1,6[Arafa1,3]Galβ1,6 side chains or truncated versions of it was identified by NMR spectroscopy. The chemical structure represents a rudiment version of an arabinogalactan, which is supported by its binding to β-D-glucosyl Yariv reagent. A proteomics approach revealed candidates of arabinogalactan proteins to which the polysaccharide is connected. The high amount of GlcA in the polysaccharide explains the unusual physical characteristics of the droplets like viscoelasticity and hygroscopy. We add a new function to arabinogalactan proteins, and discuss, why the identified polymer is well suited for catching and temporarily trapping of pollinators.

Extracellular vesicles from therapeutic grade allogeneic human placental stromal cells induce angiogenesis and modulate immunity

Project description:Allogeneic regenerative cell therapy has shown surprising results despite lack of engraftment of the transplanted cells. Their efficacy was so far considered to be mostly due to secreted trophic factors. We hypothesized that extracellular vesicles (EVs) can also contribute to their mode of action. Here we provide evidence that EVs derived from therapeutic placental-expanded (PLX) stromal cells are potent inducers of angiogenesis and modulate immune cell proliferation in a dose dependent manner. Crude EVs were enriched >100-fold from large volume PLX conditioned media via tangential flow filtration (TFF) as determined by tunable resistive pulse sensing (TRPS). Additional TFF purification was devised to separate EVs from cell-secreted soluble factors. EV identity was confirmed by western blot, calcein-based flow cytometry and electron microscopy. Surface marker profiling of tetraspanin-positive EVs identified expression of cell- and matrix-interacting adhesion molecules. Quantitative proteomics using isobaric labeling comparing PLX-EVs to PLX-derived soluble factors revealed significant differential enrichment of 495 proteins in purified PLX-EVs involved in angiogenesis, cell movement and immune system signaling. At the functional level, PLX-EVs and cells but not the corresponding soluble factors inhibited T cell mitogenesis. PLX-EVs and soluble factors displayed dose-dependent proangiogenic potential by enhancing tube-like structure formation in vitro. Our findings indicate an alternative mode of PLX action based on EV-mediated proangiogenic function and immune response modulation that may help explaining clinical efficacy beyond presence of the transplanted allogeneic cells.

ADP-heptose attenuates H. pylori-induced type I IFN signaling in DCs and impairs subsequent T cell responses

Project description:Helicobacter pylori (H. pylori) colonizes the gastric mucosa of approximately 50% of the world’s population. While it is well established that H. pylori is a major cause of gastric cancer, it has only recently been reported that H. pylori can also have adverse effects on immunological processes, including autoimmune diseases and immunotherapies. Understanding the paradoxical interplay between H. pylori and the human immune system will therefore be beneficial to improve a variety of therapeutic approaches. Here we show that ADP-heptose, a metabolite originally reported to act as a bona fide PAMP, attenuates full-fledged activation of primary human dendritic cells in the context of H. pylori infection by impairing type I IFN signaling, which in turn results in reduced DC maturation and T cell activation. Thus, this study provides novel mechanistic insights into how H. pylori utilizes ADP-heptose to mitigate host immunity.

Towards middle-up analysis of polyclonal antibodies: subclass-specific N-glycosylation profiling of murine immunoglobulin G (IgG) by means of HPLC-MS

Project description:Advanced analytical strategies including top-down and middle-up HPLC-MS approaches have become powerful alternatives to classical bottom-up analysis for the characterization of therapeutic monoclonal antibodies. Here, we assess feasibility of middle-up analysis of polyclonal IgGs posing additional challenges due to extensive sequence variability. The presented workflow is based on Fc/2 portions as conserved subunits of IgGs and enables global profiling of subclasses and their glycosylation patterns, both of which influence IgG effector functions. To obtain subunits of murine IgGs, we established digestion with the bacterial protease SpeB. The resulting Fc/2 portions characteristic of different subclasses were subsequently analysed by ion-pair reversed-phase HPLC hyphenated to high-resolution mass spectrometry allowing relative quantification of IgG subclasses and their N-glycosylation variants. In order to assess method capabilities in an immunological context, we applied the analytical workflow to polyclonal antibodies obtained from BALB/c mice immunized with the grass pollen allergen Phl p 6. This analysis simultaneously revealed a shift in IgG subclasses and Fc-glycosylation patterns in total and antigen-specific IgGs from different mouse cohorts. Eventually, Fc/2 characterization may reveal other protein modifications including oxidation, amino acid exchanges, and C-terminal lysine as demonstrated for monoclonal IgGs, which may be implemented for quality control of functional antibodies.

Angiogenesis study of Gd@C82(OH)22 nanoparticle

Project description:we report a novel nanomedicine (Gd@C82(OH)22 ) effectively inhibit human breast tumor growth by antiangiogenesis in vivo. To further identify which angiogenic factor(s) were affected on mRNA level, the "RT² Profiler™ PCR Array Mouse Angiogenesis (APMM-024, SuperArray Bioscience Corporation)" was used. The 0.9% saline soultion was used as control. At the end of the experiment, parts of the tumor tissue was collected, and further be analyzed by PCR-array.

In-Depth Proteomics Reveals Selective EV Protein Sorting and Pathway Perturbation in AML upon Synergistic FLT3 and Hedgehog Pathway Inhibition

Project description:Acute myeloid leukemia (AML) is an aggressive cancer mainly affecting bone marrow and blood with a high relapse incidence and mortality rate. Approximately one third of AML patients carry an fms-like tyrosine kinase 3 (FLT3) mutation, which is often associated with GLI expression and Hedgehog signaling, leading to tumor proliferation. AML cells shape their microenvironment into a leukemia-permissive space by releasing extracellular vesicles (EVs). EVs can transfer chemoresistance and thereby play an important role in refractory and relapsing diseases. Here, we discovered a synergistic effect of a combined treatment with the FLT3 inhibitor Crenolanib and the Hedgehog pathway inhibitor HPI-1 in the AML cell lines MOLM-14 and MV4-11. In-depth comparative proteomics revealed alterations in the cellular and the EV proteome upon single or combined inhibition of FLT3 and GLI, highlighting affected pathways. By comparing the cellular and EV proteomes, we report that transport of ribosomal proteins such as RPS26 and RPL27A and ErbB pathway members such as GAB1, GRB2 and SHC1 to EVs is selectively avoided upon treatment with Crenolanib. Ribosomal and ErbB signaling pathway proteins may play an important role in microenvironmental modulation by EVs, and Crenolanib treatment potentially acts by interfering with leukemia niche formation.

Effect of in utero undernutrition on skeletal muscle gene expression

Project description:In utero undernutrition is associated with obesity and insulin resistance, although its effect on skeletal muscle remains poorly defined. We report that, in mice, adult offspring from undernourished dams have decreased energy expenditure, decreased skeletal muscle mitochondrial content, and altered energetics in isolated mitochondria and permeabilized muscle fibers. Strikingly, when these mice are put on a 40% calorie restricted diet they lose half as much weight as calorie restricted controls. Our results reveal for the first time that in utero undernutrition alters metabolic physiology having a profound effect on skeletal muscle energetics and response to calorie restriction in adulthood. We have used a mouse model of low birth weight generated through 50% food restriction of mouse dams during the third week of gestation. We have studied in utero food restricted offspring and control offspring that were not food restricted in utero in both the ad libitum and calorie restricted states. Gene expression profiling was performed on tibialis anterior muscle from 8 mice per group, pooled in pairs.
