HCC INSIG1 Mass Spectrometric Analysis

NSD2 promotes HCC metastasis through methylating PKM2 and activating SF3B1-dependent alternative splicing

Project description:Hepatocellular carcinoma (HCC) is notorious for its early and frequent metastases. To understand the molecular mechanisms underlying HCC metastasis, we generated a pulmonary metastasis HCC mouse model and performed both time-series transcriptomics and proteomics analysis of protein methylation. We found that methyltransferase NSD2 with significant upregulation in the tipping point for metastasis was closely correlated with high numbers of methylated-proteins in HCC tissues. NSD2 promoted the invasion and metastasis of HCC cells, both in vitro and in vivo. Mechanistically, NSD2 directly bound to PKM2, a glycolysis rate-limiting enzyme, and catalyzed di-methylation of PKM2 at the lysine 336 residue. Further investigation demonstrated that NSD2-mediated di-methylation of PKM2 increased the intracellular glycolytic rate and lactate production by enhancing its pyruvate kinase activity. High-lactate level in HCC cells lead to lactylation of splicing factor 3B subunit 1 (SF3B1) at lysine 333 residue, promoting SF3B1-mediated RNA splicing of several metastasis-related genes. Further, UNC8153, a novel NSD2-targeted degrader, inhibited HCC metastasis in PDX model. Altogether, our study identifies a key methyltransferase NSD2 for HCC metastasis and reveals a protein methylation-mediated molecular mechanism catalyzed by NSD2 integrate glycolysis regulation and alternative splicing.

Project description:Tuberculosis (TB) remains a major public health problem and we lack a comprehensive understanding of how Mycobacterium tuberculosis (M. tb) infection impacts host immune responses. We compared, at two timepoints, the induced immune response to TB antigen, BCG and IL-1β stimulation between latently M. tb infected individuals (LTBI) and active TB patients. The immune response was assessed using the TruCulture system with a Null stimulation. samples were tested by Ultrahigh Performance Liquid Chromatography-Tandem Mass Spectroscopy (UPLC MS/MS) for a total of 696 metabolites.

Huh7 cells exist as a mixed population of cells with distinct patterns of gene methylation

Project description:A powerful approach to study innate antiviral response is to compare the difference between wild type Huh7 cells, which do not support robust replication of hepatitis C virus (HCV)2, versus certain subclones of Huh7 cells that are permissive for HCV replication. We generated two permissive cell lines and two independent non-permissive subclone from Huh7 cells. We compared the global methylation pattern of these different cells and find that Huh7 cells exist as a heterogeneous population of cells with distinct patterns of gene methylation. Comparison of Huh7, HRP1, HRP4, Huh7-pNeo1 and Huh7-pNeo2 cells.

Identification of differentially expressed miRNAs in HCSC cell lines compared with HCC cell lines

Project description:Hep3B and Huh7 are two types of human hepatoma cell lines (HCC). In our laboratory, we cultured their stem-like cancer cells (HCSCs), Hep3B-C and Huh7-C. And we have demonstrated that these cells had enhanced stem cell properties, drug resistance, properties of EMT, and stronger tumor-initiating capabilities. To explore functionally crucial miRNAs in HCSCs, 2 samples of HCSCs and 2 samples of HCCs were sequenced by the Illumina Genome Analyzer II. Through differential expression analysis, we finally identified 9 up- and 9 down-regulated miRNAs which were consistently up- and down-regulated in two stem cells compared to the cancer cells. Expression analysis using total RNAs extracted from 2 HCSC cell lines (Hep3B-C and Huh7-C), and 2 HCC cell lines (Hep3B and Huh7).
