Proteomics characterization of human peritoneal tissue

ABSTRACT: To identify the proteome of human peritoneal tissue, 43 FFPE and 5 fresh frozen peritoneal tissue samples at healthy or non-cancerous state were collected and subjected to mass spectrometry analysis. The resulting findings represnt a baseline catalog of the proteomic composition of the peritoneum, as a comparison dataset for future studies focused on alterations in pathologic states such as peritoneal carcinomatosis.

Single cell RNA sequencing of total peritoneal cells from Beclinf/f-Lyz2cre mice

Project description:In Beclinf/f-Lyz2cre mice, we observed proinflammatory activation of tissue-resident peritoneal macrophages. These mice were resistent to lethal bacterial infection with Listeria. It is important to understand how the peritoneal immunity is affected by Beclin deletion in myeloid cells. To characterize the immune cells in the peritoneum, we performed single cell RNA sequencing of total peritoneal cells from Beclinf/f-Lyz2cre and littermate control Beclinf/f mice. The single cell RNA-seq analyses revealed profound changes in both the myeloid compartment and bystander cells.

miRNA expression profile in subcutaneous adipose tissue from morbid obese patients

Project description:To evaluate wether miRNA expression patterns contributes to obesity total RNA were purified from subcutaneous adipose tissue (SAT)and used in miRNA microarrays. Platform contain LNA-modified probes for all human miRNAs present in release 8.2 of the miRBase microRNA Registry. Two RNA pools from 3 non obese men (CM) and from 5 non obese women (CW) were used as controls. Expression profiling revealed that a large set of miRNAs is expressed in SAT. Forty two miRNAs changed by at least 1.5 folds in 17/20 obese subjects versus non obese control pool. Particularly, 21/42 were up-regulated and 21/42 were down-regulated. Among the differentially expressed miRNA, miR-519d, miR-498 and miR-150 were up-regulated, miR-659 and miR-371-3p_MM2 were down-regulated consistently in 20/20 obese subjects. To search for miRNAs eventually associated with obesity we used a microarray to evaluate the expression of 1458 different miRNAs in 10 obese women (OBW) and 10 obese men (OBM). Two RNA pools from 3 non obese men (CM) and from 5 non obese women (CW) were used as controls.

Single cell transcriptome of peritoneal cells

Project description:To define the transcriptomic landscape of human mesothelial cells in response to PD, we analyzed single cell transcriptomes including cells dissociated from normal peritoneum (hernia surgery, n = 3), and peritoneal cells from effluent of short-term PD patients (PD less than 2 weeks, n = 6) and from long-term PD patients (PD more than 6 years, n = 4) ,we demonstrate that mesothelial cells develop hyperglycolysis, a metabolic alteration that is correlated with the development of peritoneal fibrosis.

Single cell RNASeq analysis of memory TH2 cell response to an intestianl nematode

Project description:Intestinal parasitic nematodes, affecting a quarter of the world’s population, typically elicit prominent effector Th2-driven host immune responses. However, a more detailed understanding of nematode-induced memory Th2 responses remains scarce. We investigated the activation of peritoneal memory Th2 cells and the mechanisms driving early recall responses to the enteric nematode Heligmosomoides polygyrus in mice. We show memory Th2 cells with distinct transcriptional profiles to persist systemically in lymphoid and non-lymphoid tissues following cure of infection. Moreover, peritoneal memory Th2 cells display strong cytokine production, upregulate costimulatory marker Ox40 (CD134) and IL-5 production in a parasite-specific manner as early as 3 days following challenge infection. This effect is paralleled by an influx of Ox40L+ dendritic cells (DC) and eosinophils, both appearing exclusively in the peritoneum of reinfected mice. Finally, we show that peritoneal mesothelial cells (PeM) significantly downregulate their expression of VCAM-1 and ICAM-1 in response to a secondary infection, pointing towards an involvement in immune cell efflux. Thus, our data indicate a pivotal role of the peritoneum for memory Th2 responses and a role for PeM cells in immune cell recruitment, activation and efflux during recall responses to a strictly intestinal pathogen.

Human normal and peritumor liver tissues LC-MSMS

Project description:The tumor microenvironment (TME) plays an important role in tumorigenesis1. The TME is usually studied in tumor tissue and in relation to tumor progression2,3, which can limit understanding of how the TME is involved in processes associated with initial tumorigenesis as well as tumor recurrence and metastasis after surgery. To describe the underlying mechanisms of human hepatocellular carcinoma (HCC) occurrence and progression, we propose a new concept, the peritumor microenvironment (PME). We collected normal (all biochemical indicators of liver function, imaging examination and histopathology examination were normal) and peritumor (The liver tissues adjacent to the tumor were approximately 2 cm away from the tumor and were taken from patients with HCC who did not receive tumor radiotherapy, tumor chemotherapy or targeted drug therapy before surgery) to perform a proteomic characterization of the PME in peritumor liver tissues.

Mesothelioma location influences the tumour microenvironment and immune checkpoint therapy response in preclinical models

Project description:Mesothelioma is a cancer derived from mesothelial cells, most commonly arising from the pleura or the peritoneum. Immune checkpoint therapy (ICT) has shown survival benefit for pleural mesothelioma, but little is known about the response in peritoneal mesothelioma. Most preclinical mesothelioma models involve subcutaneous cancer cell implantation, which lacks the relevant tumour microenvironment of peritoneal mesothelioma and does not resemble the clinical presentation. We therefore set out to explore the influence of location on the mesothelioma tumour microenvironment, comparing pleural, peritoneal, and subcutaneous models using identical cell line-derived syngeneic mesotheliomas. We found that the peritoneal location conferred an anti-inflammatory tumour microenvironment, characterised by low IFN, TNFα, and STAT signalling activity, low immune cell infiltration and a gene signature associated with non-response to ICT. ICT was effective in subcutaneous models, but the same cell line-derived tumours were irresponsive when inoculated intraperitoneally. Together, these findings show that peritoneal location is associated with an immune suppressive tumour microenvironment.
