Project description:Transthyretin (TTR) aggregation causes transthyretin amyloidosis, a severe condition often linked to mutations that destabilize the TTR tetramer, leading to amyloid fibril formation. Small molecules that stabilize the tetramer can prevent this process. While over 300 X-ray structures of TTR exist, they represent static snapshots and fail to show dynamic changes from mutations or ligand binding. This project uses hydrogen-deuterium exchange (HDX) and fast photochemical oxidation of proteins (FPOP) with mass spectrometry (MS) to study these conformational dynamics.

Conformational differences in the light chain constant domain of immunoglobulin G and free light chain may influence proteolysis in AL amyloidosis

Project description:Immunoglobulin light chain amyloidosis (AL) is a life-threatening disease caused by the deposition of monoclonal light chain (LC) and its fragments containing variable (VL) and portions of constant (CL) domains. AL patients feature either monoclonal free LCs circulating as covalent and noncovalent homodimers, or monoclonal immunoglobulin (Ig) wherein LC and heavy chain (HC) form disulfide-linked heterodimers, or both. The role of monoclonal Ig in AL amyloidosis is unclear as prior studies focused on full-length free LC or VL domain. We used a mammalian cell-based expression system to generate four AL patient-derived full-length IgGs, two non-AL IgG controls, and six corresponding free LC proteins derived from IGLV6-57 germline precursor. Comparison of proteins’ secondary structure, thermal stability, proteolytic susceptibility, and disulfide link reduction suggested the importance of local vs. global conformational stability. Analysis of IgGs vs. corresponding free LCs using hydrogen-deuterium exchange mass spectrometry revealed major differences in the local conformation/dynamics of CL domain. In all IgGs vs. LCs, segment containing ß-strand and -helix ßAC-ACBC was more dynamic/exposed while segment ßDC-ßEC was less dynamic/exposed. Notably, these segments overlapped proteolysis-prone regions whose in vivo cleavage generates LC fragments found in AL deposits. Altogether, the results suggest that preferential cleavage in segments ßAC-ACBC of free LC or ßDC-ßEC of IgG helps generate amyloid protein precursors. We propose that protecting these segments using small-molecule stabilizers, which bind to the interfacial cavities CL-CL in free LC or CL-CH1 in IgG, is a potential therapeutic strategy to complement current approaches targeting VL-VL or VL-CL stabilization.

Ligand induced conformational changes in the B1-Adrenergic Receptor Revealed by Hydrogen-Deuterium Exchange Mass Spectrometry

Project description:Project employ Hydrogen Deuterium Exchange Mass Spectrometry (HDX-MS), to characterise the structural dynamics of turkey β1-adrenergic receptor (tβ1AR) in complex with nine ligands, including agonists, partial agonists and antagonists.We show that dynamic signatures across the GPCR structure can be grouped by compound modality. Surprisingly, we discovered repeated destabilisation of the intracellular loop 1 (ICL1) upon full agonist binding and stabilisation upon antagonist binding, suggesting that increased dynamics in this region are an essential component for G protein recruitment. Multiple sequence alignments and molecular dynamics simulations indicate that L72 in ICL1 plays an important structural role. Differential HDX-MS experiment of tβ1AR and tβ1AR L72A construct in complex with miniGs, in response to various ligands, suggests involvement of ICL1 in stabilising the GDP bound state by influencing the stability of HG helix of miniGs.

Structural proteomics defines a sequential priming mechanism for the progesterone receptor (HDX-MS)

Project description:The progesterone receptor (PR) is a steroid-responsive nuclear receptor with two isoforms: PR-A and PR-B. Disruption of PR-A:PR-B signaling is associated with breast cancer through interactions with oncogenic co-regulatory proteins (CoRs). However, molecular details of isoform-specific PR-CoR interactions remain poorly understood. Using structural mass spectrometry, we investigate the sequential binding mechanism of purified full-length PR and intact CoRs, steroid receptor coactivator 3 (SRC3) and p300, as complexes on target DNA. Our findings reveal selective CoR NR-box binding by PR and novel interaction surfaces between PR and CoRs during complex assembly, providing a structural basis for CoR sequential binding on PR. Antagonist-bound PR showed persistent CoR interactions, challenging the classical model of nuclear receptor activation and repression. Collectively, we offer a peptide-level perspective on the organization of the PR transcriptional complex and infer the mechanisms behind the interactions of these proteins, both in active and inactive conformations.

Patient-derived anti-Siglec-6 antibodies engineered into T cell-recruiting bispecific antibody formats for treating leukemia

Project description:Despite numerous therapeutic options, safe and curative therapy is out of reach for most chronic lymphocytic leukemia (CLL) patients. We previously reported the identification of a patient-derived CLL-binding antibody, JML-1, and identified Siglec-6 as the target of JML-1. Siglec-6 is an attractive target for cancer immunotherapy due to its absence on healthy tissues and potential role in immune signaling. In this work, we identify additional patient-derived anti-Siglec-6 antibodies, RC-1 and RC-2, that bind with higher affinity than JML-1, yet exhibit similar specificity and overlapping conformational epitopes. Both JML-1 and RC-1 were employed to generate anti-Siglec-6 x anti-CD3 T cell-recruiting bispecific antibodies (T-biAbs) which were effective in vitro at activating T cells and lysing target cells. Upon engineering RC-1 into more compact and rigid T-biAb formats, we achieved sub-pM EC50 values for cell lysis in vitro. In addition, the T-biAbs mediated the killing of primary CLL cells by autologous T cells at natural effector-to-target cell ratios ex vivo. The increased potency of this T-biAb format appears to be due to the reduction in the length and/or flexibility of the cytolytic synapse. Furthermore, the anti-Siglec-6 x anti-CD3 T-biAb was effective at curing mice in a systemic in vivo model of CLL.

Project description:The biological effects of TTR proteins in the vasculature remain unknown. We used microarrays to detail the modulation of gene expression on HUVECs by V30M TTR when compared to cells exposed to WT TTR. HUVECs (passage 7) were cultured in the presence of WT or V30M TTR at 4M-BM-5M for 3 hours. RNA was extracted and hybridized on Affymetrix microarrays. We sought to obtain differentially expressed genes modulated by V30M TTR protein.
