Effect of insulin receptor knockdown on the proteome and secretome of CYLD cutaneous syndrome primary keratinocytes

ABSTRACT: CYLD cutaneous syndrome (CCS) is a rare genetic disorder characterized by multiple skin appendage tumors. This study investigated the impact of insulin receptor (INSR) knockdown on the cellular proteome and secretome of CCS primary keratinocytes. Three biological replicates each of control and INSR knockdown CCS keratinocytes were analyzed using label-free DIA mass spectrometry on a Bruker timsTOF HT. Both whole cell lysates and secreted proteins (secretome) were examined to understand the role of INSR signaling in CCS tumor biology. Data were acquired in data-independent acquisition (DIA) mode and processed using DIA-NN v2.
