Exploration of protein degradability enables fully endogenous MrTAC degraders


ABSTRACT: Methylarginine degrons mark proteins for degradation in lysosomes. We leveraged the machinery responsible for methylarginine degron addition to create a chemically inducible degrader named a Methyl Arginine Targeting Chimera (MrTAC). First, we evaluated the contents of isolated lysosomes following activation of the native methylarginine degron pathway. Next, we evaluated global proteome abundance in human cell culture during treatment with different MrTACs relative to control treatments.
