Proteomic profiling of frozen and lyophilized bovine milk-derived extracellular vesicles using LC-MS/MS

ABSTRACT: This project aimed to compare the proteomic landscapes of bovine milk-derived extracellular vesicles (EVs) preserved under two storage conditions—lyophilization and conventional freezing at −80 °C. By evaluating how different preservation strategies influence EV protein composition, the study sought to determine whether lyophilization can serve as a practical and reliable alternative to low-temperature storage for maintaining EV integrity and functionality.

Extracellular vesicles (EVs) are promising therapeutic agents due to their role in intercellular communication. This study examined the protective effects of milk-derived EVs (mEVs) on bovine oviductal epithelial cells (BOECs) under cobalt chloride (CoCl2)-induced oxidative stress (OS), comparing EVs stored at -80 °C or lyophilized. mEVs and algae-derived EVs (aEVs; negative control) were isolated via tangential flow filtration and applied at 107, 109, and 10<sup  ...[more]
